    Local Stability in the Process of Excavation Located in High Permeability Saturated Sand of Diaphragm Wall Construction

    The stability of the slurry trench is very important in the construction of the underground diaphragm wall. In the current research, the local instability of the slurry trench is mainly investigated after the excavation of a unit slot is completely completed. However, the local stability in the process of excavation has received little attention. In this paper, the local stability in the process of excavation located in high permeability strata of diaphragm wall construction is investigated. A slurry infiltration experiment was carried out to investigate the distribution of the excess pore pressure in the high permeability strata, which can determine the effective support pressure. Then, the local stability of the slurry trench in the process of excavation located in high permeability saturated sand is calculated. The results show that the same types of sand according to the design code cannot be simply treated to have the same permeability and similar distribution of the excess pore pressure, since whether the filter cake can be formed and the quality of the filter cake are the key factors to determine the distribution of the excess pore pressure. This is also crucial for the local stability in the process of excavation located in high permeability saturated sand. It is suggested that attention should be paid to the local stability in the process of excavation located in high permeability strata when the slurry infiltration mode is the pure permeable zone

    Characteristics of the O(1S) to O(1D) 557.7 nm green emission observed in an argon plasma jet

    An extensive study on the green auroral emission characterization is presented based on a single dielectric barrier discharge geometry argon plasma jet driven by a kHz sine voltage. The plasma was generated by using 99.999% pure argon and the observed 557.7 nm green line resulted from the excited O(1S) state. An optical emission spectroscopy method using line ratios of argon was used to obtain the electron density and electron temperature under different conditions in the downstream region. The characteristics of discharge and green emission with variations in interelectrode distance, applied voltage (power) and flow rate are discussed. The spatially diffuse distribution of O(1S), owing to its long lifetime, is shown by the short exposure imaging. Two discharge regimes are presented, accompanied by two distinct branches of the green emission intensity, with a clear conclusion that the 557.7 nm emission is favored in the low electron temperature environment. In this work, the intense and diffuse green plume only forms when the downstream electron density is approximately lower than 1 × 1014 cm−3 and the electron temperature is lower than 1.1 eV. By charging the two electrodes in two opposite ways, it is shown that the green emission from oxygen is favored in the case where the electric field and the electron drift are not continuous

    In order to solve the problems existing in the construction of underground structures located in the downtown of saturated soft soil area, such as insufficient construction site, complex adjacent structures and great impact on the surrounding environment, the construction method of underground bundled integrate tunnel(U-BIT) is proposed. In this method, after steel pipes jacking completed, concrete is filled into the pipes, and prestress is tensioned to make each independent pipe combined to form a whole bearing structure, so as to achieve the purpose of reducing the size of structural components, improving the structural stiffness and bearing capacity. Based on the structural mechanical properties test and the project of Wuding Road Station of Shanghai Metro Line 14, the failure mechanism of bundled integrate structure, the tension technology of prestressed tendons in narrow space and the variation rules of ground surface subsidence are systematically studied. The research shows that structural seam sections will be destroyed before pipe sections, so ensuring the mechanical performance of seam sections is very important to make sure the structural safety. Since each independent pipe is combined to form an overall stable structure under the prestress effect, the subsequent soil excavation has little influence on the tension of prestressed tendons and ground surface deformation. Therefore, the above construction method can control the ground surface subsidence effectively and reduce the influence of underground engineering construction on the surrounding environment.  &nbsp

    MiR-196a-5p facilitates progression of estrogen-dependent endometrial cancer by regulating FOXO1

    Background and Purpose. Estrogen-dependent endometrial cancer mainly occurs in younger pre-menopausal and post-menopausal women and threatens their health. Recently, microRNAs (miRNAs) have been considered as novel targets in endometrial cancer treatment. Therefore, we aimed to explore the effect of miRNA (miR)-196a-5p in estrogen-dependent endometrial cancer. Methods. 17β-estradiol (E2; 2.5, 5, 10 and 20 nM) was used to treat RL95-2, HEC-1B and ECC-1 cells followed by cell viability assessment using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT). The level of miR-196a-5p was measured by reverse transcription-quantitative PCR (RT-qPCR). We then transfected miR-196a-5p mimic/inhibitor and Forkhead box protein O1 (FOXO1) small interfering RNA (siRNA) into E2-treated cells. Apoptotic cells were measured by flow cytometry. Wound healing and Transwell assays were implemented to assess migration and invasion. Bioinformatics and luciferase reporter assays were applied to confirm the interaction between miR-196a-5p and FOXO1. Immunoblotting determined the levels of FOXO1, Bcl-2, Bax, Caspase 3. Results. E2 promoted cell viability and miR-196a-5p expression in RL95-2 and ECC-1 cells. miR-196a-5p mimic enhanced cell viability, migration and invasion but suppressed apoptosis and FOXO1, whilst miR-196a-5p inhibitor blocked these processes. In addition, miR-196a-5p upregulated Bcl-2, but down regulated Bax and Caspase 3 expression, an effect that was reversed by miR-196a-5p inhibitor. We determined that miR-196a-5p targeted FOXO1, and that si-FOXO1 blocked the effects of miR-196a-5p inhibitor on viability, apoptosis, migration and invasion of E2-treated RL95-2 and ECC-1 cells. Conclusions. Our findings suggested potential diagnostic and therapeutic applications for miR-196a-5p and its FOXO1 target in patients suffering from estrogen-dependent endometrial cancer
