    Establishment of core collection from apricot germplasm in China

    This study aimed at establishing a core collection based on the analysis of data from simple sequence repeat (SSR) alleles and morphological and agronomical traits (MOR) of the primary core collection from apricot germplasm resources. The index of genetic diversity, and frequency ratios of retention and loss of the alleles were studied between cluster and random sampling methods at five sampling ratios. The results demonstrate that the cluster sampling method preceded random sampling, and cluster sampling of SSR combined with MOR at the rate of 80% was the best sampling strategy among all the sampling methods. Based on this sampling strategy, 120 accessions were selected as the core collection of apricot, which retained 100% alleles in the primary core collection and 100% phenotypic characters. The core collection developed had also been evaluated by using the data of six quantitative traits, which showed that the established core collection could well represent the genetic diversity of the original collection of 1501 apricot accessions.Keywords: Apricot, core collection, sampling strategy, simple sequence repeat (SSR) molecular markerAfrican Journal of Biotechnology Vol. 12(37), pp. 5577-558

    P2X2 Dominant Deafness Mutations Have No Negative Effect on Wild-Type Isoform: Implications for Functional Rescue and in Deafness Mechanism

    The P2X2 receptor is an ATP-gated ion channel, assembled by three subunits. Recently, it has been found that heterozygous mutations of P2X2 V60L and G353R can cause autosomal dominant nonsyndromic hearing loss. However, the underlying mechanism remains unclear. The fact that heterozygous mutations cause deafness suggests that the mutations may have dominant-negative effect (DNE) on wild-type (WT) P2X2 isoforms and/or other partners leading to hearing loss. In this study, the effect of these dominant deafness P2X2 mutations on WT P2X2 was investigated. We found that sole transfection of both V60L and G353R deafness mutants could efficiently target to the plasma membrane, like WT P2X2, but exhibit a significantly reduced response to ATP stimulation. Both mutants reduced the channel conductance, but G353R mutation also altered the voltage dependency. Co-expression with WT P2X2 could restore the response to ATP. As the ratio of WT P2X2 vs. mutants increased, the response to ATP was also increased. Computer modeling confirmed that both V60L and G353R dominant-deafness mutant subunits do not have any negative effect on WT P2X2 subunit, when assembled as a heterotrimer. Improper docking or defective gating is the more likely mechanism for impaired channel function by these P2X2 deafness mutations. These results suggest that P2X2 dominant deafness mutations do not have negative effects on WT P2X2 isoforms, and that adding additional WT P2X2 could rescue the lost channel function caused by the deafness mutations. These P2X2 dominant deafness mutations may have negative-effects on other partners leading to hearing loss

    Effective Silencing of Sry Gene with RNA Interference in Developing Mouse Embryos Resulted in Feminization of XY Gonad

    Delivering siRNA or shRNA into the developing embryos is still a main challenge to use of RNAi in mammalian systems. Here we analyze several factors influencing RNAi-mediated silencing of Sry gene, which is a tightly controlled spatiotemporal expressed gene and only shortly expressed in developing mouse embryo gonad. A Sry gene-specific shRNAs expression vector (pSilencer4.1/Sry565) was constructed. The shRNA constructs were mixed with polyethylenimines (PEIs) to form a complex and then injected into pregnant mice though tail vein. Our results showed that Sry gene was downregulated significantly in developing embryos. Further study revealed that knocking-down of Sry expression resulted in feminization of gonad development in mouse embryos and the expression level of Sox9 and Wt1 gene was also significantly changed by downregulation of Sry. The transfection efficiency is associated with the amount of plasmid DNA injection, injection time, injection speed, and volume. Our studies suggest that transplacental RNAi could be implemented by tail vein injection of plasmid vector into pregnant mice

    Efficiency of Public Educational Expenditure in China

    The study investigates the efficiency of local public educational expenditure of 31 provinces in China during 2005-2010, using the Slack-based Measurement (SBM) directional distance function. The results show that public educational expenditure is the most efficient in eastern China, followed by middle and western areas. The inefficiency can be explained mostly by the number of master graduates, while the impacts of the number of undergraduates and graduates from secondary school are also significant. Additionally, bootstrap method is applied to explore the contextual factors influencing the efficiency. The results suggest that economic development and urbanization process increase the efficiency, while the state-owned industry obstructs the development
