    As a Chinese person, I often feel unsatisfied with some aspects of my country just like some British complaining about foggy weather in London. But I have never been picky about China's food. Moreover, sometimes I feel I'm lucky to live in China with so many great foods in varying cuisines throughout the whole big country. If you are a chowhound like me, you know what I am talking about. This is the tenth month I have been in Columbia, which also means that I have been away from home for nearly one year.     IL-1β-Mediated Up-Regulation of WT1D via miR-144-3p and Their Synergistic Effect with NF-κB/COX-2/HIF-1α Pathway on Cell Proliferation in LUAD

    Background/Aims: IL-1β is an important mediator of “inflammation-cancer" transformation through IL-1β/NF-κB/COX-2/HIF-1α signaling pathway, whereas certain portion of patients with lung adenocarcinoma (LUAD) still suffer from rapid tumor progression in clinical practice, indicating the occurrence of potential bypass. Methods: Real-time polymerase chain reaction was applied to examine the expressions of mir-144-3p, WT1, NF-κB, COX2 and HIF-1α at the mRNA level in 127 LUAD samples and corresponding adjacent tissues. miR-144-3p mimic and antagormiR were used to trigger activation and suppression of miR-144-3p in A549 cells, respectively. MTT assay and Western blotting analysis were carried out to evaluate the cell proliferation. Stable clones with over-expression or knockdown of WT1 were generated with plasmid or shRNA by lentiviral vector technology in H1568 and H1650 NSCLC cell lines, respectively. Dual luciferase reporter assay was performed to validate the effect of miR-144-3p on WT1D. Xenograft model was established for in vivo experiment, and TCGA data were extracted for validation. Results: miR-144-3p could suppress the WT1D expression at the post-transcriptional level, hence regulating cell proliferation in LUAD. WT1 and COX-2 were independent prognostic factors of LUAD patients. In addition, inhibition of IL-1β/miR-144-3p/WT1D and IL-1β/NF-κB/COX-2/HIF-1α pathways using miR-144-3p mimic and Celecoxib, respectively, displayed synergistic suppressive effect on cell proliferation in LUAD. Conclusion: A de novo IL-1β/miR-144-3p/WT1D axis was involved in proliferative regulation of LUAD. Moreover, simultaneous blockade of both IL-1β/miR-144-3p/WT1D and IL-1β/NF-κB/COX-2/ HIF-1α pathways might have synergistic suppressive effect on cell proliferation in LUAD

    Prognostic Significance of miR-181b and miR-21 in Gastric Cancer Patients Treated with S-1/Oxaliplatin or Doxifluridine/Oxaliplatin

    Background: The goal of this study is to evaluate the effectiveness of S-1/Oxaliplatin vs. Doxifluridine/Oxaliplatin regimen and to identify miRNAs as potential prognostic biomarkers in gastric cancer patients. The expression of candidate miRNAs was quantified from fifty-five late stage gastric cancer FFPE specimens. Experimental Design: Gastric cancer patients with KPS>70 were recruited for the trial. The control group was treated with 400 mg/twice/day Doxifluridine plus i.v. with Oxaliplatin at 130 mg/m 2/first day/4 week cycle. The testing group was treated with S-1 at 40 mg/twice/day/4 week cycle plus i.v. with Oxaliplatin at 130 mg/m 2/first day/4 week cycle. Total RNAs were extracted from normal and gastric tumor specimens. The levels of miRNAs were quantified using real time qRT-PCR expression analysis. Results: The overall objective response rate (CR+PR) of patients treated with S-1/Oxaliplatin was 33.3% (CR+PR) vs. 17.6% (CR+PR) with Doxifluridine/Oxaliplatin for advanced stage gastric cancer patients. The average overall survival for patients treated with S-1/Oxaliplatin was 7.80 month vs. 7.30 month with patients treated with Doxifluridine/Oxaliplatin. The expression of miR-181b (P = 0.022) and miR-21 (P = 0.0029) was significantly overexpressed in gastric tumors compared to normal gastric tissues. Kaplan-Meier survival analysis revealed that low levels of miR-21 expression (Log rank test, hazard ratio: 0.17, CI = 0.06-0.45; P = 0.0004) and miR-181b (Log rank test, hazard ratio: 0.37, CI = 0.16-0.87; P = 0.018) are closely associated with better patient's overall survival for both S-1 and Doxifluridine based regimens. Conclusion: Patients treated with S-1/Oxaliplatin had a better response than those treated with Doxifluridine/Oxaliplatin. miR-21 and miR-181b hold great potential as prognostic biomarkers in late stage gastric cancer. © 2011 Jiang et al

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    Dewatering of <i>Juglans mandshurica</i> Wood Using Supercritical Carbon Dioxide

    Supercritical carbon dioxide (ScCO2), known for such features as good solubility and mass transfer properties, can be an efficient drying medium for various materials, such as wood, by filling the pore space and dissolving water in the cell cavity without altering the microstructure. In this study, two specimens of Juglans mandshurica wood with a length of 30 mm and 140 mm were subjected to ScCO2 dewatering under four different pressure and temperature conditions. The results showed that the drying rate is mainly influenced by pressure and temperature, with pressure having the more significant effect. Moreover, the efficiency of dewatering was not dependent on the sample length under the same conditions. The moisture content (MC) was the same along the longitudinal direction throughout both the surfaces and core of the wood. While there were no significant differences in dewatering rate between tangential and radial directions and lengths of samples, significant MC gradient differences were noted along wood in radial and tangential directions. During ScCO2 dewatering, the dominant water transfer occurred from the middle towards the end surfaces along the wood’s longitudinal directions. Furthermore, ScCO2 dewatering did not result in any shrinkage or significant drying stress, but it did cause some swelling in Juglans mandshurica wood

    Prognostic Significance of MiRNA in Patients with Diffuse Large B-Cell Lymphoma: a Meta-Analysis

    Introduction: This pooled analysis study aimed to reveal the prognostic relevance of microRNAs (miRNAs) in patients with diffuse large B-cell lymphoma (DLBCL). Materials and Methods: We examined the impact of miRNAs on clinical outcome. Eligible studies were identified and quality assessed using multiple search strategies. Data were extracted from included studies which correlated survival with expression of miRNAs (serum or tissue). Results: We pooled proper studies, and combined the hazard ratios with 95% confidence intervals to estimate strength of the correlations. There were 18 studies including 1950 patients with DLBCL eligible for pooled analysis. We found significant combined HRs for poor overall survival for high expression of miR-21 and low expression of miR-224 in tumor tissue, but for favorable relapse free survival for high expression of miR-21 in serum. Progression free survival was shortened in patients with low expression of miR-199a/b, miR-146b-5p, miR-224 and high expression of miR-222. Conclusion: MiRNAs may act as independent prognostic factors in patients with DLBCL, and useful in risk stratification

    Dissociated stimulus and response conflict effect in the Stroop task Evidence from evoked brain potentials and brain oscillations

    The Stroop task is a classic paradigm that can be used to examine cognitive control as it contains conditionswith and without interference. Cumulative evidence suggests that both stimulus and response conflictcontribute to the Stroop interference effect. However, it remains unclear whether there are dissociableevent-related potential (ERP) or frequency band-specific electroencephalographic (EEG) power changesassociated with stimulus conflict and response conflict. To investigate potential markers for each formof conflict, we applied a Stroop 2-1 mapping task in 20 healthy young adults. Results showed that anegative deflection in the 350&ndash;500 ms time window (N450) and a positive deflection in the 600&ndash;900 mstime window (late positive component, LPC) were associated with response conflict and stimulus conflict,respectively. Time-frequency analyses found that both stimulus and response conflict induced theta bandpower changes and that response conflict additionally induced a beta band power change. These resultsindicate that stimulus and response conflict in the Stroop task are associated with different ERP effectsand brain oscillatory features.</p

    Phosphoric Acid Induced Controllable Nanoparticle Aggregation for Ultrasensitive SERS Detection of Malondialdehyde in a Microfluidic Chip

    Malondialdehyde (MDA), one of the most important products of lipid peroxidation, has been widely accepted as a biomarker to indicate food rancidity as well as the progress of some human diseases. However, ready detection of MDA with ultra-high sensitivity remains a challenge. In this work, a microfluidic surface-enhanced Raman scattering (SERS) sensing chip based on phosphoric acid induced nanoparticles aggregation was proposed for ultrasensitive MDA detection. The sensing chip was composed of an ultrafast microfluidic mixer, which efficiently transferred analytes to hot spots via the mixer assisted hot spots occupying (MAHSO) SERS strategy. Phosphoric acid, a reagent used in MDA detection, played the role of aggregator to induce aggregation of silver nanoparticles (Ag NPs); meanwhile, as fast as a few milliseconds mixing time effectively prevented over-aggregation of Ag NPs. Therefore, this process generated a uniform and dense SERS substrate with analyte molecules located in hot spots. As a result, the MDA SERS sensing chip possessed a limit of detection (LOD) lower than 3.3 × 10−11 M, high spot-to-spot uniformity with a relative standard deviation (RSD) of 9.0% and an excellent batch-to-batch reproducibility with a RSD of 3.9%. This method also demonstrated excellent specificity and reliability in real sample detection with recoveries of 90.4–109.8% in spiked tests
