    Challenges of scale down model for disposable bioreactors: Case studies on growth & product quality impacts

    Despite wide-spread use of disposable bioreactors, there is a lack of well-established scale-down model for larger scale SUBs. Here we report a case of NS0 cell culture process transfer from 2000L stainless steel bioreactor (SST) to 2000L disposable bioreactor (SUB). Initial attempts in trying to grow the NS0 cells in the small scale 2D bags yielded non-satisfactory results, as growth was impacted by bag material type as well as by suppliers of the same bag material type. However, 3D bags of 50L and above proved to be supportive of the NS0 cell line growth. Even for cell lines that do not have growth issues in SUBs, surprising product quality difference between SUBs and traditional bench top glass bioreactors are still being observed, thus making the bench top glass bioreactors non-ideal as scale down models. We report two cases where glycan profiles of the expressed antibody products show such dramatic differences. In one case, extensive testing of glass bioreactors from various suppliers led to a particular type being able to mimic the glycan profiles from the SUB, whereas in the other case, alternative scale down model had to be identified and the process had to be modified to maintain the glycan profiles when scaling up to the 200L SUB

    International audienceSome of the recent publications in nanofiltration modelling converge on the importance of dielectric effects and numerous models have been developed in order to take them into account. However several works reported lately in the literature suggest a screening of image charges effect at high electrolyte concentration and the predominance of the Born effect, due to the change of dielectric constant inside the confined nanopore regarding that of the feed solution. In pursuit of an exhaustive and simple model for nanofiltration, a new approach is developed that account for both dielectric phenomena. Based on the Steric, Electric and Dielectric Exclusion (SEDE), the introduction of an average potential gradient approximation is shown to greatly improve the computational performance of the model without being detrimental to its predictive accuracy. The results obtained with this simplified model (SEDE-APG) are compared to the original SEDE model and an excellent agreement is obtained even in the case of electrolyte mixtures. Ultimately this model is confronted to experimental data of separation obtained for moderately to highly concentrated feed flows and exhibits promising result

    Finite-Horizon Optimal Control of Discrete-Time Switched Linear Systems

    Finite-horizon optimal control problems for discrete-time switched linear control systems are investigated in this paper. Two kinds of quadratic cost functions are considered. The weight matrices are different. One is subsystem dependent; the other is time dependent. For a switched linear control system, not only the control input but also the switching signals are control factors and are needed to be designed in order to minimize cost function. As a result, optimal design for switched linear control systems is more complicated than that of non-switched ones. By using the principle of dynamic programming, the optimal control laws including both the optimal switching signal and the optimal control inputs are obtained for the two problems. Two examples are given to verify the theory results in this paper

    A Fast Robot Identification and Mapping Algorithm Based on Kinect Sensor

    Internet of Things (IoT) is driving innovation in an ever-growing set of application domains such as intelligent processing for autonomous robots. For an autonomous robot, one grand challenge is how to sense its surrounding environment effectively. The Simultaneous Localization and Mapping with RGB-D Kinect camera sensor on robot, called RGB-D SLAM, has been developed for this purpose but some technical challenges must be addressed. Firstly, the efficiency of the algorithm cannot satisfy real-time requirements; secondly, the accuracy of the algorithm is unacceptable. In order to address these challenges, this paper proposes a set of novel improvement methods as follows. Firstly, the ORiented Brief (ORB) method is used in feature detection and descriptor extraction. Secondly, a bidirectional Fast Library for Approximate Nearest Neighbors (FLANN) k-Nearest Neighbor (KNN) algorithm is applied to feature match. Then, the improved RANdom SAmple Consensus (RANSAC) estimation method is adopted in the motion transformation. In the meantime, high precision General Iterative Closest Points (GICP) is utilized to register a point cloud in the motion transformation optimization. To improve the accuracy of SLAM, the reduced dynamic covariance scaling (DCS) algorithm is formulated as a global optimization problem under the G2O framework. The effectiveness of the improved algorithm has been verified by testing on standard data and comparing with the ground truth obtained on Freiburg University’s datasets. The Dr Robot X80 equipped with a Kinect camera is also applied in a building corridor to verify the correctness of the improved RGB-D SLAM algorithm. With the above experiments, it can be seen that the proposed algorithm achieves higher processing speed and better accuracy

    The Advantage of PET and CT Integration in Examination of Lung Tumors

    Purpose. To evaluate the diagnosis value of integrated positron emission tomography and computed tomography (PET/CT) with lung masses, this study emphasized the correlation between tumor size and maximum standardized uptake value (SUVmax) in selected regions of interest (ROI) of lung masses. Material and Methods. A retrospective analysis was performed on 85 patients with solid pulmonary lesions, all verified by pathology. The morphology, edge (speculated margins and lobule), size, density of pulmonary masses, and on-chest CT images were reviewed. The SUVmax in ROI of pulmonary masses was calculated. Results. Among the 85 patients with lung masses, 59 patients presented with pulmonary malignant neoplasm and 26 patients with benign lesions. The sensitivity, specificity, and accuracy were 89.8%, 61.5%, 81.2%, respectively, for PET measurement only, 88.1%, 65.4%, 81.2% for CT only, and 96.6%, 80.8%, 91.8% for PET/CT. The size of pulmonary malignant neoplasm in the 59 patients was apparently correlated with the ROI's SUVmax (r=0.617, P<.001). However, the size of pulmonary benign mass in the 26 patients was not correlated with the SUVmax. Conclusion. PET/CT is of greater value in characterization of lung masses than PET and CT performed separately. The examination of lung tumor can be further specified by the correlation between the size of pulmonary malignant neoplasm and the ROI's SUVmax

    Efficient Scene Text Detection with Textual Attention Tower

    Scene text detection has received attention for years and achieved an impressive performance across various benchmarks. In this work, we propose an efficient and accurate approach to detect multioriented text in scene images. The proposed feature fusion mechanism allows us to use a shallower network to reduce the computational complexity. A self-attention mechanism is adopted to suppress false positive detections. Experiments on public benchmarks including ICDAR 2013, ICDAR 2015 and MSRA-TD500 show that our proposed approach can achieve better or comparable performances with fewer parameters and less computational cost.Comment: Accepted by ICASSP 202
