    Is birthweight associated with total and aggressive/lethal prostate cancer risks? A systematic review and meta-analysis

    BACKGROUND: It has been hypothesised that intrauterine exposures are important for subsequent prostate cancer risk. Prior epidemiological studies have used birthweight as a proxy of cumulative intrauterine exposures to test this hypothesis, but results have been inconsistent partly because of limited statistical power. METHODS: We investigated birthweight in relation to prostate cancer in the Medical Research Council (MRC) National Survey of Health and Development (NSHD) using Cox proportional hazards models. We then conducted a meta-analysis of birthweight in relation to total and aggressive/lethal prostate cancer risks, combining results from the NSHD analysis with 13 additional studies on this relationship identified from a systematic search in four major scientific literature databases through January 2015. RESULTS: Random-effects models found that per kg increase in birthweight was positively associated with total (OR=1.02, 95% confidence interval (95% CI)=1.00, 1.05; I(2)=13%) and aggressive/lethal prostate cancer (OR=1.08, 95% CI=0.99, 1.19; I(2)=40%). Sensitivity analyses restricted to studies with birthweight extracted from medical records demonstrated stronger positive associations with total (OR=1.11, 95% CI=1.03, 1.19; I(2)=0%) and aggressive/lethal (OR=1.37, 95% CI=1.09, 1.74; I(2)=0%) prostate cancer. These studies heavily overlapped with those based in Nordic countries. CONCLUSIONS: This study provides evidence that heavier birthweight may be associated with modest increased risks of total and aggressive/lethal prostate cancer, which supports the hypothesis that intrauterine exposures may be related to subsequent prostate cancer risks

    Cloud computing service providers have large numbers of notices, consents, and controls (NCCs). It is infeasible for engineers to understand most or the entirety of the NCCs, when or if they should be checked, and how to implement logic that appropriately complies with them. This disclosure describes techniques that define a mechanism for, and enforce a requirement to, provide structured information about NCCs that data are collected under. NCCs are described in a structured fashion using code. Prior to initiating upload from a client device, the code ascertains if upload of the data is permissible. If permissible, the data upload is allowed and metadata relating to the reasons for data collection is persisted to ensure compliant downstream usage of data. If data upload is not permissible, the upload is prevented, and indicators generated for the appropriate developer team to review

    Predictors of Students’ Achievement in a Blended Learning Course

    Massive Open Online Courses (MOOCs) have been at the center of media attention and hyperbole since 2012. Physics I: Classical Mechanics was a physics course offered by an elite private university in the northeastern United States in the fall of 2014. This course was offered in a blended learning format using a MOOC platform. There were 476 students enrolled in the course, but only 266 of them completed the course. In order to better understand the behavior and academic performance of students in this course, several mathematical models were used to examine the relationship between their grades and their use of the course materials. More specifically, the focus of this paper is the relationship between students’ online problem checking behaviors for questions that required handwritten solutions and their academic performance in the course. All of the data comprising students’ behaviors on the MITx platform are available as a “click stream” log of every action taken by each individual. Several mathematical models were used to create features describing these behavior patterns, and all of these features together were used to build a regression tree, which predicted students’ academic achievement. The results from this model indicated that students who checked their answers more frequently generally received better grades on average. Moreover, students who had more interaction with course materials generally had higher average grades than those who interacted with the platform less. Results also indicated that the group of students whose last problem check was correct received better grades than the other group whose last problem check was not correct. The group of students who started to do the homework earlier in the week also received better grades. The results of this paper are going to help the instructor to build better course outlines and provide students more useful instructions to achieve better grades

    Synaptic Mitochondria Are Critical for Mobilization of Reserve Pool Vesicles at Drosophila Neuromuscular Junctions

    SummaryIn a forward screen for genes affecting neurotransmission in Drosophila, we identified mutations in dynamin-related protein (drp1). DRP1 is required for proper cellular distribution of mitochondria, and in mutant neurons, mitochondria are largely absent from synapses, thus providing a genetic tool to assess the role of mitochondria at synapses. Although resting Ca2+ is elevated at drp1 NMJs, basal synaptic properties are barely affected. However, during intense stimulation, mutants fail to maintain normal neurotransmission. Surprisingly, FM1-43 labeling indicates normal exo- and endocytosis, but a specific inability to mobilize reserve pool vesicles, which is partially rescued by exogenous ATP. Using a variety of drugs, we provide evidence that reserve pool recruitment depends on mitochondrial ATP production downstream of PKA signaling and that mitochondrial ATP limits myosin-propelled mobilization of reserve pool vesicles. Our data suggest a specific role for mitochondria in regulating synaptic strength

    Genomic Inference of the Metabolism and Evolution of the Archaeal Phylum Aigarchaeota

    Microbes of the phylum Aigarchaeota are widely distributed in geothermal environments, but their physiological and ecological roles are poorly understood. Here we analyze six Aigarchaeota metagenomic bins from two circumneutral hot springs in Tengchong, China, to reveal that they are either strict or facultative anaerobes, and most are chemolithotrophs that can perform sulfide oxidation. Applying comparative genomics to the Thaumarchaeota and Aigarchaeota, we find that they both originated from thermal habitats, sharing 1154 genes with their common ancestor. Horizontal gene transfer played a crucial role in shaping genetic diversity of Aigarchaeota and led to functional partitioning and ecological divergence among sympatric microbes, as several key functional innovations were endowed by Bacteria, including dissimilatory sulfite reduction and possibly carbon monoxide oxidation. Our study expands our knowledge of the possible ecological roles of the Aigarchaeota and clarifies their evolutionary relationship to their sister lineage Thaumarchaeota

    Exploring Embeddings for Measuring Text Relatedness: Unveiling Sentiments and Relationships in Online Comments

    After the COVID-19 pandemic caused internet usage to grow by 70%, there has been an increased number of people all across the world using social media. Applications like Twitter, Meta Threads, YouTube, and Reddit have become increasingly pervasive, leaving almost no digital space where public opinion is not expressed. This paper investigates sentiment and semantic relationships among comments across various social media platforms, as well as discusses the importance of shared opinions across these different media platforms, using word embeddings to analyze components in sentences and documents. It allows researchers, politicians, and business representatives to trace a path of shared sentiment among users across the world. This research paper presents multiple approaches that measure the relatedness of text extracted from user comments on these popular online platforms. By leveraging embeddings, which capture semantic relationships between words and help analyze sentiments across the web, we can uncover connections regarding public opinion as a whole. The study utilizes pre-existing datasets from YouTube, Reddit, Twitter, and more. We made use of popular natural language processing models like Bidirectional Encoder Representations from Transformers (BERT) to analyze sentiments and explore relationships between comment embeddings. Additionally, we aim to utilize clustering and Kl-divergence to find semantic relationships within these comment embeddings across various social media platforms.     Male Pattern Baldness in Relation to Prostate Cancer–Specific Mortality: A Prospective Analysis in the NHANES I Epidemiologic Follow-up Study

    We used male pattern baldness as a proxy for long-term androgen exposure and investigated the association of dermatologist-assessed hair loss with prostate cancer–specific mortality in the first National Health and Nutrition Examination Survey Epidemiologic Follow-up Study. From the baseline survey (1971–1974), we included 4,316 men who were 25–74 years of age and had no prior cancer diagnosis. We estimated hazard ratios and used Cox proportional hazards regressions with age as the time metric and baseline hazard stratified by baseline age. A hybrid framework was used to account for stratification and clustering of the sample design, with adjustment for the variables used to calculate sample weights. During follow-up (median, 21 years), 3,284 deaths occurred; prostate cancer was the underlying cause of 107. In multivariable models, compared with no balding, any baldness was associated with a 56% higher risk of fatal prostate cancer (hazard ratio = 1.56; 95% confidence interval: 1.02, 2.37), and moderate balding specifically was associated with an 83% higher risk (hazard ratio = 1.83; 95% confidence interval: 1.15, 2.92). Conversely, patterned hair loss was not statistically significantly associated with all-cause mortality. Our analysis suggests that patterned hair loss is associated with a higher risk of fatal prostate cancer and supports the hypothesis of overlapping pathophysiological mechanisms

    The Proteasome Is a Molecular Target of Environmental Toxic Organotins

    BACKGROUND: Because of the vital importance of the proteasome pathway, chemicals affecting proteasome activity could disrupt essential cellular processes. Although the toxicity of organotins to both invertebrates and vertebrates is well known, the essential cellular target of organotins has not been well identified. We hypothesize that the proteasome is a molecular target of environmental toxic organotins. OBJECTIVES: Our goal was to test the above hypothesis by investigating whether organotins could inhibit the activity of purified and cellular proteasomes and, if so, the involved molecular mechanisms and downstream, events. RESULTS: We found that some toxic organotins [e.g., triphenyltin (TPT)] can potently and preferentially inhibit the chymotrypsin-like activity of purified 20S proteasomes and human breast cancer cellular 26S proteasomes. Direct binding of tin atoms to cellular proteasomes is responsible for the observed irreversible inhibition. Inhibition of cellular proteasomes by TPT in several human cell lines results in the accumulation of ubiquitinated proteins and natural proteasome target proteins, accompanied by induction of cell death. CONCLUSIONS: The proteasome is one of the molecular targets of environmental toxic organotins in human cells, and proteasome inhibition by organotins contributes to their cellular toxicity

    Differential effects of Radix Paeoniae Rubra (Chishao) on cytokine and chemokine expression inducible by mycobacteria

    <p>Abstract</p> <p>Background</p> <p>Upon initial infection with mycobacteria, macrophages secrete multiple cytokines and chemokines, including interleukin-6 (IL-6), IL-8 and tumor necrosis factor-α (TNF-α), to mediate host immune responses against the pathogen. Mycobacteria also induce the production of IL-10 <it>via </it>PKR activation in primary human monocytes and macrophages. As an anti-inflammatory cytokine, over-expression of IL-10 may contribute to mycobacterial evasion of the host immunity. <it>Radix Paeoniae Rubra </it>(RPR, <it>Chishao</it>), a Chinese medicinal herb with potentials of anti-inflammatory, hepatoprotective and neuroprotective effects, is used to treat tuberculosis. This study investigates the immunoregulatory effects of RPR on primary human blood macrophages (PBMac) during mycobacterial infection.</p> <p>Methods</p> <p>The interaction of Bacillus Calmette-Guerin (BCG) with PBMac was used as an experimental model. A series of procedures involving solvent extraction and fractionation were used to isolate bioactive constituents in RPR. RPR-EA-S1, a fraction with potent immunoregulatory effects was obtained with a bioactivity guided fractionation scheme. PBMac were treated with crude RPR extracts or RPR-EA-S1 before BCG stimulation. The expression levels of IL-6, IL-8, IL-10 and TNF-α were measured by qPCR and ELISA. Western blotting was used to determine the effects of RPR-EA-S1 on signaling kinases and transcriptional factors in the BCG-activated PBMac.</p> <p>Results</p> <p>In BCG-stimulated macrophages, crude RPR extracts and fraction RPR-EA-S1 specifically inhibited IL-10 production while enhanced IL-8 expression at both mRNA and protein levels without affecting the expressions of IL-6 and TNF-α. Inhibition of BCG-induced IL-10 expression by RPR-EA-S1 occurred in a dose- and time-dependent manner. RPR-EA-S1 did not affect the phosphorylation of cellular protein kinases including MAPK, Akt and GSK3β. Instead, it suppressed the degradation of IκBα in the cytoplasm and inhibited the translocation of transcription factor NF-κB1 p50 to the nucleus.</p> <p>Conclusion</p> <p>RPR crude extracts and its fraction RPR-EA-S1 inhibited anti-inflammatory cytokine IL-10 and enhanced pro-inflammatory chemokine IL-8 expression in BCG-activated PBMac. The inhibitory effects of RPR-EA-S1 on IL-10 expression in BCG-activated PBMac may be due to the reduced nuclear translocation of NF-κB1 p50.</p
