Extracellular RNA in a single droplet of human serum reflects physiologic and disease states.
Extracellular RNAs (exRNAs) are present in human serum. It remains unclear to what extent these circulating exRNAs may reflect human physiologic and disease states. Here, we developed SILVER-seq (Small Input Liquid Volume Extracellular RNA Sequencing) to efficiently sequence both integral and fragmented exRNAs from a small droplet (5 μL to 7 μL) of liquid biopsy. We calibrated SILVER-seq in reference to other RNA sequencing methods based on milliliters of input serum and quantified droplet-to-droplet and donor-to-donor variations. We carried out SILVER-seq on more than 150 serum droplets from male and female donors ranging from 18 y to 48 y of age. SILVER-seq detected exRNAs from more than a quarter of the human genes, including small RNAs and fragments of mRNAs and long noncoding RNAs (lncRNAs). The detected exRNAs included those derived from genes with tissue (e.g., brain)-specific expression. The exRNA expression levels separated the male and female samples and were correlated with chronological age. Noncancer and breast cancer donors exhibited pronounced differences, whereas donors with or without cancer recurrence exhibited moderate differences in exRNA expression patterns. Even without using differentially expressed exRNAs as features, nearly all cancer and noncancer samples and a large portion of the recurrence and nonrecurrence samples could be correctly classified by exRNA expression values. These data suggest the potential of using exRNAs in a single droplet of serum for liquid biopsy-based diagnostics

Identification and pathogenicity of Aeromonas veronii isolated from sexually mature female Hypophthalmichthys molitrix
An outbreak of bacterial septicemia in female silver carp *Hypophthalmichthys molitrix*, which caused significant death of the fish in the Yantian Reservoir, was investigated. The pathogen was isolated from diseased fish and identified as *Aeromonas veronii* utilizing biochemical characteristics and molecular methods analyses. An artificial infection experiment indicated that the strain caused 100% mortality of juvenile silver carp and mature female fish with eggs but no death of the mature male fish. Silver carp challenged with *A. veronii* showed similar clinical signs with naturally infected fish. The histopathological study revealed that *A. veronii* infection caused the increment of hemosiderin granules and the vacuole formation in tissues of the spleen and livers, as well as the collapse, deformation, and disintegration of egg cells. The ACP, AKP, and CAT enzyme activities in the serum of both naturally and artificially infected silver carp were decreased significantly with the severe infection. In this study, *A. veronii* was isolated and identified as the primary bacterial pathogen causing the mass death of sexually mature female silver carp with eggs in Yantian Reservoir
Output entanglement and squeezing of two-mode fields generated by a single atom

unitary squeezing operator of two-mode fields. Employing the input-output
theory, we find that the entanglement and squeezing of the output fields can be
achieved. By analyzing the squeezing spectrum, we show that asymmetric detuning
and asymmetric atomic initial state split the squeezing spectrum from one
valley into two minimum values, and appropriate leakage of the cavity is needed
for obtaining output entangled fields
Recombinant human insulin-like growth factor-1 promotes osteoclast formation and accelerates orthodontic tooth movement in rats
Background: IGF-1 may be an important factor in bone remodeling, but its mechanism of action on osteoclasts during orthodontic tooth movement is complex and unclear. Methodology: The closed-coil spring was placed between the left maxillary first molar and upper incisors with a force of 50 g to establish an orthodontic movement model. Eighty SD rats were randomized to receive phosphate buffer saline or 400 ng rhIGF-1 in the lateral buccal mucosa of the left maxillary first molar every two days. Tissue sections were stained for tartrate-resistant acidic phosphatase (TRAP), the number of TRAP-positive cells was estimated and tooth movement measured. Results: The rhIGF-1 group exhibited evidential bone resorption and lacuna appeared on the alveolar bone compared to the control group. Moreover, the number of osteoclasts in compression side of the periodontal ligament in the rhIGF-1 group peaked at day 4 (11.37±0.95 compared to 5.28±0.47 in the control group) after the orthodontic force was applied and was significantly higher than that of the control group (p<0.01). Furthermore, the distance of tooth movement in the rhIGF-1 group was significantly larger than that of the control group from day 4 to day 14 (p<0.01), suggesting that rhIGF-1 accelerated orthodontic tooth movement. Conclusion: Our study has showed that rhIGF-1 could stimulate the formation of osteoclasts in the periodontal ligament, and accelerate bone remodeling and orthodontic tooth movement
The Improved Algorithm of Fast Panorama Stitching for Image Sequence and Reducing the Distortion Errors
The traditional image stitching result based on the SIFT feature points extraction, to a certain extent, has distortion errors. The panorama, especially, would get more seriously distorted when compositing a panoramic result using a long image sequence. To achieve the goal of creating a high-quality panorama, the improved algorithm is proposed in this paper, including altering the way of selecting the reference image and putting forward a method that can compute the transformation matrix for any image of the sequence to align with the reference image in the same coordinate space. Additionally, the improved stitching method dynamically selects the next input image based on the number of SIFT matching points. Compared with the traditional stitching process, the improved method increases the number of matching feature points and reduces SIFT feature detection area of the reference image. The experimental results show that the improved method can not only accelerate the efficiency of image stitching processing, but also reduce the panoramic distortion errors, and finally we can obtain a pleasing panoramic result
Screening Quality Evaluation Factors of Freeze-Dried Peach ( Prunus Persica
The quality evaluation of processed products is complex. To simplify the quality evaluation process and improve the efficiency, fourteen evaluation factors of freeze-dried powders of seventeen cultivars of peach at different ripening times were analyzed. The most important evaluation indicators and criteria were obtained by analysis of variance (ANOVA), correlation analysis (CA), principal component analysis (PCA), system cluster analysis (SCA), and analytic hierarchy process (AHP). Results showed that the peach powders had the significant differences in quality (P<0.05), and some processing factors were related with some physicochemical and nutritional factors. Five principle components were extracted by PCA and the cumulative contribution achieved was 84.46%. Through the score plot of the first two principal components, a clear differentiation among ripening times was found and three distinct groups were separated according to ripening time. Five characteristic factors were obtained as titratable acid, browning index, hemicellulose, hygroscopicity, and vitamin C by SCA. Their weights of 0.1249, 0.3007, 0.0514, 0.4916, and 0.0315 were obtained by AHP, respectively. The peach cultivars were divided into four evaluation grades by the comprehensive quality score
Early detection of stroke at the sudden sensorineural hearing loss stage
Background and purposeSudden sensorineural hearing loss (SSNHL) can be a prodromal symptom of ischemic stroke, especially posterior circulation strokes in the anterior inferior cerebellar artery (AICA) area. Early diagnosis and optimal treatment for vascular SSNHL provide an opportunity to prevent more extensive area infarction. The objective of our research was to find clues that suggest stroke at the stage of isolated sudden hearing loss.MethodsWe retrospectively investigated the medical records of patients who received an initial diagnosis of sudden sensorineural hearing loss upon admission from January 2017 to December 2022 at Capital Medical University Affiliated Beijing Tiantan Hospital. Among these patients, 30 individuals who developed acute ischemic stroke during their hospital stay were enrolled as the case group. To create a control group, we matched individuals from the nonstroke idiopathic SSNHL patients to the case group in terms of age (±3 years old) at a ratio of 1:4. We collected the clinical characteristics, pure tone hearing threshold test results, and imaging information for all patients included in the study.ResultsThree models were constructed to simulate different clinical situations and to identify vascular sudden sensorineural hearing loss (SSNHL). The results revealed that patients with SSNHL who had three or more stroke risk factors, bilateral hearing loss, moderately severe to total hearing loss, and any intracranial large artery stenosis and occlusion (≥50%) were at a higher risk of developing ischemic stroke during hospitalization. Consistent with previous studies, the presence of vertigo at onset also played a significant role in the early detection of upcoming stroke.ConclusionClinicians should be alert to SSNHL patients with bilateral hearing loss, moderately severe to total hearing loss and other aforementioned features. Early pure tone audiometric hearing assessment and vascular assessment are necessary for high-risk patients with SSNHL
