    The linkages with fires, vegetation composition and human activity in response to climate changes in the Chinese Loess Plateau during the Holocene

    Holocene paleo-records of the Chinese Loess Plateau loess-soil profiles were used to reconstruct wildfire patterns and landscape evolution. We examine black carbon and charcoal influx, combined with the Magnetic susceptibility, delta C-13 values of soil organic matter, pollen counts and other paleo-environmental proxies to discuss interactions with biomass-climate during the Holocene. The history of fires from the charcoal and black carbon (BC, char and soot) influx at the two sites demonstrates a transition from climate-controlled low amplitude variations with peaks during the Early and Middle Holocene (11-3.1kyearsB.P.) to higher amplitude variability in fire occurrence decoupled from climate and tied to human activities during the Late Holocene (3.1-0kyearsB.P.). The difference in fire patterns was attributed to regional effective moisture and human land use over the entre Loess Plateau; meanwhile, fire activities observed during the Holocene are consistent with variations in vegetation composition inferred from delta C-13 values in soil organic matter, pollen counts, and paleoclimate proxies. Regional wildfires rarely occurred on the desert steppe dominated by a weedy C-3 taxon (Artemisia, Compositae, and Chenopodiaceae dominated)during the late glacial period. A limited biomass would not meet fire propagation in the extreme colder and drier environment of the Loess Plateau during those periods, though. As the climate became ameliorated during the early Holocene, there was an increasing biomass and a sufficient contribution do to high fuel accumulation from C-4 taxon (Gramineae). As the middle Holocene progressed toward warmer and wetter conditions, fire events were less frequent on the steppe and forest-steppe (e.g. expansion of trees C-3,C- Quercus, Corylus) of the Loess Plateau. Subsequently, the number of local and regional fire events have largely increased with the colder and drier climate conditions (e.g. expansion of C-3 weedy), which have been decoupling with intensive anthropogenic burning for farming since the past 3kyr. These data suggests that the regional fire patterns vary strongly along environmental gradients in the effective moisture and regional fuel availability as well as the spatial and temporal distributions of Neolithic burning practices over the Loess Plateau in response to the weakening East Asian monsoon during the Holocene

    Effect of ecological restoration programs on dust concentrations in the North China Plain: a case study

    In recent decades, the Chinese government has made a great effort in initiating large-scale ecological restoration programs (ERPs) to reduce the dust concentrations in China, especially for dust storm episodes. Using the Moderate Resolution Imaging Spectroradiometer (MODIS) land cover product, the ERP-induced land cover changes are quantitatively evaluated in this study. Two obvious vegetation protective barriers arise throughout China from the southwest to the northeast, which are well known as the "Green Great Wall" (GGW). Both the grass GGW and forest GGW are located between the dust source region (DSR) and the densely populated North China Plain (NCP). To assess the effect of ERPs on dust concentrations, a regional transport/dust model (WRF-DUST, Weather Research and Forecast model with dust) is applied to investigate the evolution of dust plumes during a strong dust storm episode from 2 to 8 March 2016. The WRF-DUST model generally performs reasonably well in reproducing the temporal variations and spatial distributions of near-surface [PMC] (mass concentration of particulate matter with aerodynamic diameter between 2.5 and 10 mu m) during the dust storm event. Sensitivity experiments have indicated that the ERP-induced GGWs help to reduce the dust concentration in the NCP, especially in BTH (Beijing, Tianjin, and Hebei). When the dust storm is transported from the upwind DSR to the downwind NCP, the [PMC] reduction ranges from -5 to -15% in the NCP, with a maximum reduction of -12.4% (-19.2 mu gm(3)) in BTH and -7.6% (-10.1 mu g m(3)) in the NCP. We find the dust plumes move up to the upper atmosphere and are transported from the upwind DSR to the downwind NCP, accompanied by dust decrease. During the episode, the forest GGW is nonsignificant in dust concentration control because it is of benefit for dry deposition and not for emission. Conversely, the grass GGW is beneficial in controlling dust erosion and is the dominant reason for [PMC] decrease in the NCP. Because the air pollution is severe in eastern China, especially in the NCP, and the contribution of dust episodes is significant, the reduction of dust concentrations will have important effects on severe air pollution.     Peach allergen Pru p 1 content is generally low in fruit but with large variation in different varieties

    Background: Pru p 1 is a major allergen in peach and nectarine, and the different content in varieties may affect the degree of allergic reactions. This study aimed to quantify Pru p 1 levels in representative peach varieties and select hypoallergenic Pru p 1 varieties. Methods: To obtain monoclonal and polyclonal antibodies, mice and rabbits, respectively, were immunized with recombinant Pru p 1.01 and Pru p 1.02. The Pru p 1 levels in fruits from 83 representative peach varieties was quantified by sandwich enzyme‐linked immunosorbent assay (sELISA). nPru p 1 was obtained through specific monoclonal antibody affinity purification and confirmed by Western blot and mass spectrometry. The variable Pru p 1 content of selected varieties was evaluated by Western blot and the expression level of encoding Pru p 1 genes by quantitative polymerase chain reaction. Results: A sELISA method with monoclonal and polyclonal antibodies was built for quantifying Pru p 1 levels in peach. Pru p 1 was mainly concentrated in the peel (0.20–73.44 μg/g, fresh weight), being very low in the pulp (0.05–9.62 μg/g) and not detected in wild peach. For the 78 peach and nectarine varieties, Pru p 1 content varied widely from 0.12 to 6.45 μg/g in whole fruit. We verified that natural Pru p 1 is composed of 1.01 and 1.02 isoallergens, and the Pru p 1 expression level and Pru p 1 band intensity in the immunoblots were in agreement with protein quantity determined by ELISA for some tested varieties. In some cases, the reduced levels of Pru p 1 did not coincide with low Pru p 3 in the same variety in whole fruit, while some ancient wild peach and nectarines contained low levels of both allergens, and late‐ripening yellow flesh varieties were usually highly allergenic. Conclusion: Pru p 1 content is generally low in peach compared to Pru p 3. Several hypoallergenic Pru p 1 and Pru p 3 varieties, “Zi Xue Tao,” “Wu Yue Xian,” and “May Fire,” were identified, which could be useful in trials for peach allergy patients.info:eu-repo/semantics/publishedVersio

    Decoupled Land and Ocean Temperature Trends in the Early-Middle Pleistocene

    Record of long-term land temperature changes remains ephemeral, discontinuous, and isolated, thus leaving the common view that Pleistocene land temperature evolution should have followed ocean temperatures unconfirmed. Here, we present a continuous land surface temperature reconstruction in the Asian monsoon region over the past 3.0 Myr based on the distribution of soil bacterial lipids from the Chinese Loess Plateau. The land temperature record indicates an unexpected warming trend over the Pleistocene, which is opposite to the cooling trend in Pleistocene ocean temperatures, resulting in increased land-sea thermal contrast. We propose that the previously unrecognized increase of land-sea thermal contrast during much of the Pleistocene is a regional climate phenomenon that provides a likely mechanism in favor of the long-term enhancement of the Pleistocene East Asian summer monsoon

    Nutritional Interventions Improved Rumen Functions and Promoted Compensatory Growth of Growth-Retarded Yaks as Revealed by Integrated Transcripts and Microbiome Analyses

    Growth retardation reduces the incomes of livestock farming. However, effective nutritional interventions to promote compensatory growth and the mechanisms involving digestive tract microbiomes and transcripts have yet to be elucidated. In this study, Qinghai plateau yaks, which frequently suffer from growth retardation due to malnutrition, were used as an experimental model. Young growth-retarded yaks were pastured (GRP), fed basal ration (GRB), fed basal ration addition cysteamine hydrochloride (CSH; GRBC) or active dry yeast (ADY; GRBY). Another group of growth normal yak was pastured as a positive control (GNP). After 60-day nutritional interventions, the results showed that the average daily gain (ADG) of GRB was similar to the level of GNP, and the growth rates of GRBC and GRBY were significantly higher than the level of GNP (P < 0.05). Basal rations addition of CSH or ADY either improved the serum biochemical indexes, decreased serum LPS concentration, facilitated ruminal epithelium development and volatile fatty acids (VFA) fermentation of growth-retarded yaks. Comparative transcriptome in rumen epithelium between growth-retarded and normal yaks identified the differentially expressed genes mainly enriched in immune system, digestive system, extracellular matrix and cell adhesion pathways. CSH addition and ADY addition in basal rations upregulated ruminal VFA absorption (SLC26A3, PAT1, MCT1) and cell junction (CLDN1, CDH1, OCLN) gene expression, and downregulated complement system (C2, C7) gene expression in the growth-retarded yaks. 16S rDNA results showed that CSH addition and ADY addition in basal rations increased the rumen beneficial bacterial populations (Prevotella_1, Butyrivibrio_2, Fibrobacter) of growth-retarded yaks. The correlation analysis identified that ruminal VFAs and beneficial bacteria abundance were significantly positively correlated with cell junction and VFA absorption gene expressions and negatively correlated with complement system gene expressions on the ruminal epithelium. Therefore, CSH addition and ADY addition in basal rations promoted rumen health and body growth of growth-retarded yaks, of which basal ration addition of ADY had the optimal growth-promoting effects. These results suggested that improving nutrition and probiotics addition is a more effective method to improve growth retardation caused by gastrointestinal function deficiencies

    The genome sequence of Streptomyces rochei 7434AN4, which carries a linear chromosome and three characteristic linear plasmids

    Streptomyces rochei 7434AN4 produces two structurally unrelated polyketide antibiotics, lankacidin and lankamycin, and carries three linear plasmids, pSLA2-L (211 kb), -M (113 kb), and -S (18 kb), whose nucleotide sequences were previously reported. The complete nucleotide sequence of the S. rochei chromosome has now been determined using the long-read PacBio RS-II sequencing together with short-read Illumina Genome Analyzer IIx sequencing and Roche 454 pyrosequencing techniques. The assembled sequence revealed an 8,364,802-bp linear chromosome with a high G + C content of 71.7% and 7,568 protein-coding ORFs. Thus, the gross genome size of S. rochei 7434AN4 was confirmed to be 8,706,406 bp including the three linear plasmids. Consistent with our previous study, a tap-tpg gene pair, which is essential for the maintenance of a linear topology of Streptomyces genomes, was not found on the chromosome. Remarkably, the S. rochei chromosome contains seven ribosomal RNA (rrn) operons (16S-23S-5S), although Streptomyces species generally contain six rrn operons.
