    Photoacoustic signal enhancement in dual-contrast gastrin-releasing peptide receptor-targeted nanobubbles

    Translatable imaging agents are a crucial element of successful molecular imaging. Photoacoustic molecular imaging relies on optical absorbing materials to generate a sufficient signal. However, few materials approved for human use can generate adequate photoacoustic responses. Here we report a new nanoengineering approach to further improve photoacoustic response from biocompatible materials. Our study shows that when optical absorbers are incorporated into the shell of a gaseous nanobubble, their photoacoustic signal can be significantly enhanced compared to the original form. As an example, we constructed nanobubbles using biocompatible indocyanine green (ICG) and biodegradable poly(lactic-co-glycolic acid) (PLGA). We demonstrated that these ICG nanobubbles generate a strong ultrasound signal and almost four-fold photoacoustic signal compared to the same concentration of ICG solution; our theoretical calculations corroborate this effect and elucidate the origin of the photoacoustic enhancement. To demonstrate their molecular imaging performance, we conjugated gastrin-releasing peptide receptor (GRPR) targeting ligands with the ICG nanobubbles. Our dual photoacoustic/ultrasound molecular imaging shows a more than three-fold enhancement in targeting specificity of the GRPR-targeted ICG nanobubbles, compared to untargeted nanobubbles or prostate cancer cells not expressing GRPR, in a prostate cancer xenograft mouse model in vivo

    Does PGA external stenting reduce compliance mismatch in venous grafts?

    BACKGROUND: Autogenous vein grafting is widely used in regular bypassing procedures. Due to its mismatch with the host artery in both mechanical property and geometry, the graft often over expands under high arterial blood pressure and forms a step-depth where eddy flow develops, thus causing restenosis, fibrous graft wall, etc. External stents, such as sheaths being used to cuff the graft, have been introduced to eliminate these mismatches and increase the patency. Although histological and immunochemical studies have shown some positive effects of the external stent, the mechanical mismatch under the protection of an external stent remains poorly analyzed. METHODS: In this study, the jugular veins taken from hypercholesterolemic rabbits were transplanted into the carotid arteries, and non-woven polyglycolic acid (PGA) fabric was used to fabricate the external stents to study the effect of the biodegradable external stent. Eight weeks after the operation, the grafts were harvested to perform mechanical tests and histological examinations. An arc tangent function was suggested to describe the relationship between pressure and cross-sectional area to analyse the compliance of the graft. RESULTS: The results from the mechanical tests indicated that grafts either with or without external stents displayed large compliance in the low-pressure range and were almost inextensible in the high-pressure range. This was very different from the behavior of the arteries or veins in vivo. The data from histological tests showed that, with external stents, collagen fibers were more compact, whilst those in the graft without protection were looser and thicker. No elastic fiber was found in either kind of grafts. Furthermore, grafts without protection were over-expanded which resulted in much bigger cross-sectional areas. CONCLUSION: The PGA external extent contributes little to the reduction of the mechanical mismatch between the graft and its host artery while remodeling develops. For the geometric mismatch, it reduces the cross-section area, therefore matching with the host artery much better.     The Online Data Quality Monitoring System at BESIII

    The online Data Quality Monitoring (DQM) plays an important role in the data taking process of HEP experiments. BESIII DQM samples data from online data flow, reconstructs them with offline reconstruction software, and automatically analyzes the reconstructed data with user-defined algorithms. The DQM software is a scalable distributed system. The monitored results are gathered and displayed in various formats, which provides the shifter with current run information that can be used to find problems early.     The links between ecosystem multifunctionality and above- and belowground biodiversity are mediated by climate

    Plant biodiversity is often correlated with ecosystem functioning in terrestrial ecosystems. However, we know little about the relative and combined effects of above-and belowground biodiversity on multiple ecosystem functions (for example, ecosystem multifunctionality, EMF) or how climate might mediate those relationships. Here we tease apart the effects of biotic and abiotic factors, both above- and belowground, on EMF on the Tibetan Plateau, China. We found that a suite of biotic and abiotic variables account for up to 86% of the variation in EMF, with the combined effects of above-and belowground biodiversity accounting for 45% of the variation in EMF. Our results have two important implications: first, including belowground biodiversity in models can improve the ability to explain and predict EMF.     Fabricating core-shell WC@C/Pt structures and its enhanced performance for methanol electrooxidation

The spray-dried spheres within a W/Pt multi-separation can be used to prepare discrete core-shell WC@C/Pt catalysts through a typical carburization production mechanism at 800 °C. In contrast with previous studies of the WC/Pt synthesis, the reaction observed here proceeds through an indirect annealing mechanism at 600 °C wherein species diffuse, thereby resulting in core-shell structure, and Pt nanoparticles were successfully dispersed in size/shape and randomly scattered across the in situ produced C spheres. Through direct carburization or at higher initial hydrochloroplatinic acid concentrations, however, complete reaction with core-shell spheres was not observed. Indirect carburization reduces the strain felt by the bonds featuring the larger WC particles and allows the motion of carbon around WC and Pt nanoparticles to be reserved, influencing the electrocatalytic performance and stability toward methanol oxidation

    On the basis of similar structure of solution for a second-order linear differential equation's boundary value problem, and our analysis of the relationship between this similar structure and its kernel function, the differential equation, and the boundary conditions, we propose a new simple solution—similar constructive method of solution (SCMS)—and sum up its detailed steps. We set up a porous media model under three kinds of outer boundary conditions (infinite, constant pressure, and closed), in which the influences of fractal dimension, spherical flow, skin effect, and storage are taken into consideration. And then we use SCMS to solve it. The SCMS is a straightforward method for differential equation's boundary value problems with complex boundary conditions, especially for solving the reservoir models in petroleum engineering

    Evaluation of ferritin and TfR level in plasma neural-derived exosomes as potential markers of Parkinson’s disease

    IntroductionEarly diagnosis of Parkinson’s disease (PD) remains challenging. It has been suggested that abnormal brain iron metabolism leads to excessive iron accumulation in PD, although the mechanism of iron deposition is not yet fully understood. Ferritin and transferrin receptor (TfR) are involved in iron metabolism, and the exosome pathway is one mechanism by which ferritin is transported and regulated. While the blood of healthy animals contains a plentiful supply of TfR-positive exosomes, no studies have examined ferritin and TfR in plasma neural-derived exosomes.MethodsPlasma exosomes were obtained from 43 patients with PD and 34 healthy controls. Neural-derived exosomes were isolated with anti-human L1CAM antibody immunoabsorption. Transmission electron microscopy and western blotting were used to identify the exosomes. ELISAs were used to quantify ferritin and TfR levels in plasma neural-derived exosomes of patients with PD and controls. Receivers operating characteristic (ROC) curves were applied to map the diagnostic accuracy of ferritin and TfR. Independent predictors of the disease were identified using logistic regression models.ResultsNeural-derived exosomes exhibited the typical exosomal morphology and expressed the specific exosome marker CD63. Ferritin and TfR levels in plasma neural-derived exosomes were significantly higher in patients with PD than controls (406.46 ± 241.86 vs. 245.62 ± 165.47 ng/μg, P = 0.001 and 1728.94 ± 766.71 vs. 1153.92 ± 539.30 ng/μg, P < 0.001, respectively). There were significant positive correlations between ferritin and TfR levels in plasma neural-derived exosomes in control group, PD group and all the individuals (rs = 0.744, 0.700, and 0.752, respectively). The level of TfR was independently associated with the disease (adjusted odds ratio 1.002; 95% CI 1.000–1.003). ROC performances of ferritin, TfR, and their combination were moderate (0.730, 0.812, and 0.808, respectively). However, no relationship was found between the biomarkers and disease progression.ConclusionIt is hypothesized that ferritin and TfR in plasma neural-derived exosomes may be potential biomarkers for PD, and that they may participate in the mechanism of excessive iron deposition in PD

    The LAMOST Survey of Background Quasars in the Vicinity of the Andromeda and Triangulum Galaxies -- II. Results from the Commissioning Observations and the Pilot Surveys

    We present new quasars discovered in the vicinity of the Andromeda and Triangulum galaxies with the LAMOST during the 2010 and 2011 observational seasons. Quasar candidates are selected based on the available SDSS, KPNO 4 m telescope, XSTPS optical, and WISE near infrared photometric data. We present 509 new quasars discovered in a stripe of ~135 sq. deg from M31 to M33 along the Giant Stellar Stream in the 2011 pilot survey datasets, and also 17 new quasars discovered in an area of ~100 sq. deg that covers the central region and the southeastern halo of M31 in the 2010 commissioning datasets. These 526 new quasars have i magnitudes ranging from 15.5 to 20.0, redshifts from 0.1 to 3.2. They represent a significant increase of the number of identified quasars in the vicinity of M31 and M33. There are now 26, 62 and 139 known quasars in this region of the sky with i magnitudes brighter than 17.0, 17.5 and 18.0 respectively, of which 5, 20 and 75 are newly-discovered. These bright quasars provide an invaluable collection with which to probe the kinematics and chemistry of the ISM/IGM in the Local Group of galaxies. A total of 93 quasars are now known with locations within 2.5 deg of M31, of which 73 are newly discovered. Tens of quasars are now known to be located behind the Giant Stellar Stream, and hundreds behind the extended halo and its associated substructures of M31. The much enlarged sample of known quasars in the vicinity of M31 and M33 can potentially be utilized to construct a perfect astrometric reference frame to measure the minute PMs of M31 and M33, along with the PMs of substructures associated with the Local Group of galaxies.
