    Origin Identification and Quantitative Analysis of Honeys by Nuclear Magnetic Resonance and Chemometric Techniques

    The combination of H-1 NMR spectroscopy and multivariate statistical analysis has become a promising method for the discrimination of food origins. In this paper, this method has been successfully employed to analyze 70 Chinese honey samples from eight botanic origins, three geographical origins, and five production dates. Thirty-three components in honey samples were detected and identified from their H-1 NMR spectra, and 20 of them were accurately quantified by comparing their integral area with that of internal standards with relaxation time correction. Nontargeted principal component analysis (PCA) has been applied to distinguish the honeys from different botanical and geographical origins. The variations of components in the honeys, including saccharides and all kind of amino and organic carboxylic acids, confirmed their clustering according to their origins in PCA scores plots. Orthogonal partial least squares discriminant analysis (OPLS-DA) based on the NMR data for the different pairwise honey samples allows to identify the compositional variations contributed to geographical discrimination and storage time. Hence, NMR spectroscopy coupled with chemometric techniques offers an efficient tool for quality control of honey, and it could further serve to the classification, qualitative and quantitative control of other foods

    Penyakit kuning pada kacang panjang berdampak pada penurunan produksi. Gejala serangan diawali dari gejala daun keriting serta mengakibatkan polong berwarna kuning. Penelitian ini bertujuan mengetahui nilai heritabilitas dan ragam genetik serta menduga jumlah gen pengendali ketahanan kacang panjang terhadap penyakit kuning. Penelitian dilaksanakan di Kabupaten Kediri pada bulan April sampai Juli 2013. Bahan penelitian adalah populasi UB 715 A (P1), Hitam Putih (P2), populasi F1 dan populasi F2. Berdasarkan hasil penelitian, populasi UB 715 A (P1 ) menunjukkan respon tahan terhadap penyakit kuning, populasi Hitam Putih (P2) menunjukkan respon rentan, dan populasi F1 dan F2 menunjukkan respon sedang. Karakter jumlah polong dan jumlah biji per tanaman memiliki keragaman yang sempit sedangkan karakter panjang polong, bobot segar polong, umur berbunga, dan umur panen memiliki keragaman yang luas. Karakter panjang polong dan jumlah biji per polong memiliki nilai heritabilitas rendah, sedangkan karakter jumlah polong, bobot segar polong, umur berbunga, dan umur panen memiliki nilai heritabilitas tinggi. Rasio sifat ketahanan terhadap penyakit kuning pada populasi F2 adalah 9 tahan : 3 sedang : 4 rentan yang berarti ketahanan terhadap penyakit kuning dikendalikan oleh dua gen dengan aksi gen epistasis resesif

However, many details of the molecular mechanisms that regulate fish oogenesis remain elusive. Here, we performed a single-cell RNA sequencing (scRNA-seq) analysis of the molecular signatures of distinct ovarian cell categories in adult Chinese tongue sole (Cynoglossus semilaevis). We characterized the successive stepwise development of three germ cell subtypes. Notably, we identified the cellular composition of fish follicle walls, including four granulosa cell types and one theca cell type, and we proposed important transcription factors (TFs) showing high activity in the regulation of cell identity. Moreover, we found that the extensive niche–germline bidirectional communications regulate fish oogenesis, whereas ovulation in fish is accompanied by the coordination of simultaneous and tightly sequential processes across different granulosa cells. Additionally, a systems biology analysis of the homologous genes shared by Chinese tongue sole and macaques revealed remarkably conserved biological processes in germ cells and granulosa cells across vertebrates.     Protocol for a Single-Center Randomized Controlled Trial of Percutaneous Coronary Intervention Via Distal Transradial Access Versus Transradial Access

    Background: Although transradial access (TRA) has become the main vascular access for coronary intervention, its high radial artery occlusion rate limits its application in some patients. Studies have shown that compared with TRA, distal transradial access (dTRA) with the snuffbox area or the Hegu acupoint area as the puncture point significantly decreases the incidence of radial artery occlusion. However, no randomized controlled study has confirmed the safety and efficacy of coronary artery intervention via dTRA in China. Methods and analyses: This single-center, prospective, randomized controlled, superiority open-label study will enroll 428 consecutive patients with coronary heart disease undergoing percutaneous coronary intervention as the study population. After preoperative evaluation, the participants will be randomly divided into a study group (dTRA) and control group (TRA) in a 1:1 ratio. The primary endpoint (radial artery occlusion at 24 hours after operation) and secondary endpoint events will be evaluated and recorded.     A novel single nucleotide polymorphism within the NOD2 gene is associated with pulmonary tuberculosis in the Chinese Han, Uygur and Kazak populations

    <p>Abstract</p> <p>Background</p> <p>The present study aimed to investigate the genetic polymorphisms in exon 4 of the <it>NOD2 </it>gene in tuberculosis patients and healthy controls, in order to clarify whether polymorphisms in the <it>NOD2 </it>gene is associated with tuberculosis.</p> <p>Methods</p> <p>A case-control study was performed on the Chinese Han, Uygur and Kazak populations. Exon 4 of the <it>NOD2 </it>gene was sequenced in 425 TB patients and 380 healthy controls to identify SNPs.</p> <p>Results</p> <p>The frequency of T/G genotypes for the Arg587Arg (CGT → CGG) single nucleotide polymorphism (SNP) in <it>NOD2 </it>was found to be significantly higher in the Uygur (34.9%) and Kazak (37.1%) populations than the Han population (18.6%). Also, the frequency of G/G genotypes for the Arg587Arg SNP was significantly higher in the Uyghur (8.3%) and Kazak (5.4%) populations than the Han population (0.9%). Meanwhile, no significant difference was found in the Arg587Arg polymorphism between the tuberculosis patients and healthy controls in the Uyghur and Kazak populations (<it>P </it>> 0.05) whereas, a significant difference was observed in the Arg587Arg polymorphism between the tuberculosis patients and healthy controls in the Han population (<it>P </it>< 0.01). The odd ratio of 2.16 (95% CI = 1.31-3.58; <it>P </it>< 0.01) indicated that the Arg587Arg SNP in <it>NOD2 </it>may be associated with susceptibility to tuberculosis in the Chinese Han population.</p> <p>Conclusions</p> <p>Our study is the first to demonstrate that the Arg587Arg SNP in <it>NOD2 </it>is a new possible risk factor for tuberculosis in the Chinese Han population, but not in the Uyghur and Kazak populations. Our results may reflect racial differences in genetic susceptibility to tuberculosis.</p

    Fatigue Cracking Resistance of Engineered Cementitious Composites (ECC) under Working Condition of Orthotropic Steel Bridge Decks Pavement

    In order to investigate the fatigue cracking resistance of engineered cementitious composites (ECC) used in in total life pavement, the semi-circular bending (SCB) test and improved three-point bending fatigue test (ITBF) were utilized in this study. The digital image correlation (DIC) method was also utilized to track the surface strain fields of specimens during the SCB test. X-ray computed tomography (CT) and digital image processing (DIP) technologies were applied to measure the internal-crack distribution of the ITBF specimen. The results of the SCB test showed that the fatigue cracking damage process of ECC can be divided into three stages and that the cracking stable propagating stages occupied the main part, which indicates that ECC has excellent ductility and toughness and could work very well with existing cracks. The ITBF results showed that the fatigue cracking resistance of ECC was better than epoxy asphalt concrete (EAC). In addition, the internal-crack distribution along the depth direction of the ITBF specimen could be presented well by the image pixel statistical (IPS) method based on CT scanning of image slices. It could be found that multiple cracks propagate simultaneously in ECC, instead of a single crack, under the OSBD pavement working condition

    Database server workload characterization in an e-commerce environment

    In an E-commerce system, the database server performance is crucial. Dynamic cache is often used to reduce the load on the database server, which reduces the need for scalability. A good understanding of the workload characteristics of the database server in an E-commerce environment is important to the design, tuning, and capacity planning of the database server. In this paper, we characterize the database server workloads in a benchmark E-commerce system. We focus on the response time, CPU utilization, the database page reference characteristics, and disk I/Os of the database server. We found that using dynamic cache can substantially reduce the CPU utilization but not always the number of disk I/Os of the database server. In most cases, using dynamic cache reduces temporal locality in database page references, but to a smaller degree than that reported in file servers and web proxies. Interestingly, in certain Ecommerce workloads, using dynamic cache results in better temporal locality. 1

    Numerical Simulation of Cavitation Characteristics of a Centrifugal Pump Based on an Improved ZGB Model

    We modified the Zwart–Gerber–Belamri (ZGB) cavitation and RNG turbulence models based on their rotational motion characteristics, as well as simulating the phenomenon of small fluctuations in rotational speed due to the action of hydraulic excitation force, to increase the precision of numerical simulations of the cavitation characteristics of centrifugal pumps. According to the theory behind the enhanced model, the pressure gradient in the impeller runner changes uniformly, and the cavitation bubble initially manifests itself at the front edge of the blade’s suction. With the reduction in the effective margin, changes in the impeller flow channel’s pressure gradient increased, the blade’s suction-side cavitation area expanded, and the flow field’s internal disturbance enhanced, resulting in hydraulic loss to the centrifugal pump, considerably reduced operating performance, and the blade pressure side by the cavitation-affected area becoming smaller. The blade’s suction-surface pressure–load curve can be divided into a low-pressure cavitation zone, a pressure fluctuation zone, and a pressure stabilization zone. The pressure–frequency-domain diagram of each monitoring point has a discrete distribution, and the main frequency is the blade-passing frequency ƒ = 293 Hz and its n times frequency. With the deepening of the cavitation, the amplitude of the main frequency in the low-frequency band showed a trend of increasing and then decreasing, and the amplitude of the high-frequency band showed an increasing trend. When we applied the ZGB cavitation model based on the improved rotational motion characteristics to our numerical calculation of the centrifugal pumps’ cavitation characteristics, our calculated cavitation performance curve was closer to the experimental value than the ZGB model, and when the centrifugal pump’s cavitation degree was increased, the area covered by the vacuole area was 1/4 of the suction side of the vane

    Sustainable Urban Street Comprising Permeable Pavement and Bioretention Facilities: A Practice

    Roadside bioretention and permeable pavements have proven effectiveness in rainwater filtration and waterlogging mitigation, but conventional street design approach could not accommodate their work in conjunction. In this research, possible roadside facilities allowing water transmission from permeable pavements and bioretention to the pipe system are proposed. Hydraulic properties of the comprised elements were analyzed, including rainfall intensity, permeable pavements, soil layers and pipe systems. A transformation method was formulated to obtain a successive time-intensity formula from conventional design parameters to describe the rainfall behavior, and therefore the water retention capacity of the bioretention could be considered. A test section of 1.6 km combining permeable pavements and roadside bioretention was constructed, and its hydraulic performance was predicted based on the proposed design method and Storm Water Management Model (SWMM). The research results suggest that the bioretention facilities and permeable pavements cooperate well in the test section. In a light rain event, the proposed street has favorable performance in rainwater collection and filtration. In a relatively intense rainstorm event, the street collects and filters water in the initial stage, but will have similar hydraulic performance to a conventional street once the retention facilities are saturated. Thus, no reduction in diameters of drainage pipes from conventional designs is suggested in similar projects
