    AbstractFor a connected graph G=(V,E), an edge set S⊂E is a 3-restricted edge cut if G−S is disconnected and every component of G−S has order at least three. The cardinality of a minimum 3-restricted edge cut of G is the 3-restricted edge connectivity of G, denoted by λ3(G). A graph G is called minimally 3-restricted edge connected if λ3(G−e)<λ3(G) for each edge e∈E. A graph G is λ3-optimal if λ3(G)=ξ3(G), where ξ3(G)=max{ω(U):U⊂V(G),G[U] is connected,|U|=3}, ω(U) is the number of edges between U and V∖U, and G[U] is the subgraph of G induced by vertex set U. We show in this paper that a minimally 3-restricted edge connected graph is always λ3-optimal except the 3-cube

    B Cell Activation in Insulin Resistance and Obesity

    Our group has demonstrated that inflammatory diseases such as type 2 diabetes (DM), inflammatory bowel disease (IBD), and periodontal disease (PD) are associated with altered B cell function that may contribute to disease pathogenesis. B cells were found to be highly activated with characteristics of inflammatory cells. Obesity is a pre-disease state for cardiovascular disease and type 2 diabetes and is considered a state of chronic inflammation. Therefore, we sought to better characterize B cell function and phenotype in obese patients. We demonstrate that (Toll-like receptor) TLR4 and CD36 expression by B cells is elevated in obese subjects, suggesting increased sensing of lipopolysaccharide (LPS) and other TLR ligands. These ligands may be of microbial, from translocation from a leaky gut, or host origin. To better assess microbial ligand burden and host response in the bloodstream, we measured LPS binding protein (LBP), bacterial/permeability increasing protein (BPI), and high mobility group box 1 (HMGB1). Thus far, our data demonstrate an increase in LBP in DM and obesity indicating increased responses to TLR ligands in the blood. Interestingly, B cells responded to certain types of LPS by phosphorylating extracellular-signal-regulated kinases (ERK) 1/2. A better understanding of the immunological state of obesity and the microbial and endogenous TLR ligands that may be activating B cells will help identify novel therapeutics to reduce the risk of more dangerous conditions, such as cardiovascular disease

    Mechanical Performance of Simple Supported Concrete Beam-Cable Composite Element with External Prestress

    A new reinforcement technology of external prestress based on stretch tilted belly poles has been presented. Taking simply supported beam, which is reinforced by three titled belly poles, as a research object to establish a model of reinforced simply supported beam. Relationship expressions about deflection and internal force increment of external cable or about load and deflection have been deduced. Finite element model is established by ABAQUS. The influence of structure performance of reinforced simply supported beam with cable section, cable sag and initial internal force value was investigated. Three tests are carried out to testify the results of theoretical analysis and numerical simulation. The results show that the redistributions of internal force and sectional stress have occurred, and the stiffness, crack load, ultimate load, and structure ductility are all improved with the increase of three design parameters. For example, the crack load, ultimate load, and structure ductility have increased, respectively, by 24%~40%, 15%~42%, and 14%~40%. High initial internal force, small section, and big cable sag should be avoided, because the probability of brittle failure of structure will increase. The analytical result shows that the reliability of internal increment expression of external cable and carrying capacity expression can be used in the engineering practice

    Molecular Evolution of the Deuterolysin (M35) Family Genes in Coccidioides

    Coccidioides is a primary fungal pathogen of humans, causing life-threatening respiratory disease known as coccidioidomycosis (Valley fever) in immunocompromised individuals. Recently, Sharpton et al (2009) found that the deuterolysin (M35) family genes were significantly expanded in both the Coccidioides genus and in U. reesii, and that Coccidioides has acquired three more M35 family genes than U. reesii. In the present work, phylogenetic analyses based on a total of 28 M35 family genes using different alignments and tree-building methods consistently revealed five clades with high nodal supports. Interestingly, likelihood ratio tests suggested significant differences in selective pressure on the ancestral lineage of three additional duplicated M35 family genes from Coccidioides species compared to the other lineages in the phylogeny, which may be associated with novel functional adaptations of M35 family genes in the Coccidioides species, e.g., recent pathogenesis acquisition. Our study adds to the expanding view of M35 family gene evolution and functions as well as establishes a theoretical foundation for future experimental investigations

    Characteristics of CD8+ T cell subsets in Chinese patients with chronic HIV infection during initial ART

    <p>Abstract</p> <p>Background</p> <p>CD8+ T cells may play an important role in protecting against HIV. However, the changes of CD8+ T cell subsets during early period of ART have not been fully studied.</p> <p>Methods</p> <p>Twenty-one asymptomatic treatment-naive HIV-infected patients with CD4 T+ cells less than 350 cells/μl were enrolled in the study. Naïve, central memory(CM), effective memory(EM) and terminally differentiated effector (EMRA) CD8+ cell subsets and their activation and proliferation subsets were evaluated in blood samples collected at base line, and week 2, 4, 8 and 12 of ART.</p> <p>Results</p> <p>The total CD8+ T cells declined and the Naïve and CM subsets had a tendency of increase. Activation levels of all CD8+ T cell subsets except EMRA subset decreased after ART. However, proliferation levels of total CD8+ T cells, EMRA, EM and CM subsets increased at the first 4 weeks of ART, then decreased. Proliferation level of the naïve cells decreased after ART.</p> <p>Conclusion</p> <p>The changes of CD8+ T cell subsets during initial ART are complex. Our results display a complete phenotypical picture of CD8+ cell subsets during initial ART and provide insights for understanding of immune status during ART.</p
