Simulation model to determine ratios between quay, yard and intra-terminal transfer equipment in an integrated container handling system
This paper presents a generic simulation model to determine the equipment mix (quay, yard and intra-terminal transfer) for a Container Terminal Logistics Operations System (CTLOS). The simulation model for the CTLOS, a typical type of discrete event dynamic system (DEDS), consists of three sub-models: ship queue, loading-unloading operations and yard-gate operations. The simulation model is empirically applied to phase 1 of the Yangshan Deep Water Port in Shanghai. This study considers different scenarios in terms of container throughput levels, equipment utilization rates, and operational bottle-necks, and presents a sensitivity analysis to evaluate and choose reasonable equipment ratio ranges under different operational conditions
Analysis of nonlinear suspension power harvest potential
Because the power consumption of a controlled suspension is huge, the power harvest potential of a nonlinear controlled suspension is analyzed. Instead of simplifying the suspension to a linear model or adopting some control strategies to solve the problem, this paper investigates the effect of the nonlinear characteristics on the power harvesting potential. A mathematic model is introduced to calculate the nonlinear vibration, and the amount of harvested power was obtained using the multi-scale method. A numerical validation is carried out at the end of this study. The results show that the investigated mechanical parameters affect both the vibration amplitude and the induced current, while the electric parameters only affect the induced current. The power harvesting potential of the nonlinear suspension is generally greater than the linear suspension because the frequency band of the actual pavement also contains bandwidth surrounding the body resonance point. The only exception occurs if the vehicle travels on a road with a particular profile, e.g. a sine curve. To optimize harvested power, it is better to consider the nonlinear characteristics rather than simplifying the suspension to a linear model

Prompting Fab Yeast Surface Display Efficiency by ER Retention and Molecular Chaperon Co-expression.
For antibody discovery and engineering, yeast surface display (YSD) of antigen-binding fragments (Fabs) and coupled fluorescence activated cell sorting (FACS) provide intact paratopic conformations and quantitative analysis at the monoclonal level, and thus holding great promises for numerous applications. Using anti-TNFα mAbs Infliximab, Adalimumab, and its variants as model Fabs, this study systematically characterized complementary approaches for the optimization of Fab YSD. Results suggested that by using divergent promoter GAL1-GAL10 and endoplasmic reticulum (ER) signal peptides for co-expression of light chain and heavy chain-Aga2 fusion, assembled Fabs were functionally displayed on yeast cell surface with sigmoidal binding responses toward TNFα. Co-expression of a Hsp70 family molecular chaperone Kar2p and/or protein-disulfide isomerase (Pdi1p) significantly improved efficiency of functional display (defined as the ratio of cells displaying functional Fab over cells displaying assembled Fab). Moreover, fusing ER retention sequences (ERSs) with light chain also enhanced Fab display quality at the expense of display quantity, and the degree of improvements was correlated with the strength of ERSs and was more significant for Infliximab than Adalimumab. The feasibility of affinity maturation was further demonstrated by isolating a high affinity Fab clone from 1:103 or 1:105 spiked libraries
Full-length enriched multistage cDNA library construction covering floral bud development in Populus tomentosa
Flowering involves expression of a suite of genes associated with floral development. The genome of the Chinese white poplar (Populus trichocarpa) was sequenced because of its importance as a model tree for genetic studies as well as being an economically important woody plant. However, information on expressed genes involved in poplar floral bud development is insufficient to allow annotation of genes and use of the genomic information. To isolate and characterize genes involved in flowering of Populus tomentosa, floral bud samples were collected at different developmental stages from floral bud initiation to flower maturity, and full-length enriched cDNA libraries from both male and female floral buds were constructed. The results of titer analysis showed that the titer of the female and male primary libraries were 8.00 × 105 and 7.20 × 105 pfu/ml, respectively, and the titer of the amplified libraries were 2.60 × 108 and 2.56 × 108 pfu/ml, respectively. The combination ratio reached 90% and the insert size was 400 to 2000 bp. In the structure of the title compound, 2[Ag(C10H8N2)(H2O)](C7H6NO2)(NO3)·H2O, the AgI atom is three-coordinated in a T-shaped configuration by two N atoms from two symmetry-related 4,4′-bipyridine (bipy) ligands at short distances and by one water O atom at a longer distance. Each bipy ligand bridges two neighbouring AgI atoms, forming a chain structure extending parallel to [101]. The complete 4-aminobenzoate anion, the nitrate anion and the uncoordinated water molecule are located on mirror planes: together with the coordinated water molecule, they form N—H⋯O, O—H⋯O and O—H⋯N hydrogen bonds, stabilizing the crystal structure
Suan Zao Ren Tang
Insomnia is a serious worldwide health problem that is often comorbid with anxiety. The purpose of the present study was to evaluate the efficacy of a Chinese formula containing Suan Zao Ren Tang (SZRT) and Zhi Zi Chi Tang (ZZCT; SZR-ZZC) for improving sleep quality and anxiety states with four indices of Polysomnography (PSG), the Insomnia Severity Index (ISI), the Pittsburgh Sleep Quality Index (PSQI), and the Self Rating Anxiety Scale (SAS). Methods. A randomized, parallel-controlled trial compared SZR-ZZC to lorazepam tablet in insomniacs with anxiety. Patients were randomized to the SZR-ZZC treatment group (n=60) and the lorazepam tablet treatment group (n=59). Results. SZR-ZZC significantly improved scores on all four treatment indices. Compared with lorazepam, treatment with SZR-ZZC resulted in a significant reduction in the ISI (P=0.029), the PSQI (P=0.017), and wake after sleep onset (WASO; P=0.008) scores and improved sleep architecture (P=0.000–0.003) after a 4-week treatment period. Only one subject in the SZR-ZZC group experienced adverse side effects. Conclusion. Treatment with SZR-ZZC for 4 weeks appears to be a relatively safe and effective complementary therapeutic option when aiming to improve sleep quality and anxiety in insomniacs with anxiety
