    Why Is Maternal Control Harmful? The Relation between Maternal Control, Insecure Attachment and Antisocial Personality Disorder Features in Chinese College Students:A Sequential Mediation Model

    Background: Previous work has indicated that a negative parenting style is associated with antisocial personality disorder features in Chinese college students, yet few studies have explored the unique role of negative mothering in children’s antisocial personality disorder. Methods: The current study mainly examined the sequential mediation effect of parental antipathy and neglect (PAN) and mother negative loving (a form of insecure attachment) in the association between mother control and adulthood antisocial personality disorder features (ASPD features) in the framework of attachment theory and cognitive–behavioral theory. A community sample of 1547 Chinese college students filled in the Parental Bonding Instrument, the Childhood Experience of Care and Abuse Questionnaire, the Adult Attachment Questionnaire and the Personality Diagnostic Questionnaire-4+. Results: A sequential mediation model analysis showed that maternal control significantly predicted PAN, mother negative loving, as well as ASPD features. Conclusions: Mother control and mother negative loving appear to advance on the development and exacerbation of ASPD features in college students

    Limit Equilibrium Method-Based Shear Strength Prediction for Corroded Reinforced Concrete Beam with Inclined Bars

    Shear strength is a widely investigated parameter for reinforced concrete structures. The corrosion of reinforcement results in shear strength reduction. Corrosion has become one of the main deterioration factors in reinforced concrete beam. This paper proposes a shear strength model for beams with inclined bars based on a limit equilibrium method. The proposed model can be applied to both corroded and uncorroded reinforced concrete beams. Besides the tensile strength of longitudinal steel bars, the shear capacity provided by the concrete on the top of the diagonal crack, the tensile force of the shear steel at the diagonal crack, the degradation of the cross-sectional area and strength of the reinforcements induced by corrosion are all considered. An experimental work on two groups accelerated corroded beams was performed. Good agreements were found between the proposed theoretical predictions and experimental observations

    Isolation and characterization of α-amylase from marine Pseudomonas sp. K6-28-040

    The α-amylase of marine Pseudomonas sp. K6-28-040 was purified through a series of three steps and the purity of enzymes was checked by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The results showed that, the enzyme was purified 4.7-fold with a specific activity of 134.6 U/mg protein and a yield of 44%. When it was subjected to SDS-PAGE, a single band near 58 kDa appeared. The optimum temperature and pH were 50°C and 7.0, respectively. The addition of Ca2+, Mn2+ and Co2+ could improve the enzyme activity, while Cu2+, Hg2+, Fe3+ and Al3+ decreased the activity. The enzyme was inhibited by ethylenediaminetetraacetic acid (EDTA), ethylenebis(oxonitrilo)]tetra-acetate (EGTA), SDS and dimethyl sulfoxide (DMSO), but was not affected by phenylmethane-sulfonyl fluoride (PMSF) and 1,4-dithiothreitol (DTT). Km and Vmax values of the purified enzyme for soluble starch were 1.73 ± 0.3 mg/ml and 1.24 ± 0.02 mg/ml/min, respectively. The degradation ability of wild type α amylase on starch granules was examined by thin layer chromatography. The final purified enzyme had an isoelectric point of 7.5-7.8 and α-helix of 28%, β-sheet of 32% and random coil of 40%.Keywords: α-Amylase; Pseudomonas sp., purification, enzyme characterizatio

    Parenting styles as a moderator of the association between pubertal timing and chinese adolescents’ drinking behavior

    Background: Pubertal timing refers to the timing of an individual regarding pubertal sexual maturation, both physiologically and psychologically. Existing research shows that pubertal timing is associated with behavioral problems. This study investigated the role of parenting style in the relationship between pubertal timing and Chinese adolescents’ smoking behavior. Methods: The study examined the association of pubertal timing, parenting style and adolescents’ smoking behavior, using the Pubertal Development Scale (Chinese version), Simplified Parenting Style Scale-Chinese version, and three items related to adolescents’ smoking situation. Participants were 1391 Chinese adolescents aged 11–16 years old (53.41% boys). Hierarchical linear regression analyses assessed the moderating role of parenting style on the association between pubertal timing and adolescent smoking behavior. Results: The results indicated that parenting style moderates the relationship between pubertal timing and adolescent smoking behavior. For male adolescents, father rejection moderated the relationship between early pubertal timing and smoking behavior. For female adolescents, father rejection, father emotional warmth, and mother emotional warmth moderated the relationship between pubertal timing and smoking behavior. Conclusions: Findings from the study highlight the importance of parenting style, which may influence the negative outcomes associated with early pubertal timing and can help improve interventions aimed at reducing these negative outcomes

    Tumor-directed gene therapy in mice using a composite nonviral gene delivery system consisting of the piggyBac transposon and polyethylenimine

    <p>Abstract</p> <p>Background</p> <p>Compared with viral vectors, nonviral vectors are less immunogenic, more stable, safer and easier to replication for application in cancer gene therapy. However, nonviral gene delivery system has not been extensively used because of the low transfection efficiency and the short transgene expression, especially <it>in vivo</it>. It is desirable to develop a nonviral gene delivery system that can support stable genomic integration and persistent gene expression <it>in vivo</it>. Here, we used a composite nonviral gene delivery system consisting of the <it>piggyBac </it>(PB) transposon and polyethylenimine (PEI) for long-term transgene expression in mouse ovarian tumors.</p> <p>Methods</p> <p>A recombinant plasmid PB [Act-RFP, HSV-tk] encoding both the herpes simplex thymidine kinase (HSV-tk) and the monomeric red fluorescent protein (mRFP1) under PB transposon elements was constructed. This plasmid and the PBase plasmid were injected into ovarian cancer tumor xenografts in mice by <it>in vivo </it>PEI system. The antitumor effects of HSV-tk/ganciclovir (GCV) system were observed after intraperitoneal injection of GCV. Histological analysis and TUNEL assay were performed on the cryostat sections of the tumor tissue.</p> <p>Results</p> <p>Plasmid construction was confirmed by PCR analysis combined with restrictive enzyme digestion. mRFP1 expression could be visualized three weeks after the last transfection of pPB/TK under fluorescence microscopy. After GCV admission, the tumor volume of PB/TK group was significantly reduced and the tumor inhibitory rate was 81.96% contrasted against the 43.07% in the TK group. Histological analysis showed that there were extensive necrosis and lymphocytes infiltration in the tumor tissue of the PB/TK group but limited in the tissue of control group. TUNEL assays suggested that the transfected cells were undergoing apoptosis after GCV admission <it>in vivo</it>.</p> <p>Conclusion</p> <p>Our results show that the nonviral gene delivery system coupling PB transposon with PEI can be used as an efficient tool for gene therapy in ovarian cancer.</p
