    Metabolic fluxes in the central carbon metabolism of Dinoroseobacter shibae and Phaeobacter gallaeciensis, two members of the marine Roseobacter clade

    <p>Abstract</p> <p>Background</p> <p>In the present work the central carbon metabolism of <it>Dinoroseobacter shibae </it>and <it>Phaeobacter gallaeciensis </it>was studied at the level of metabolic fluxes. These two strains belong to the marine <it>Roseobacter </it>clade, a dominant bacterial group in various marine habitats, and represent surface-associated, biofilm-forming growth (<it>P. gallaeciensis</it>) and symbiotic growth with eukaryotic algae (<it>D. shibae</it>). Based on information from recently sequenced genomes, a rich repertoire of pathways has been identified in the carbon core metabolism of these organisms, but little is known about the actual contribution of the various reactions <it>in vivo</it>.</p> <p>Results</p> <p>Using <sup>13</sup>C labelling techniques in specifically designed experiments, it could be shown that glucose-grown cells of <it>D. shibae </it>catabolise the carbon source exclusively via the Entner-Doudoroff pathway, whereas alternative routes of glycolysis and the pentose phosphate pathway are obviously utilised for anabolic purposes only. Enzyme assays confirmed this flux pattern and link the lack of glycolytic flux to the absence of phosphofructokinase activity. The previously suggested formation of phosphoenolpyruvate from pyruvate during mixotrophic CO<sub>2 </sub>assimilation was found to be inactive under the conditions studied. Moreover, it could be shown that pyruvate carboxylase is involved in CO<sub>2 </sub>assimilation and that the <it>cyclic </it>respiratory mode of the TCA cycle is utilised. Interestingly, the use of intracellular pathways was highly similar for <it>P. gallaeciensis</it>.</p> <p>Conclusion</p> <p>The present study reveals the first insight into pathway utilisation within the <it>Roseobacter </it>group. Fluxes through major intracellular pathways of the central carbon metabolism, which are closely linked to the various important traits found for the <it>Roseobacter </it>clade, could be determined. The close similarity of fluxes between the two physiologically rather different species might provide the first indication of more general key properties among members of the <it>Roseobacter </it>clade which may explain their enormous success in the marine realm.</p

    Wicked facets of the German energy transition – examples from the electricity, heating, transport, and industry sectors

    We shed light on wicked problems in the German energy transition. Our methods consist of a multiple-case study and multi-criteria analysis, utilising the wicked problems theoretical framework introduced by Horst Rittel and Melvin Webber [1973. “Dilemmas in a General Theory of Planning.” Policy Sciences 4 (2): 155–169. Results from the energy supply, heating/cooling, transport, and industry sectors illustrate where and how the 10-point frame of wicked problems manifests in the German energy transition. The four cases exhibit more wicked tendencies in the governance domain than the technical domain and differ in their degrees of technology maturity, policy regulation, and knowledge states. We do not find that the German energy transition is inherently wicked. However, wickedness unfolds through the social setting into which technical solutions of the energy transition are embedded. We aim to highlight these intricacies and encourage scrutinising these wicked facets early on

    Effect of high-dose glucocorticoid treatment on human brown adipose tissue activity: a randomised, double-blinded, placebo-controlled cross-over trial in healthy men

    BACKGROUND Glucocorticoids (GCs) are widely applied anti-inflammatory drugs that are associated with adverse metabolic effects including insulin resistance and weight gain. Previous research indicates that GCs may negatively impact brown adipose tissue (BAT) activity in rodents and humans. METHODS We performed a randomised, double-blinded cross-over trial in 16 healthy men (clinicaltrials.govNCT03269747). Participants received 40 mg of prednisone per day for one week or placebo. After a washout period of four weeks, participants crossed-over to the other treatment arm. Primary endpoint was the increase in resting energy expenditure (EE) in response to a mild-cold stimulus (cold-induced thermogenesis, CIT). Secondary outcomes comprised mean 18^{18}F-FDG uptake into supraclavicular BAT (SUVmean_{mean}) as determined by FDG-PET/CT, volume of the BAT depot as well as fat content determined by MRI. The plasma metabolome and the transcriptome of supraclavicular BAT and of skeletal muscle biopsies after each treatment period were analysed. FINDINGS Sixteen participants were recruited to the trial and completed it successfully per protocol. After prednisone treatment resting EE was higher both during warm and cold conditions. However, CIT was similar, 153 kcal/24 h (95% CI 40-266 kcal/24 h) after placebo and 186 kcal/24 h (95% CI 94-277 kcal/24 h, p = 0.38) after prednisone. SUVmean_{mean} of BAT after cold exposure was not significantly affected by prednisone (3.36 g/ml, 95% CI 2.69-4.02 g/ml, vs 3.07 g/ml, 95% CI 2.52-3.62 g/ml, p = 0.28). Results of plasma metabolomics and BAT transcriptomics corroborated these findings. RNA sequencing of muscle biopsies revealed higher expression of genes involved in calcium cycling. No serious adverse events were reported and adverse events were evenly distributed between the two treatments. INTERPRETATION Prednisone increased EE in healthy men possibly by altering skeletal muscle calcium cycling. Cold-induced BAT activity was not affected by GC treatment, which indicates that the unfavourable metabolic effects of GCs are independent from thermogenic adipocytes. The project is a joined effort by the German Institute for Economic Research (DIW Berlin), the Workgroup for Infrastructure Policy (WIP) at Technische UniversitĂ€t Berlin (TUB), the Chair of Energy Economics (EE2) at Technische UniversitĂ€t Dresden (TUD), and the House of Energy Markets & Finance at University of Duisburg-Essen. The project was funded by the German Federal Ministry for Economic Affairs and Energy through the grant ‘LKD-EU’, FKZ 03ET4028A. The objective of this paper is to document a reference data set representing the status quo of the German energy sector. We also update and extend parts of the previous DIW Data Documentation 75 (Egerer et al. 2014). While the focus is on the electricity sector, the heat and natural gas sectors are covered as well. With this reference data set, we aim to increase the transparency of energy infrastructure data in Germany. On the one hand, this documentation presents sources of original data and information used for the data set. On the other hand, it elaborates on the methodologies which have been applied to derive the data from respective sources in order to make it useful for modeling purposes and to promote a discussion about the underlying assumptions. Furthermore, we briefly discuss the underlying regulations with regard to data transparency in the energy sector.     Ausgehend von der Feststellung, dass die Integration mittelalterlicher Texte im Deutschunterricht in der Schulpraxis weitgehend ein Desiderat darstellt, prĂ€sentierten Beitragende aus Schule und Wissenschaft bei der Tagung „Helden in der Schule“ im Oktober 2014 ihre Projekte und Ideen zu und Erfahrungen mit der Implementation germanistisch-mediĂ€vistischer Inhalte im Deutschunterricht. Dabei reichen die BeitrĂ€ge von allgemeinen Überlegungen zum Nutzen mittelalterlicher Literatur in der Schule ĂŒber konkrete UnterrichtsentwĂŒrfe bis hin zur Umsetzung in der Waldorfschule und der Integration schulbezogener Lehrveranstaltungen in der UniversitĂ€t. Bei aller Verschiedenheit in den Herangehensweisen wird in allen BeitrĂ€gen eindrucksvoll deutlich gemacht, dass mittelalterliche Literatur auch im 21. Jahrhundert ĂŒberaus lohnend in die Unterrichtspraxis einbezogen werden kann.Since medieval texts are not treated enough in school, lecturers working in school and university presented their projects, ideas and experiences concerning the implementation of German-mediavistic contents in German school lessons during the conference “Heroes in School” in October 2014. The topics concern general reflection about the profitability of medieval literature in school, concrete lesson plans, the implementation in Rudolf Steiner schools, the integration of seminars in university that deal with the work in school etc. All articles demonstrate that the integration of medieval literature in school worth the effort – also in the 21st century
