    O geoplano em aulas de Matemática: diálogos entre geometria e língua materna

    Neste artigo se descreve um estudo desenvolvido em sala de aula com base nas pesquisas realizadas pelos autores Hoffman (2012) e Zanon (2011) mediante sua inserção no Grupo de Estudos em Educação Matemática do Espírito Santo [GEEM-ES]. Traz-se um recorte de uma pesquisa qualitativa desenvolvida, em 2016, com 28 alunos de uma turma do quinto ano, em uma escola pública municipal de Vitória-ES. O texto mostra que conceitos de geometria e de língua materna podem ser construídos em um processo dialógico que utiliza o geoplano quadricular e as interlocuções entre aulas de matemática de caráter interdisciplinar. Apresenta e analisa aulas de noções básicas de geometria que foram direcionadas na perspectiva do diálogo realizado ante a manipulação do material concreto, da observação e da pesquisa sobre a própria prática, na ação, como parte do processo de ensino, aprendizagem e avaliação em matemática. Assim, mostra como conduzir atividades interdisciplinares que utilizam leitura e escrita na busca pela construção de conceitos de geometria a partir da exploração do geoplano quadricular e de outros recursos em diferentes espaços. A pesquisa apontou que, em uma perspectiva mais discursiva, é possível mediar a construção de conceitos de geometria, fazendo interlocução entre a língua materna e matemática com suas regras e simbologias próprias. E sugere que o processo de formação do estudante deve ser visto como um todo, em que as diversas áreas do conhecimento dialoguem entre si, evitando sua fragmentação e atribuindo sentido aos conteúdos escolares

    Minthostachys verticillata essentials oil and its major components: antiherpetic selective action in HEp-2 cells

    The medicinal herb Minthostachys verticillata was studied in various fields of natural medicine. Numerous studies were made to evaluate its antimicrobial properties. This work emphasizes in the analysis of the antiviral capacity of its Essential Oil (EO). The aim of this study was to determine the Therapeutic Index (TI) of the EO and its main chemical components. Suid herpesvirus type 1 or Pseudorabies virus (Prv) was proliferated in HEp-2 cells in presence of non-toxic concentrations of the oily samples. Antiviral action was quantified by reducing the number of plaques of lysis. Previously, the cytotoxicity of the samples was evaluated and 50% Cytotoxic Concentration values (CC50) were determined. The TI was calculated by relating the cytotoxic concentrations of each sample vs. those concentrations active against the virus. The results showed that the major components of the EO were pulegone, menthone and limonene, with CC50 of 352, 835 and 1155 μg/mL respectively vs. the CC50 of EO: 613 μg/mL. The antiviral action was exerted by pulegone and the EO at concentrations 20 to 30 times lowers than the toxic concentrations.     The early expansion of anergic NKG2Apos/CD56dim/CD16neg natural killer cells represents a therapeutic target in haploidentical haematopoietic stem cell transplantation

    Natural Killer cells are the first lymphocyte population to reconstitute early after non myelo-ablative and T cell-replete haploidentical hematopoietic stem cell transplantation with post-transplant infusion of cyclophosphamide. The present study characterizes the transient and predominant expansion starting from the 2nd week after haploidentical hematopoietic stem cell transplantation of a donor-derived unconventional subset of NKp46neg-low/CD56dim/CD16neg natural killer cells expressing remarkable high levels of CD94/NKG2A. Both transcription and phenotypic profiles indicated that unconventional NKp46neg-low/CD56dim/CD16neg natural killer cells are a distinct natural killer cell subpopulation with features of late stage differentiation, yet retaining proliferative capability and functional plasticity to generate conventional NKp46pos/CD56bright/CD16pos natural killer cells in response to interleukin-15 plus interleukin-18. While present at low frequency in healthy donors, unconventional NKp46neg-low/CD56dim/CD16neg natural killer cells are greatly expanded in the following 7 weeks after haploidentical hematopoietic stem cell transplantation and express high levels of the activating receptors NKGD and NKp30 as well as of the lytic granules Granzyme-B and Perforin. Nonetheless, NKp46neg-low/CD56dim/CD16neg natural killer cells displayed a markedly defective cytotoxicity that could be reversed by blocking the inhibitory receptor CD94/NKG2A. These data open new important perspectives to better understand the ontogenesis/homeostasis of human natural killer cells and to develop a novel immune-therapeutic approach that targets the inhibitory NKG2A check point, thus unleashing natural killer cell alloreactivity early after haploidentical hematopoietic stem cell transplantation

    Two subsets of stem-like CD8+ memory T cell progenitors with distinct fate commitments in humans

    T cell memory relies on the generation of antigen-specific progenitors with stem-like properties. However, the identity of these progenitors has remained unclear, precluding a full understanding of the differentiation trajectories that underpin the heterogeneity of antigen-experienced T cells. We used a systematic approach guided by single-cell RNA-sequencing data to map the organizational structure of the human CD8+ memory T cell pool under physiological conditions. We identified two previously unrecognized subsets of clonally, epigenetically, functionally, phenotypically and transcriptionally distinct stem-like CD8+ memory T cells. Progenitors lacking the inhibitory receptors programmed death-1 (PD-1) and T cell immunoreceptor with Ig and ITIM domains (TIGIT) were committed to a functional lineage, whereas progenitors expressing PD-1 and TIGIT were committed to a dysfunctional, exhausted-like lineage. Collectively, these data reveal the existence of parallel differentiation programs in the human CD8+ memory T cell pool, with potentially broad implications for the development of immunotherapies and vaccines
