    Genetic diversity of Escherichia coli isolated from commercial swine farms revealed by enterobacterial repetitive intergenic consensus PCR (ERIC-PCR) and repetitive extragenic palindrome PCR (REP-PCR)

    The objective of this study was to use enterobacterial repetitive intergenic consensus PCR (ERIC-PCR) and repetitive extragenic palindrome PCR (REP-PCR) for the analysis of genetic diversity among Escherichia coli strains isolated from commercial swine farms in Sichuan province of China. Thirty four strains of E. coli were selected by selective medium and conventional biochemical test from fresh stool samples of swines in five farms in Sichuan province. The isolates were identified by 160 kinds of E. coli O serums. The results show that 30 strains were determined among 34 E. coli isolates, 12 kinds of O serogroups were obtained on the basis of the agglutination test. The predominant types are O23, O113 and O120, representing 35.4%. Furthermore, the genotypes and phylogenetic relationship of all isolates were analysed by Enterobacterial repetitive intergenic consensus PCR (ERIC-PCR) and repetitive extragenic palindrome PCR (REP-PCR), 34 E. coli isolates were clustered to 19 ERIC-PCR genotypes and 13 REP-PCR genotypes. The isolates from the same farm or sharing the same serotyping showed different genotype. And the isolates which could not be serotyped were genotyped by ERIC-PCR and REP-PCR. The analysis of genetic type and original source revealed that isolates from different farms had different genetic types. The subtypes of E. coli are also different within a single farm. Genetic variability with E. coli strains isolated from swine farms in China has been demonstrated. The presence of ERIC-PCR and REP-PCR sequences in the genome of E. coli was confirmed. ERIC-PCR and REP-PCR techniques are more rapid methods for molecular typing of E. coli strain. They are also useful methods for diversity survey of E. coli and the two methods analyzes genetic diversity of E. coli isolated in Sichuan of China.Key words: Escherichia coli, serotype, enterobacterial repetitive intergenic consensus PCR (ERIC-PCR), repetitive extragenic palindrome PCR (REP-PCR)

    Plasma Hormones Facilitated the Hypermotility of the Colon in a Chronic Stress Rat Model

    Objective: To study the relationship between brain-gut peptides, gastrointestinal hormones and altered motility in a rat model of repetitive water avoidance stress (WAS), which mimics the irritable bowel syndrome (IBS). Methods: Male Wistar rats were submitted daily to 1-h of water avoidance stress (WAS) or sham WAS (SWAS) for 10 consecutive days. Plasma hormones were determined using Enzyme Immunoassay Kits. Proximal colonic smooth muscle (PCSM) contractions were studied in an organ bath system. PCSM cells were isolated by enzymatic digestion and IKv and IBKca were recorded by the patch-clamp technique. Results: The number of fecal pellets during 1 h of acute restraint stress and the plasma hormones levels of substance P (SP), thyrotropin-releasing hormone (TRH), motilin (MTL), and cholecystokinin (CCK) in WAS rats were significantly increased compared with SWAS rats, whereas vasoactive intestinal peptide (VIP), calcitonin gene-related peptide (CGRP) and corticotropin releasing hormone (CRH) in WAS rats were not significantly changed and peptide YY (PYY) in WAS rats was significantly decreased. Likewise, the amplitudes of spontaneous contractions of PCSM in WAS rats were significantly increased comparing with SWAS rats. The plasma of WAS rats (100 ml) decreased the amplitude of spontaneous contractions of controls. The IKv and IBKCa of PCSMs were significantly decreased in WAS rats compared with SWAS rats and the plasma of WAS rats (100 ml) increased the amplitude of IKv and IBKCa in normal rats

    Quantitative impacts of regenerative vibration and abrasive wheel eccentricity on surface grinding dynamic performance

    In grinding, regenerative vibration and forced vibration due to grinding wheel eccentric rotation are main excited-vibration sources that interact with grinding material removal mechanism. In the paper, instantaneous undeformed chip thickness in down-grinding cutting phase may consist of two components, i.e. linear kinetic thickness and nonlinear dynamic thickness. Considering abrasive grit-workpiece interaction in the grinding contact zone, the grinding vibration system is presented by a new set of differential equations of two degrees of freedom (DOF) with a close-loop feedback control system models. Conventional grinding control parameters, including wheel spindle speed, work-speed in feed direction and radial cutting depth, are often regarded as linear constants in many existing simplified models. When considering time delay, they can be transferred to nonlinear variables, so the capability of prediction and the accuracy of solution of the grit-workpiece dynamic performance are improved. Based on quantitative comparison of force and vibration magnitudes, the influence of the eccentric rotation of abrasive wheel and the negative rake angle of working grit cutting edges on grinding performance are demonstrated in the paper.     A Human TREK-1/HEK Cell Line: A Highly Efficient Screening Tool for Drug Development in Neurological Diseases

    TREK-1 potassium channels are involved in a number of physiopathological processes such as neuroprotection, pain and depression. Molecules able to open or to block these channels can be clinically important. Having a cell model for screening such molecules is of particular interest. Here, we describe the development of the first available cell line that constituvely expresses the TREK-1 channel. The TREK-1 channel expressed by the h-TREK-1/HEK cell line has conserved all its modulation properties. It is opened by stretch, pH, polyunsaturated fatty acids and by the neuroprotective molecule, riluzole and it is blocked by spadin or fluoxetine. We also demonstrate that the h-TREK-1/HEK cell line is protected against ischemia by using the oxygen-glucose deprivation model
