    Low-molecular-weight components of olive oil mill wastewaters

    A new lignan 1-hydroxy-2-(4-hydroxy-3-methoxyphenyl)-6-(3-acetyl-4-hydroxy-5-methoxyphenyl)-3,7-dioxabicyclo[ 3.3.0]octane, the secoiridoid 2H-pyran-4-acetic acid,3-hydroxymethyl-2,3-dihydro-5-(methoxycarbonyl)-2- methyl-, methyl ester, the phenylglycoside 4-[β -D-xylopyranosyl-(1→6)]-β -D-glucopyranosyl-1,4-dihydroxy-2- methoxybenzene and the lactone 3-[1-(hydroxymethyl)-1-propenyl] δ -glutarolactone were isolated and identified on the basis of spectroscopic data including two-dimensional NMR, as components of olive oil mill waste-waters. The known aromatic compounds catechol, 4-hydroxybenzoic acid, protocatechuic acid, vanillic acid, 4-hydroxy- 3,5-dimethoxybenzoic acid, 4-hydroxyphenylacetic acid, 3,4-dihydroxyphenylacetic acid, tyrosol, hydroxytyrosol, 2-(4-hydroxy-3-methoxy)phenylethanol, 2-(3,4-dihydroxy)phenyl-1,2-ethandiol, p-coumaric acid, caffeic acid, ferulic acid, sinapic acid, 1-O-[2-(3,4-dihydroxy)phenylethyl]-(3,4-dihydroxy)phenyl-1,2-ethandiol, 1-O-[2-(4- hydroxy)phenylethyl]-(3,4-dihydroxy)phenyl-1,2-ethandiol, D(+)-erythro-1-(4-hydroxy-3-methoxy)-phenyl-1,2,3- propantriol, p-hydroxyphenethyl-β-D-glucopyranoside, 2(3,4-dihydroxyphenyl)ethanol 3β-D-glucopyranoside, and 2(3,4-dihydroxyphenyl)ethanol 4β-D-glucopyranoside were also confirmed as constituents of the waste-waters

The C6C3 units are linked by an oxygen atom at C4–C40 or C4–C20 and they are dihydrophenylpropanoid acid units. Their effects on germination and growth of Lactuca sativa L. have been studied in the range concentration 10K4–10K7 M

    TG, FT-IR and NMR characterization of n-C16H34 contaminated alumina and silica after mechanochemical treatment

    This paper deals with the application of mechanochemistry to model systems composed of alumina or silica artificially contaminated with n-C16H34. The mechanochemical treatment was carried out by means of a ring mill for times ranging from 10 to 40 h. Thermogravimetry and infrared and nuclear magnetic resonance spectroscopies were used for the characterization of the mechanochemical products. The results have indicated that, in the case of alumina, almost all the contaminant n-C16H34 undergoes a complex oxidative reaction path whose end products are strongly held on the surface. These end products are most likely made of crosslinked, partially oxidized hydrocarbon chains bond to the solid surface via COO− groups. In the case of silica, the hydrocarbon undergoes a different, equally complex reaction path, but to a lower extent. In this case the end products are most probably carbonylic compounds and graphitic carbon. Then, for both solid matrices, the mechanochemical treatment promotes significant modification of the chemical nature of the polluting hydrocarbon with end products much more difficult to remove from the surface. As the systems studied are models of sites contaminated by aliphatic hydrocarbon, the results are worthy of consideration in relation to the mobility of the contaminants in the environment

    A Randomized Clinical Trial Evaluating the Efficacy of an Anthocyanin–Maqui Berry Extract (Delphinol®) on Oxidative Stress Biomarkers

    Objective: Berries are a rich source of anthocyanins, and clinical data suggest that a polyphenol-rich diet may exert health-promoting effects by reducing oxidative stress. The aim of this study was to elucidate the effects of dietary supplementation with Delphinol (trademark owned by MNL Chile) standardized maqui berry (Aristotelia chilensis) extract on products of lipid peroxidation in healthy, overweight, and smoker subjects.Methods: In a double-blind, placebo-controlled design, 42 participants (age 45-65years) consumed in random order either a standardized extract of maqui berry (162mg anthocyanins) or a matched placebo, given 3times daily for 4weeks. The samples were collected at baseline, after the end of the supplementation, and 40days after the end of the study. Primary outcome was the measure of oxidized low-density lipoprotein (Ox-LDL) and F-2-isoprostanes in plasma and urine, respectively. Secondary outcomes included anthropometric measures, blood pressure, and lipid profile.Results: Delphinol supplementation was associated with reduced levels of Ox-LDL in the anthocyanin group compared to baseline (p < 0.05). There was also a decrease in urinary F-2-isoprostanes (8-iso-prostaglandin F2) at 4weeks versus baseline in the Delphinol-supplemented group (p < 0.05). However, no differences in primary outcomes were evident at 40days of follow-up. In the fourth week of the intervention, no significant differences were noted for anthropometric characteristics, ambulatory blood pressure, and lipid profile.Conclusions: Our observations suggest that dietary interventions with maqui berry extract may improve oxidative status (Ox-LDL and F-2-isoprostanes) in healthy adults, overweight adults, and adult smoke

    Gymnema sylvestre R. Br. is one of the most important medicinal plants that grows in tropical forests in India and South East Asia. Its active ingredients and extracts of leaves and roots are used in traditional medicine to treat various ailments and they are present in the market for pharmaceutical and parapharmaceutical products. Commercial products based on substances of plant origin that are generally connoted as natural have to be subjected to monitoring and evaluation by health authorities for their potential impacts on public health. The monitoring and evaluation of these products are critical because the boundary between a therapeutic action and a functional or healthy activity has not yet been defined in a clear and unambiguous way. Therefore, these products are considered borderline products, and they require careful and rigorous studies, in order to use them as complement and/or even replacement of synthetic drugs that are characterized by side effects and high economic costs. This review explores the traditional uses, chemical composition and biological activity of G. sylvestre extracts, providing a general framework on the most interesting extracts and what are the necessary studies for a complete definition of the range of activities

    New findings on the d(TGGGAG) sequence: Surprising anti-HIV-1 activity

    The biological relevance of tetramolecular G-quadruplexes especially as anti-HIV agents has been extensively reported in the literature over the last years. In the light of our recent results regarding the slow G-quadruplex folding kinetics of ODNs based on d(TGGGAG) sequence, here, we report a systematic anti-HIV screening to investigate the impact of the G-quadruplex folding on their anti-HIV activity. In particular, varying the single stranded concentrations of ODNs, it has been tested a pool of ODN sample solutions with different G-quadruplex concentrations. The anti-HIV assays have been designed favouring the limited kinetics involved in the tetramolecular G4-association based on the d(TGGGAG) sequence. Aiming to determine the stoichiometry of G-quadruplex structures in the same experimental conditions of the anti-HIV assays, a native gel electrophoresis was performed. The gel confirmed the G-quadruplex formation for almost all sample solutions while showing the formation of high order G4 structures for the more concentrated ODNs solutions. The most significant result is the discovery of a potent anti-HIV activity of the G-quadruplex formed by the natural d(TGGGAG) sequence (IC50 = 14 nM) that, until now, has been reported to be completely inactive against HIV infection

    Synthesis of Oligonucleotide Conjugates and Phosphorylated Nucleotide Analogues: An Improvement to a Solid Phase Synthetic Approach

    An improvement to our solid phase strategy to generate pharmacologically interesting molecule libraries is proposed here. The synthesis of newo-chlorophenol-functionalised solid supports with very high loading (0.18–0.22 meq/g for control pore glass (CPG) and 0.25–0.50 meq/g for TG) is reported. To test the efficiency of these supports, we prepared nucleotide and oligonucleotide models, and their coupling yields and the purity of the crude detached materials were comparable to previously available results. These supports allow the facile and high-yield preparation of highly pure phosphodiester and phosphoramidate monoester nucleosides, conjugated oligonucleotides, and other yet unexplored classes of phosphodiester and phosphoramidate molecules

    Dietary phytochemicals and neuro-inflammaging: From mechanistic insights to translational challenges

    An extensive literature describes the positive impact of dietary phytochemicals on overall health and longevity. Dietary phytochemicals include a large group of non-nutrients compounds from a wide range of plant-derived foods and chemical classes. Over the last decade, remarkable progress has been made to realize that oxidative and nitrosative stress (O&NS) and chronic, low-grade inflammation are major risk factors underlying brain aging. Accumulated data strongly suggest that phytochemicals from fruits, vegetables, herbs, and spices may exert relevant negative immunoregulatory, and/or anti-O&NS activities in the context of brain aging. Despite the translational gap between basic and clinical research, the current understanding of the molecular interactions between phytochemicals and immune-inflammatory and O&NS (IO&NS) pathways could help in designing effective nutritional strategies to delay brain aging and improve cognitive function. This review attempts to summarise recent evidence indicating that specific phytochemicals may act as positive modulators of IO&NS pathways by attenuating pro-inflammatory pathways associated with the age-related redox imbalance that occurs in brain aging. We will also discuss the need to initiate long-term nutrition intervention studies in healthy subjects. Hence, we will highlight crucial aspects that require further study to determine effective physiological concentrations and explore the real impact of dietary phytochemicals in preserving brain health before the onset of symptoms leading to cognitive decline and inflammatory neurodegeneration

    Litter Inhibitory Effects on Soil Microbial Biomass, Activity, and Catabolic Diversity in Two Paired Stands of Robinia pseudoacacia L. and Pinus nigra Arn.

    Research Highlights: Plant cover drives the activity of the microbial decomposer community and affects carbon (C) sequestration in the soil. Despite the relationship between microbial activity and C sequestration in the soil, potential inhibition of soil microbial activity by plant cover has received little attention to date. Background and Objectives: Differences in soil microbial activity between two paired stands on soil at a very early stage of formation and a common story until afforestation, can be traced back to the plant cover. We hypothesized that in a black locust (Robinia pseudoacacia L.) stand the high-quality leaf litter of the tree, and that of the blackberry (Rubus fruticosus L.) understory had an inhibitory effect on soil microbial community resulting in lower mineralization of soil organic matter compared to the paired black pine (Pinus nigra Arn.) stand. Materials and Methods: We estimated potential mineralization rates (MR), microbial (MB), and active fungal biomass (AFB) of newly-shed litter, forest floor, and mineral soil. We tested the effects of litters' water extracts on soil MR, MB, AFB and its catabolic response profile (CRP). Results: Newly-shed litter of black locust had higher MR than that of blackberry and black pine; MR, MB, and AFB were higher in forest floor and in mineral soil under black pine than under black locust. Water extracts of black locust and blackberry litter had a negative effect on the amount, activity of microorganisms, and CRP. Conclusions: The results demonstrate the potential for black locust and blackberry litter to have a marked inhibitory effect on decomposer microorganisms that, in turn, reduce organic matter mineralization with possible consequences at the ecosystem level, by increasing C sequestration in mineral soil.Peer reviewe

    New silibinin glyco-conjugates: Synthesis and evaluation of antioxidant properties

    New silibinin glyco-conjugates have been synthesized by efficient method and in short time. Exploiting our solution phase strategy, several structurally diverse silibinin glyco-conjugates (gluco, manno, galacto, and lacto-) were successfully realized in very good yields and in short time. In preliminary study to evaluate their antioxidant and neuroprotective activities new derivatives were subjected to DPPH free radical scavenging assay and the Xanthine oxidase (XO) inhibition models assay. Irrespective of the sugar moiety examined, new glyco-conjugates are more than 50 times water-soluble of silibinin. In the other hand they exhibit a radical scavenging activities slightly higher than to silibinin and XO inhibition at least as silibinin. (C) 2014 Elsevier Ltd. All rights reserved
