    Association of IGF1 and KDM5A polymorphisms with performance, fatness and carcass traits in chickens

    Two functional and positional candidate genes were selected in a region of chicken chromosome 1 (GGA1), based on their biological roles, and also where several quantitative trait loci (QTL) have been mapped and associated with performance, fatness and carcass traits in chickens. The insulin-like growth factor 1 (IGF1) gene has been associated with several physiological functions related to growth. The lysine (K)-specific demethylase 5A (KDM5A) gene participates in the epigenetic regulation of genes involved with the cell cycle. Our objective was to find associations of selected single-nucleotide polymorphisms (SNPs) in these genes with performance, fatness and carcass traits in 165 F chickens from a resource population. In the IGF1 gene, 17 SNPs were detected, and in the KDM5A gene, nine SNPs were detected. IGF1 SNP c. 47673G > A was associated with body weight and haematocrit percentage, and also with feed intake and percentages of abdominal fat and gizzard genotype × sex interactions. KDM5A SNP c. 34208C > T genotype × sex interaction affected body weight, feed intake, percentages of abdominal fat (p = 0. 0001), carcass, gizzard and haematocrit. A strong association of the diplotype × sex interaction (p < 0. 0001) with abdominal fat was observed, and also associations with body weight, feed intake, percentages of carcass, drums and thighs, gizzard and haematocrit. Our findings suggest that the KDM5A gene might play an important role in the abdominal fat deposition in chickens. The IGF1 and KDM5A genes are strong candidates to explain the QTL mapped in this region of GGA1

    Depletion of B2 but Not B1a B Cells in BAFF Receptor-Deficient ApoE−/− Mice Attenuates Atherosclerosis by Potently Ameliorating Arterial Inflammation

    We have recently identified conventional B2 cells as atherogenic and B1a cells as atheroprotective in hypercholesterolemic ApoE−/− mice. Here, we examined the development of atherosclerosis in BAFF-R deficient ApoE−/− mice because B2 cells but not B1a cells are selectively depleted in BAFF-R deficient mice. We fed BAFF-R−/− ApoE−/− (BaffR.ApoE DKO) and BAFF-R+/+ApoE−/− (ApoE KO) mice a high fat diet (HFD) for 8-weeks. B2 cells were significantly reduced by 82%, 81%, 94%, 72% in blood, peritoneal fluid, spleen and peripheral lymph nodes respectively; while B1a cells and non-B lymphocytes were unaffected. Aortic atherosclerotic lesions assessed by oil red-O stained-lipid accumulation and CD68+ macrophage accumulation were decreased by 44% and 50% respectively. B cells were absent in atherosclerotic lesions of BaffR.ApoE DKO mice as were IgG1 and IgG2a immunoglobulins produced by B2 cells, despite low but measurable numbers of B2 cells and IgG1 and IgG2a immunoglobulin concentrations in plasma. Plasma IgM and IgM deposits in atherosclerotic lesions were also reduced. BAFF-R deficiency in ApoE−/− mice was also associated with a reduced expression of VCAM-1 and fewer macrophages, dendritic cells, CD4+ and CD8+ T cell infiltrates and PCNA+ cells in lesions. The expression of proinflammatory cytokines, TNF-α, IL1-β and proinflammatory chemokine MCP-1 was also reduced. Body weight and plasma cholesterols were unaffected in BaffR.ApoE DKO mice. Our data indicate that B2 cells are important contributors to the development of atherosclerosis and that targeting the BAFF-R to specifically reduce atherogenic B2 cell numbers while preserving atheroprotective B1a cell numbers may be a potential therapeutic strategy to reduce atherosclerosis by potently reducing arterial inflammation

    Modulation of 11β-hydroxysteroid dehydrogenase as a strategy to reduce vascular inflammation

    Atherosclerosis is a chronic inflammatory disease in which initial vascular damage leads to extensive macrophage and lymphocyte infiltration. Although acutely glucocorticoids suppress inflammation, chronic glucocorticoid excess worsens atherosclerosis, possibly by exacerbating systemic cardiovascular risk factors. However, glucocorticoid action within the lesion may reduce neointimal proliferation and inflammation. Glucocorticoid levels within cells do not necessarily reflect circulating levels due to pre-receptor metabolism by 11β-hydroxysteroid dehydrogenases (11β-HSDs). 11β-HSD2 converts active glucocorticoids into inert 11-keto forms. 11β-HSD1 catalyses the reverse reaction, regenerating active glucocorticoids. 11β-HSD2-deficiency/ inhibition causes hypertension, whereas deficiency/ inhibition of 11β-HSD1 generates a cardioprotective lipid profile and improves glycemic control. Importantly, 11β-HSD1-deficiency/ inhibition is atheroprotective, whereas 11β-HSD2-deficiency accelerates atherosclerosis. These effects are largely independent of systemic risk factors, reflecting modulation of glucocorticoid action and inflammation within the vasculature. Here, we consider whether evidence linking the 11β-HSDs to vascular inflammation suggests these isozymes are potential therapeutic targets in vascular injury and atherosclerosis

    Genes Involved in Systemic and Arterial Bed Dependent Atherosclerosis - Tampere Vascular Study

    BACKGROUND: Atherosclerosis is a complex disease with hundreds of genes influencing its progression. In addition, the phenotype of the disease varies significantly depending on the arterial bed. METHODOLOGY/PRINCIPAL FINDINGS: We characterized the genes generally involved in human advanced atherosclerotic (AHA type V-VI) plaques in carotid and femoral arteries as well as aortas from 24 subjects of Tampere Vascular study and compared the results to non-atherosclerotic internal thoracic arteries (n=6) using genome-wide expression array and QRT-PCR. In addition we determined genes that were typical for each arterial plaque studied. To gain a comprehensive insight into the pathologic processes in the plaques we also analyzed pathways and gene sets dysregulated in this disease using gene set enrichment analysis (GSEA). According to the selection criteria used (>3.0 fold change and p-value <0.05), 235 genes were up-regulated and 68 genes down-regulated in the carotid plaques, 242 genes up-regulated and 116 down-regulated in the femoral plaques and 256 genes up-regulated and 49 genes down-regulated in the aortic plaques. Nine genes were found to be specifically induced predominantly in aortic plaques, e.g., lactoferrin, and three genes in femoral plaques, e.g., chondroadherin, whereas no gene was found to be specific for carotid plaques. In pathway analysis, a total of 28 pathways or gene sets were found to be significantly dysregulated in atherosclerotic plaques (false discovery rate [FDR] <0.25). CONCLUSIONS: This study describes comprehensively the gene expression changes that generally prevail in human atherosclerotic plaques. In addition, site specific genes induced only in femoral or aortic plaques were found, reflecting that atherosclerotic process has unique features in different vascular beds

    Medical-Environmental Assessment and Forecasting of Socially Significant Pathology of the Population of the Republic of Dagestan

    Aim. The environmental-epidemiological monitoring of socially significant pathology and its forecasting for the development of effective preventive measures and environmental quality management methods in the Republic of Dagestan.Material and Methods. An epidemiological analysis of public health indicators was carried out in 41 municipalities and 10 urban districts of the Republic of Dagestan. This was carried out through current and retrospective analysis of regional health indicators, medical-geographical and mathematical statistical analysis. The data sources for the main indicators reflect the epidemiological situation in the Republic of Dagestan from 1997 to 2016.Results. An understanding of the status of the main epidemiological indicators of the condition of public health of the region's rural and urban population was obtained and future dynamics predicted. An integrated assessment of the well-being of the territory of the republic revealed areas and cities with adverse trends in public health in the main groups of socially significant and environmentally-related pathologies.Conclusions. An integrated assessment of the state of well-being in the Republic of Dagestan by groups of epidemiological indicators of general morbidity, general morbidity of various age groups of the population, socially significant and environmentally-caused morbidity, as well as a forecast of their dynamics, revealed adverse trends in public health in rural populations of Tsuntinskiy, Akhtynskiy, Gunibskiy, Kulinskiy, Tabasaranskiy, Nogayskiy, Novolakskiy, Suleiman-Stalskiy, Khasavyurtskiy, Tarumovskiy districts and of the cities of Kaspiysk, Dagestanskie Ogni, Makhachkala and Kizilyurt. An analysis of the sources of supplies of drinking water revealed a positive correlation between heavy metals content and adverse trends in socially significant pathology in the regions of Northern Dagestan

    The aim is to analyze the long-term results of the avian influenza virus surveillance monitoring of influenza virus in birds of one of the key Northern Eurasia points Lake. Uvs Nuur, the Republic of Tyva. Methods. The analysis of the available sources and our own research results is conducted. We used MEGA 5.2 software to construct a phylogenetic dendrogram. Tree topology is constructed by the method of maximum likelihood. Genetic distance matrix is calculated using the Kimura two-parameter metric method. Results. We conducted a biogeographical analysis of the Great Lakes basin, and an overview of the literature and the original results of highly pathogenic H5N1 influenza circulation and molecular epidemiology at Uvs Nuur Lake. Conclusion. Long-term observations at Lake Uvs Nuur revealed the important role of the biogeocoenose for the preservation and evolution of influenza A virus in wild bird populations. Planned ecological and virological monitoring is the basis for correct conclusions about the dynamics of epizootic process, infection control, as well as for the evaluation of the epidemic and pandemic potential of novel viral strains
