The first and second chapters analyze the impact of the financial shocks and anti-corruption campaign on Chinese firms through the bank lending channel. The third chapter provides a new method to predict the cash flow from operations (CFO) via semi-parametric estimation and machine learning. The first chapter explores the impact of the financial crisis and sovereign debt crisis on Chinese firms through the bank lending channel and firm borrowing channel. Using new data linking Chinese firms to their bank(s) and four different measurements of exposure to the international markets (international borrowing, importance of lending to foreign listed companies, share of trade settlement, and exchange/income), I find that banks with higher exposure to the international markets cut lending more during the recent financial crisis. In addition, state-owned bank loans are more pro-cyclical compared with private bank loans. Moreover, banks with higher exposure to the international markets cut lending more when there is a negative shock in OECD GDP growth. With regard to firm borrowing channel, I find that firms with higher weighted aggregate exposure to the international markets through banks have lower net debt, cash, employment, and capital investment during the financial crisis. Firms with higher weighted aggregate exposure to the global markets have higher net debt and lower cash, employment, and capital investment when there is a negative shock in OECD GDP growth. This paper also provides a theoretical model to explain the mechanism in a partially opened economy like China. The second chapter discusses the impact of the anti-corruption campaign on Chinese firms through the bank lending channel. Using confidential data linking Chinese firms to their bank(s) and prefecture-level corruption index, I find that banks located in more corrupted prefectures offer significantly less credits before the anti-corruption investigation, and this effect changes the direction after the investigation. Moreover, banks located in more corrupted prefectures tend to use higher interest rates, longer maturity, and more collateral before the campaign, all of these effects change the direction after the campaign. This paper suggests that the banks located in more corrupted prefectures have stronger monopoly power (or higher markup, and lower efficiency). This monopoly effect could be proved by that the bank concentration ratio is higher, and the bad loans of the banks are higher in the more corrupted areas, and all of these effects disappear after the campaign. The third chapter considers the methods of prediction of Cash flow from operations (CFO). Forecasting CFO is an essential topic in financial econometrics and empirical accounting. It impacts a variety of economic decisions, including valuation methodologies employing discounted cash flows, distress prediction, risk assessment, the accuracy of credit-rating predictions, and the provision of value-relevant information to security markets. Existing literature on statistically-based cash-flow prediction has pursued cross-sectional versus time-series estimation procedures in a mutually exclusive fashion. Cumulated empirical evidence indicates that the beta value varies across firms of different sizes, and the cross-sectional regression can not capture an idiosyncratic beta. However, although a time series based predictive model has the advantage of allowing for firm-specific variability in beta, it requires a long enough time series data. In this paper, we extend the literature on statistically-based, cash-flow prediction models by introducing an estimation procedure that, in essence, combine the favorable attributes of both cross-sectional estimation via the use of "local" cross-sectional data for firms of similar size and time-series estimation via the capturing of firm-specific variability in the beta parameters for the independent variables. The local learning approach assumes no a priori knowledge on the constancy of the beta coefficient. It allows the information about coefficients to be represented by only a subset of observations. This feature is particularly relevant in the CFO model, where the beta values are only related to cross-sectional data information that is "local" to its size. We provide empirical evidence that the prediction of cash flows from operations is enhanced by jointly adopting features specific to both cross-sectional and time-series modeling simultaneously.Thesis (PhD) — Boston College, 2020.Submitted to: Boston College.     R&D modes and firm performance in high-tech companies: A research based on cross-boundary ambidexterity and network structures

    This paper draws on the cross-boundary ambidexterity theory to propose that four different R&D modes impact firm performance differently and that cooperative network structure moderates the above relationships. The theoretical model is tested by using financial and patent data of 587 high-tech firms for 10 consecutive years in China. We find that different R&D modes have different impacts on a firm’s financial and innovative performance, and network structure plays different moderating roles. Practically, this work guides high-tech enterprises to optimize their resource allocation, select the most appropriate R&D mode, and establish efficient cooperative networks

    The metabolic repression effect of carbon-ion radiotherapy in synchronous hormone-sensitive oligometastatic prostate cancer

    BackgroundMetastatic prostate cancer (PCa) poses a significant public health concern. While radiation therapy (RT) is commonly utilized in the treatment of synchronous oligometastatic hormone sensitive prostate cancer (OM-HSPC), the occurrence of biochemical recurrence still remains. To deepen our understanding and optimize the outcome of OM-HSPC, we conducted this study to investigate the characteristics of PCa progression and explore potential synergistic mechanisms involving carbon-ion radiotherapy (CIRT) and neoadjuvant androgen deprivation treatment (naADT) in OM-HSPC.MethodsMetabolomic analysis was conducted with 72 urinary samples (at different timepoints) from 33 Patients (T2-3N0M0-1b) and 18 healthy volunteers by using liquid chromatography-tandem mass spectrometry (LC-MS/MS). MetaboAnalyst website and R software were employed for metabolomic analysis and visualization (using the criteria of p value < 0.05 and |FC|>1.5). The impact of CIRT on metabolism were further verified and explored through in vitro and in vivo experiments.ResultsWe found that most metabolites (223 out of 233) were upregulated in treatment-naïve PCa samples compared to healthy samples. After naADT, 60 core risk metabolites were still significantly related to PCa’s progression, and the glutamine level which was significantly higher in OM-HSPC compared to other groups. Remarkably, after CIRT treatment, the glutamine levels in OM-HSPC were significantly reduced to the level of healthy samples. Experiments further confirmed CIRT’s ability to suppress glutamine levels in PCa tumors and its potential enhancement with glutamine deprivation intervention.ConclusionCIRT with naADT might synergistically inhibit HS-OMPC development, progression and even the ADT resistance through glutamine metabolism repression, moreover, the glutamine metabolism might be a novel target to further improved the efficacy of CIRT

    Potential bioactive compounds and mechanisms of Fibraurea recisa Pierre for the treatment of Alzheimer’s disease analyzed by network pharmacology and molecular docking prediction

    IntroductionHeat-clearing and detoxifying Chinese medicines have been documented to have anti-Alzheimer’s disease (AD) activities according to the accumulated clinical experience and pharmacological research results in recent decades. In this study, Fibraurea recisa Pierre (FRP), the classic type of Heat-clearing and detoxifying Chinese medicine, was selected as the object of research.Methods12 components with anti-AD activities were identified in FRP by a variety of methods, including silica gel column chromatography, multiple databases, and literature searches. Then, network pharmacology and molecular docking were adopted to systematically study the potential anti-AD mechanism of these compounds. Consequently, it was found that these 12 compounds could act on 235 anti-AD targets, of which AKT and other targets were the core targets. Meanwhile, among these 235 targets, 71 targets were identified to be significantly correlated with the pathology of amyloid beta (Aβ) and Tau.Results and discussionIn view of the analysis results of the network of active ingredients and targets, it was observed that palmatine, berberine, and other alkaloids in FRP were the key active ingredients for the treatment of AD. Further, Kyoto encyclopedia of genes and genomes (KEGG) pathway enrichment analysis revealed that the neuroactive ligand-receptor interaction pathway and PI3K-Akt signaling pathway were the most significant signaling pathways for FRP to play an anti-AD role. Findings in our study suggest that multiple primary active ingredients in FRP can play a multitarget anti-AD effect by regulating key physiological processes such as neurotransmitter transmission and anti-inflammation. Besides, key ingredients such as palmatine and berberine in FRP are expected to be excellent leading compounds of multitarget anti-AD drugs

    Knowledge and attitudes of healthcare workers in Chinese intensive care units regarding 2009 H1N1 influenza pandemic

    <p>Abstract</p> <p>Background</p> <p>To describe the knowledge and attitudes of critical care clinicians during the 2009 H1N1 influenza pandemic.</p> <p>Methods</p> <p>A survey conducted in 21 intensive care units in 17 provinces in China.</p> <p>Results</p> <p>Out of 733 questionnaires distributed, 695 were completed. Three hundred and fifty-six respondents (51.2%) reported their experience of caring for H1N1 patients. Despite the fact that 88.5% of all respondents ultimately finished an H1N1 training program, only 41.9% admitted that they had the knowledge of 2009 H1N1 influenza. A total of 572 respondents (82.3%) expressed willingness to care for H1N1 patients. Independent variables associated with increasing likelihood to care for patients in the logistic regression analysis were physicians or nurses rather than other professionals (odds ratio 4.056 and 3.235, p = 0.002 and 0.007, respectively), knowledge training prior to patient care (odds ratio 1.531, p = 0.044), and the confidence to know how to protect themselves and their patients (odds ratio 2.109, p = 0.001).</p> <p>Conclusion</p> <p>Critical care clinicians reported poor knowledge of H1N1 influenza, even though most finished a relevant knowledge training program. Implementation of appropriate education program might improve compliance to infection control measures, and willingness to work in a pandemic.</p

    Haemophilus parasuis Infection Disrupts Adherens Junctions and Initializes EMT Dependent on Canonical Wnt/β-Catenin Signaling Pathway

    In this study, animal experimentation verified that the canonical Wnt/β-catenin signaling pathway was activated under a reduced activity of p-β-catenin (Ser33/37/Thr41) and an increased accumulation of β-catenin in the lungs and kidneys of pigs infected with a highly virulent strain of H. parasuis. In PK-15 and NPTr cells, it was also confirmed that infection with a high-virulence strain of H. parasuis induced cytoplasmic accumulation and nuclear translocation of β-catenin. H. parasuis infection caused a sharp degradation of E-cadherin and an increase of the epithelial cell monolayer permeability, as well as a broken interaction between β-catenin and E-cadherin dependent on Wnt/β-catenin signaling pathway. Moreover, Wnt/β-catenin signaling pathway also contributed to the initiation of epithelial-mesenchymal transition (EMT) during high-virulence strain of H. parasuis infection with expression changes of epithelial/mesenchymal markers, increased migratory capabilities as well as the morphologically spindle-like switch in PK-15 and NPTr cells. Therefore, we originally speculated that H. parasuis infection activates the canonical Wnt/β-catenin signaling pathway leading to a disruption of the epithelial barrier, altering cell structure and increasing cell migration, which results in severe acute systemic infection characterized by fibrinous polyserositis during H. parasuis infection

    An Analytical Framework for Runtime of a Class of Continuous Evolutionary Algorithms

    Although there have been many studies on the runtime of evolutionary algorithms in discrete optimization, relatively few theoretical results have been proposed on continuous optimization, such as evolutionary programming (EP). This paper proposes an analysis of the runtime of two EP algorithms based on Gaussian and Cauchy mutations, using an absorbing Markov chain. Given a constant variation, we calculate the runtime upper bound of special Gaussian mutation EP and Cauchy mutation EP. Our analysis reveals that the upper bounds are impacted by individual number, problem dimension number n, searching range, and the Lebesgue measure of the optimal neighborhood. Furthermore, we provide conditions whereby the average runtime of the considered EP can be no more than a polynomial of n. The condition is that the Lebesgue measure of the optimal neighborhood is larger than a combinatorial calculation of an exponential and the given polynomial of n

    The Effect of the Vertex Angles of Wedged Indenters on Deformation during Nanoindentation

    In order to study the effect of the angle of wedged indenters during nanoindentation, indenters with half vertex angles of 60°, 70° and 80° are used for the simulations of nanoindentation on FCC aluminum (Al) bulk material by the multiscale quasicontinuum method (QC). The load-displacement responses, the strain energy-displacement responses, and hardness of Al bulk material are obtained. Besides, atomic configurations for each loading situation are presented. We analyze the drop points in the load-displacement responses, which correspond to the changes of microstructure in the bulk material. From the atom images, the generation of partial dislocations as well as the nucleation and the emission of perfect dislocations have been observed with wedged indenters of half vertex angles of 60° and 70°, but not 80°. The stacking faults move beneath the indenter along the direction [ 1 1 ¯ 0 ] . The microstructures of residual displacements are also discussed. In addition, hardness of the Al bulk material is different in simulations with wedged indenters of half vertex angles of 60° and 70°, and critical hardness in the simulation with the 70° indenter is bigger than that with the 60° indenter. The size effect of hardness in plastic wedged nanoindentation is observed. There are fewer abrupt drops in the strain energy-displacement response than in the load-displacement response, and the abrupt drops in strain energy-displacement response reflect the nucleation of perfect dislocations or extended dislocations rather than partial dislocations. The wedged indenter with half vertex angle of 70° is recommended for investigating dislocations during nanoindentation
