    Manganese is widely used in many fields. Many efforts have been made to recover manganese from low-grade pyrolusite due to the depletion of high-grade manganese ore. Thus, it is of practical significance to develop a clean, energy-saving and environmentally friendly technical route to reduce the low-grade pyrolusite. The reported results show that biomass wastes from crops, crop waste, wood and wood waste are environmentally friendly, energy-saving, and low-cost reducing agents for roasting reduction of low-grade pyrolusite. Kinetics of the reduction reactions is necessary for an efficient design of biomass reduction of pyrolusite. Therefore, it is important to look for a general kinetics equation to describe the reduction of pyrolusite by different kinds of biomass, because there is a wide variety of biomass wastes, meaning that it is impossible to investigate the kinetics for each biomass waste. In this paper, thermal gravimetric analysis and differential thermal analysis were applied to study the overall reduction kinetics of pyrolusite using a mixture of hemicellulose and lignin, two major components of biomass. Overall reduction process is the overlap of the respective reduction processes. A new empirical equation based on the Johnsonā€“Mehlā€“Avrami equation can be used to describe the respective reduction kinetics using hemicellulose and lignin as reductants, and the corresponding apparent activation energy is 30.14 kJ molāˆ’1 and 38.91 kJ molāˆ’1, respectively. The overall kinetic model for the reduction of pyrolusite by the mixture of hemicellulose and lignin can be simulated by the summation of the respective kinetics by considering their mass-loss fractions, while a unit step function was used to avoid the invalid conversion data. The obtained results in this work are necessary to understand the biomass reduction of pyrolusite and provide valuable assistance in the development of a general kinetics equation.Ukupna kinetika redukcije piroluzita istraživana je termogravimetrijom i diferencijalnom termogravimetrijom. Kao redukcijsko sredstvo upotrijebljeni su hemiceluloza i lignin, glavni sastojci poljoprivrednog biljnog biootpada, drva i drvnog otpada. Ukupnu redukciju čine isprepleteni pojedinačni redukcijski procesi. Kinetika redukcije piroluzita smjesom hemiceluloze i lignina može se opisati novom empirijskom jednadžbom temeljenoj na jednadžbi Johnsonā€“Mehlā€“Avrami, a odgovarajuća prividna energija aktivacije iznosi 30.14 kJ molāˆ’1, odnosno 38.91 kJ molāˆ’1.     Movable Fiber-Integrated Hybrid Plasmonic Waveguide on Metal Film

    A waveguide structure consisting of a tapered nanofiber on a metal film is proposed and analyzed to support highly localized hybrid plasmonic modes. The hybrid plasmonic mode can be efficiently excited through the in-line tapered fiber based on adiabatic conversion and collected by the same fiber, which is very convenient in the experiment. Due to the ultrasmall mode area of plasmonic mode, the local electromagnetic field is greatly enhanced in this movable waveguide, which is potential for enhanced coherence light emitter interactions, such as waveguide quantum electrodynamics, single emitter spectrum and nonlinear optics

    Molecular Cloning of the Genes Encoding the PR55/BĪ²/Ī“ Regulatory Subunits for PP-2A and Analysis of Their Functions in Regulating Development of Goldfish, Carassius auratus

    The protein phosphatase-2A (PP-2A), one of the major phosphatases in eukaryotes, is a heterotrimer, consisting of a scaffold A subunit, a catalytic C subunit and a regulatory B subunit. Previous studies have shown that besides regulating specific PP-2A activity, various B subunits encoded by more than 16 different genes, may have other functions. To explore the possible roles of the regulatory subunits of PP-2A in vertebrate development, we have cloned the PR55/B family regulatory subunits: Ī² and Ī“, analyzed their tissue specific and developmental expression patterns in Goldfish ( Carassius auratus). Our results revealed that the full-length cDNA for PR55/BĪ² consists of 1940 bp with an open reading frame of 1332 nucleotides coding for a deduced protein of 443 amino acids. The full length PR55/BĪ“ cDNA is 2163 bp containing an open reading frame of 1347 nucleotides encoding a deduced protein of 448 amino acids. The two isoforms of PR55/B display high levels of sequence identity with their counterparts in other species. The PR55/BĪ² mRNA and protein are detected in brain and heart. In contrast, the PR55/BĪ“ is expressed in all 9 tissues examined at both mRNA and protein levels. During development of goldfish, the mRNAs for PR55/BĪ² and PR55/BĪ“ show distinct patterns. At the protein level, PR55/BĪ“ is expressed at all developmental stages examined, suggesting its important role in regulating goldfish development. Expression of the PR55/BĪ“ anti-sense RNA leads to significant downregulation of PR55/BĪ“ proteins and caused severe abnormality in goldfish trunk and eye development. Together, our results suggested that PR55/BĪ“ plays an important role in governing normal trunk and eye formation during goldfish development

    Using histogram analysis of the intrinsic brain activity mapping to identify essential tremor

    BackgroundEssential tremor (ET) is one of the most common movement disorders. Histogram analysis based on brain intrinsic activity imaging is a promising way to identify ET patients from healthy controls (HCs) and further explore the spontaneous brain activity change mechanisms and build the potential diagnostic biomarker in ET patients.MethodsThe histogram features based on the Resting-state functional magnetic resonance imaging (Rs-fMRI) data were extracted from 133 ET patients and 135 well-matched HCs as the input features. Then, a two-sample t-test, the mutual information, and the least absolute shrinkage and selection operator methods were applied to reduce the feature dimensionality. Support vector machine (SVM), logistic regression (LR), random forest (RF), and k-nearest neighbor (KNN) were used to differentiate ET and HCs, and classification performance of the established models was evaluated by the mean area under the curve (AUC). Moreover, correlation analysis was carried out between the selected histogram features and clinical tremor characteristics.ResultsEach classifier achieved a good classification performance in training and testing sets. The mean accuracy and area under the curve (AUC) of SVM, LR, RF, and KNN in the testing set were 92.62%, 0.948; 92.01%, 0.942; 93.88%, 0.941; and 92.27%, 0.939, respectively. The most power-discriminative features were mainly located in the cerebello-thalamo-motor and non-motor cortical pathways. Correlation analysis showed that there were two histogram features negatively and one positively correlated with tremor severity.ConclusionOur findings demonstrated that the histogram analysis of the amplitude of low-frequency fluctuation (ALFF) images with multiple machine learning algorithms could identify ET patients from HCs and help to understand the spontaneous brain activity pathogenesis mechanisms in ET patients

    Decreased Intrinsic Functional Connectivity of the Salience Network in Drug-NaĆÆve Patients With Obsessive-Compulsive Disorder

    Obsessive-compulsive disorder (OCD) patients have difficulty in switching between obsessive thought and compulsive behavior, which may be related to the dysfunction of the salience network (SN). However, little is known about the changes in intra- and inter- intrinsic functional connectivity (iFC) of the SN in patients with OCD. In this study, we parceled the SN into 19 subregions and investigated iFC changes for each of these subregions in 40 drug-naĆÆve patients with OCD and 40 healthy controls (HCs) using seed-based functional connectivity resting-state functional magnetic resonance imaging (rs-fMRI). We found that patients with OCD exhibited decreased iFC strength between subregions of the SN, as well as decreased inter-network connectivity between SN and DMN, and ECN. These findings highlight a specific alteration in iFC patterns associated with SN in patients with OCD and provide new insights into the dysfunctional brain organization of the SN in patients with OCD

    Chronic Pseudomonas aeruginosa (PA) infection significantly contributes to morbidity and mortality in bronchiectasis patients. Initiating antibiotics early may lead to the eradication of PA. Here we outline the design of a trial (ERASE; NCT06093191) assessing the efficacy and safety of inhaled tobramycin, alone or with oral ciprofloxacin, in bronchiectasis patients with a new isolation of PA. This multicentre, 2Ɨ2 factorial randomised, double-blind, placebo-controlled, parallel-group trial includes a 2-week screening period, a 12-week treatment phase (with a combination of ciprofloxacin or a placebo at initial 2ā€…weeks) and a 24-week follow-up. 364 adults with bronchiectasis and a new PA isolation will be randomly assigned to one of four groups: placebo (inhaled saline and ciprofloxacin placebo twice daily), ciprofloxacin alone (750ā€…mg ciprofloxacin and inhaled saline twice daily), inhaled tobramycin alone (inhaled 300ā€…mg tobramycin and ciprofloxacin placebo twice daily) or a combination of both drugs (inhaled 300ā€…mg tobramycin and 750ā€…mg ciprofloxacin twice daily). The primary objective of this study is to assess the proportion of patients successfully eradicating PA in each group by the end of the study. Efficacy will be evaluated based on the eradication rate of PA at other time points (12, 24 and 36ā€…weeks), the occurrence of exacerbations and hospitalisations, time to first pulmonary exacerbations, patient-reported outcomes, symptom measures, pulmonary function tests and the cost of hospitalisations. To date no randomised trial has evaluated the benefit of different PA eradication strategies in bronchiectasis patients. The ERASE trial will therefore generate crucial data to inform future clinical guidelines.</p

    Yersinia pseudotuberculosis Exploits CD209 Receptors for Promoting Host Dissemination and Infection

    Yersinia pseudotuberculosis is a Gram-negative enteropathogen and causes gastrointestinal infections. It disseminates from gut to mesenteric lymph nodes (MLNs), spleen, and liver of infected humans and animals. Although the molecular mechanisms for dissemination and infection are unclear, many Gram-negative enteropathogens presumably invade the small intestine via Peyer's patches to initiate dissemination. In this study, we demonstrate that Y. pseudotuberculosis utilizes its lipopolysaccharide (LPS) core to interact with CD209 receptors, leading to invasion of human dendritic cells (DCs) and murine macrophages. These Y. pseudotuberculosis CD209 interactions result in bacterial dissemination to MLNs, spleens, and livers of both wild-type and Peyer's patch-deficient mice. The blocking of the Y. pseudotuberculosis CD209 interactions by expression of 0-antigen and with oligosaccharides reduces infectivity. Based on the well-documented studies in which HIV-CD209 interaction leads to viral dissemination, we therefore propose an infection route for Y. pseudotuberculosis where this pathogen, after penetrating the intestinal mucosal membrane, hijacks the Y. pseudotuberculosis CD209 interaction antigen-presenting cells to reach their target destinations, MLNs, spleens, and livers.Peer reviewe

    Natural Coevolution of Tumor and Immunoenvironment in Glioblastoma.

    Isocitrate dehydrogenase (IDH) wild-type glioblastoma (GBM) has a dismal prognosis. A better understanding of tumor evolution holds the key to developing more effective treatment. Here we study GBM\u27s natural evolutionary trajectory by using rare multifocal samples. We sequenced 61,062 single cells from eight multifocal IDH wild-type primary GBMs and defined a natural evolution signature (NES) of the tumor. We show that the NES significantly associates with the activation of transcription factors that regulate brain development, including MYBL2 and FOSL2. Hypoxia is involved in inducing NES transition potentially via activation of the HIF1A-FOSL2 axis. High-NES tumor cells could recruit and polarize bone marrow-derived macrophages through activation of the FOSL2-ANXA1-FPR1/3 axis. These polarized macrophages can efficiently suppress T-cell activity and accelerate NES transition in tumor cells. Moreover, the polarized macrophages could upregulate CCL2 to induce tumor cell migration. SIGNIFICANCE: GBM progression could be induced by hypoxia via the HIF1A-FOSL2 axis. Tumor-derived ANXA1 is associated with recruitment and polarization of bone marrow-derived macrophages to suppress the immunoenvironment. The polarized macrophages promote tumor cell NES transition and migration. This article is highlighted in the In This Issue feature, p. 2711
