    Assessing the impacts of climate change on domestic crop production: Experience and perception of local farmers in North Malaita, Solomon Islands.

    The aim of this research was to: 1) gather farmers' experiences and observations of climate change impacts on domestic crop yields during the last thirty years (1988-2018); 2) study climatic projections (2050) and their potential impacts on Sikwafata and Malu'u's crops; and 3) understand the knowledge engaged with, and adaptations taken, to avoid susceptibility to climate change. The data was collected using the snowball approach, in which the researcher contacts informants using information provided by previous informants. A thematic analysis was performed using NVIVO 10 software. For both sites, soil samples were taken to determine moisture content. Farmers reported changes in rainfall patterns (73.5%) and temperature (44.9%), while 26.5% indicated no change in rainfall or temperature (55.1%). About 83.7% of farmers anticipate increasing temperatures and rainfall to occur by 2050, with extreme impacts on crop productivity. The majority of the gardens (87.5%) were on hillsides, while 8.5% were on the lowland, with an average of three garden per farmer. The gardens (57.0%) were on rotational sites that had been abandoned for two to five years, while 43.0% had never been rotated. In comparison to Malu'u moisture content (47.4%), Sikwafata moisture content is 66.4% higher. Furthermore, farmers are aware of adaptation measures to reduce the risk of crop failure. However, the absence of scientific information on climate change and agricultural resilience has increased the vulnerability to extreme climate-related events harming food security and nutrition. Climate change will undoubtedly intensify, resulting in a global and local drop in crop production, thus compromising livelihoods in the future

    “You Might as well Call it Planet of the Sioux”: Indigenous Youth, Imagination, and Decolonization

    Colonial institutions, such as residential schools, suppressed First Nations peoples’ imaginations, punishing those who sought to exercise their imaginations. Creating imaginary spaces is an important aspect of the process of decolonization and includes the reclamation of traditional modes of relationships in new forms, the co-creation of new possibilities, and the transformation of political and personal histories. In this article, we describe a workshop with First Nations youth, focusing on two arts-based activities used to evoke imaginative spaces for First Nations youth to explore, critique, and re-imagine their histories, current realities, and futures in a safe and comfortable environment. The process of imagery and imagining facilitated awareness of things not easily expressed in words as youths’ imaginative ideas emerged through their drawings. Through these activities, youth expressed their view of healthy communities that included cultural traditions and leadership. They described leadership as enacting good communication, organization, protection, and maintenance of cultural traditions, as well as providing guidance and defending the rights of the community. These imagined possibilities provided meaningful blueprints that youth can use, alter, and be inspired by as they move towards being leaders of healthy communities

    Policy Brief 2: Building Community Resilience

    Building resilience in Ocean Cities of the Pacific region is key to converting challenges and risks communities face into opportunities for systemic transformation toward sustainable development. Island settlements at the nexus of ocean health, climate change and urban development will need to build anticipatory, adaptive, absorptive and transformative capacity in the face of complex stressors. The interlinked challenges and resource constraints in rapidly growing Ocean Cities in the Pacific means that the ability to absorb shocks and bounce back relevant partners and stakeholders to promote equitable opportunities for all people in urban communities to access resources and use them sustainably, and to apply standardized methods contextualized for the Pacific to assess the relative vulnerability of communities in Ocean Cities. It also recognizes the importance of promoting stronger governance regimes within urban communities, aligning these with the policies of local government, and encouraging partnerships between urban and rural communities. This second policy brief in the Ocean Cities series offers guidance in this direction

    All-codon scanning identifies p53 cancer rescue mutations

    In vitro scanning mutagenesis strategies are valuable tools to identify critical residues in proteins and to generate proteins with modified properties. We describe the fast and simple All-Codon Scanning (ACS) strategy that creates a defined gene library wherein each individual codon within a specific target region is changed into all possible codons with only a single codon change per mutagenesis product. ACS is based on a multiplexed overlapping mutagenesis primer design that saturates only the targeted gene region with single codon changes. We have used ACS to produce single amino-acid changes in small and large regions of the human tumor suppressor protein p53 to identify single amino-acid substitutions that can restore activity to inactive p53 found in human cancers. Single-tube reactions were used to saturate defined 30-nt regions with all possible codon changes. The same technique was used in 20 parallel reactions to scan the 600-bp fragment encoding the entire p53 core domain. Identification of several novel p53 cancer rescue mutations demonstrated the utility of the ACS approach. ACS is a fast, simple and versatile method, which is useful for protein structure–function analyses and protein design or evolution problems

    Towards a global partnership model in interprofessional education for cross-sector problem-solving

    Objectives A partnership model in interprofessional education (IPE) is important in promoting a sense of global citizenship while preparing students for cross-sector problem-solving. However, the literature remains scant in providing useful guidance for the development of an IPE programme co-implemented by external partners. In this pioneering study, we describe the processes of forging global partnerships in co-implementing IPE and evaluate the programme in light of the preliminary data available. Methods This study is generally quantitative. We collected data from a total of 747 health and social care students from four higher education institutions. We utilized a descriptive narrative format and a quantitative design to present our experiences of running IPE with external partners and performed independent t-tests and analysis of variance to examine pretest and posttest mean differences in students’ data. Results We identified factors in establishing a cross-institutional IPE programme. These factors include complementarity of expertise, mutual benefits, internet connectivity, interactivity of design, and time difference. We found significant pretest–posttest differences in students’ readiness for interprofessional learning (teamwork and collaboration, positive professional identity, roles, and responsibilities). We also found a significant decrease in students’ social interaction anxiety after the IPE simulation. Conclusions The narrative of our experiences described in this manuscript could be considered by higher education institutions seeking to forge meaningful external partnerships in their effort to establish interprofessional global health education

    Large-scale mapping of human protein–protein interactions by mass spectrometry

    Mapping protein–protein interactions is an invaluable tool for understanding protein function. Here, we report the first large-scale study of protein–protein interactions in human cells using a mass spectrometry-based approach. The study maps protein interactions for 338 bait proteins that were selected based on known or suspected disease and functional associations. Large-scale immunoprecipitation of Flag-tagged versions of these proteins followed by LC-ESI-MS/MS analysis resulted in the identification of 24 540 potential protein interactions. False positives and redundant hits were filtered out using empirical criteria and a calculated interaction confidence score, producing a data set of 6463 interactions between 2235 distinct proteins. This data set was further cross-validated using previously published and predicted human protein interactions. In-depth mining of the data set shows that it represents a valuable source of novel protein–protein interactions with relevance to human diseases. In addition, via our preliminary analysis, we report many novel protein interactions and pathway associations

    A Multicentre Molecular Analysis of Hepatitis B and Blood-Borne Virus Coinfections in Viet Nam

    Hepatitis B (HBV) infection is endemic in Viet Nam, with up to 8.4 million individuals estimated to be chronically infected. We describe results of a large, multicentre seroepidemiological and molecular study of the prevalence of HBV infection and blood-borne viral coinfections in Viet Nam. Individuals with varying risk factors for infection (n = 8654) were recruited from five centres; Ha Noi, Hai Phong, Da Nang, Khanh Hoa and Can Tho. A mean prevalence rate of 10.7% was observed and levels of HBsAg were significantly higher in injecting drug users (IDUs) (17.4%, n = 174/1000) and dialysis patients (14.3%, n = 82/575) than in lower-risk groups (9.4%; p<0.001). Coinfection with HIV was seen in 28% of HBV-infected IDUs (n = 49/174) and 15.2% of commercial sex workers (CSWs; n = 15/99). HCV infection was present in 89.8% of the HBV-HIV coinfected IDUs (n = 44/49) and 40% of HBV-HIV coinfected CSWs (n = 16/40). Anti-HDV was detected in 10.7% (n = 34/318) of HBsAg positive individuals. Phylogenetic analysis of HBV S gene (n = 187) showed a predominance of genotype B4 (82.6%); genotypes C1 (14.6%), B2 (2.7%) and C5 (0.5%) were also identified. The precore mutation G1896A was identified in 35% of all specimens, and was more frequently observed in genotype B (41%) than genotype C (3%; p<0.0001). In the immunodominant ‘a’ region of the surface gene, point mutations were identified in 31% (n = 58/187) of sequences, and 2.2% (n = 4/187) and 5.3% (n = 10/187) specimens contained the major vaccine escape mutations G145A/R and P120L/Q/S/T, respectively. 368 HBsAg positive individuals were genotyped for the IL28B SNP rs12979860 and no significant association between the IL28B SNP and clearance of HBsAg, HBV viral load or HBeAg was observed. This study confirms the high prevalence of HBV infection in Viet Nam and also highlights the significant levels of blood-borne virus coinfections, which have important implications for hepatitis-related morbidity and development of effective management strategies
