    A Dominant Complement Fixation Pathway for Pneumococcal Polysaccharides Initiated by SIGN-R1 Interacting with C1q

    The intricate system of serum complement proteins provides resistance to infection. A pivotal step in the complement pathway is the assembly of a C3 convertase, which digests the C3 complement component to form microbial binding C3 fragments recognized by leukocytes. The spleen and C3 provide resistance against blood-borne S. pneumoniae infection. To better understand the mechanisms involved, we studied SIGN-R1, a lectin that captures microbial polysaccharides in spleen. Surprisingly, conditional SIGN-R1 knockout mice developed deficits in C3 catabolism when given S. pneumoniae or its capsular polysaccharide intravenously. There were marked reductions in proteolysis of serum C3, deposition of C3 on organisms within SIGN-R1+ spleen macrophages, and formation of C3 ligands. We found that SIGN-R1 directly bound the complement C1 subcomponent, C1q, and assembled a C3 convertase, but without the traditional requirement for either antibody or factor B. The transmembrane lectin SIGN-R1 therefore contributes to innate resistance by an unusual C3 activation pathway

    Two ways teachers can develop greater harmonious passion

    Background Passion is highly prized. The Dualistic Model of Passion provides a general roadmap for how people develop passion, at least under conditions in which they can freely schedule their daily activity, abandon activities they no longer value, and have flexible time to invest as they see fit. But teaching is a different, because many aspects of this activity are fixed and pre-set by circumstances. Recognizing this unique condition of classroom PE teaching, we designed the present investigation to test the merits of two antecedents teachers can utilize to develop greater (harmonious) passionā€”namely, participate in an autonomy-supportive teaching workshop (Study 1) and incorporate intrinsic instructional goals into their lesson plans (Study 2). Purpose The over-arching purpose was to investigate the potential of two malleable and personally controllable catalysts to greater harmonious teaching passion. Method Study 1 used a randomized control trial. We randomly assigned 104 Korean PE teachers to participate (or not) in an autonomy-supportive teaching (AST) workshop. PE teachers self-reported their harmonious and obsessive passion at the beginning, middle, and end of an academic year. Study 2 used a longitudinal research design and a sample that included both PE and non-PE teachers. These 134 secondary-grade level teachers self-reported their intrinsic instructional goals, autonomy-supportive teaching, and harmonious and obsessive passion across three waves. Results In Study 1, a repeated-measures ANCOVA showed that teachers in the experimental condition, compared to teachers in the control condition, showed a longitudinal increase in autonomy-supportive teaching and harmonious passion and a longitudinal decrease in obsessive passion. A mediation analysis confirmed that participants in the AST workshop experienced greater autonomy-supportive teaching that then explained their greater harmonious passion and lesser obsessive passion. In Study 2, a structural equation modeling analysis showed that adopting intrinsic instructional goals early in the year longitudinally increased harmonious passion (but did not decrease obsessive passion). A mediation analysis confirmed that teachers who more adopted intrinsic instructional goals experienced greater autonomy-supportive teaching that then explained their greater harmonious passion. Conclusion Teachers can gain personal control over their harmonious passion. They can do this through greater autonomy-supportive teaching. And teachers can become more autonomy supportive in two ways: Participate in an expert-provided professional development experience (Study 1) or incorporate intrinsic instructional goals into the delivery of their instruction (Study 2). Additional ways teachers can develop harmonious passion may be possible, so we encourage future research to continue this search with additional samples, such as sport coaches

    Near-Infrared Spectroscopy of Carbon-Enhanced Metal-Poor Stars. I. A SOAR/OSIRIS Pilot Study

    We report on an abundance analysis for a pilot study of seven Carbon-Enhanced Metal-Poor (CEMP) stars, based on medium-resolution optical and near-infrared spectroscopy. The optical spectra are used to estimate [Fe/H], [C/Fe], [N/Fe], and [Ba/Fe] for our program stars. The near-infrared spectra, obtained during a limited early science run with the new SOAR 4.1m telescope and the Ohio State Infrared Imager and Spectrograph (OSIRIS), are used to obtain estimates of [O/Fe] and 12C/13C. The chemical abundances of CEMP stars are of importance for understanding the origin of CNO in the early Galaxy, as well as for placing constraints on the operation of the astrophysical s-process in very low-metallicity Asymptotic Giant Branch (AGB) stars. This pilot study includes a few stars with previously measured [Fe/H], [C/Fe], [N/Fe],[O/Fe], 12C/13C, and [Ba/Fe], based on high-resolution optical spectra obtained with large-aperture telescopes. Our analysis demonstrates that we are able to achieve reasonably accurate determinations of these quantities for CEMP stars from moderate-resolution optical and near-infrared spectra. This opens the pathway for the study of significantly larger samples of CEMP stars in the near future. Furthermore, the ability to measure [Ba/Fe] for (at least the cooler) CEMP stars should enable one to separate stars that are likely to be associated with s-process enhancements (the CEMP-s stars) from those that do not exhibit neutron-capture enhancements (the CEMP-no stars).Comment: 27 pages, including 5 tables, 6 figures, accepted for publication in The Astronomical Journa

    Goal Content Analysis for Middle and High School Students With Disabilities

    Goal setting is associated with self-determination and student involvement in learning. Middle and high school students receiving special education services and corresponding special education teachers across six states were asked to name goals on which the students were working. A comparison of the academic, transition, and social goals named by 332 students was conducted to determine differences between groups. Results are reported by school level and the ability levels of students, evaluating both student- and teacher-reported goals from a written evaluation scenario. Relative grade level in school was shown to be a significant factor for content variation but not ability level.     Hippocampal glucose uptake as a surrogate of metabolic change of microglia in Alzheimers disease

    Abstract Background Dynamically altered microglia play an important role in the progression of Alzheimers disease (AD). Here, we found a close association of the metabolic reconfiguration of microglia with increased hippocampal glucose uptake on [18F]fluorodeoxyglucose (FDG) PET. Methods We used an AD animal model, 5xFAD, to analyze hippocampal glucose metabolism using both animal FDG PET and ex vivo FDG uptake test. Cells of the hippocampus were isolated to perform single-cell RNA-sequencing (scRNA-seq). The molecular features of cells associated with glucose metabolism were analyzed at a single-cell level. In order to apply our findings to human brain imaging study, brain FDG PET data obtained from the Alzheimers Disease Neuroimaging Initiative were analyzed. FDG uptake in the hippocampus was compared according to the diagnosis, AD, mild cognitive impairment, and controls. The correlation analysis between hippocampal FDG uptake and soluble TREM2 in cerebrospinal fluid was performed. Results In the animal study, 8- and 12-month-old 5xFAD mice showed higher FDG uptake in the hippocampus than wild-type mice. Cellular FDG uptake tests showed that FDG activity in hippocampal microglia was increased in the AD model, while FDG activity in non-microglial cells of the hippocampus was not different between the AD model and wild-type. scRNA-seq data showed that changes in glucose metabolism signatures including glucose transporters, glycolysis and oxidative phosphorylation, mainly occurred in microglia. A subset of microglia with higher glucose transporters with defective glycolysis and oxidative phosphorylation was increased according to disease progression. In the human imaging study, we found a positive association between soluble TREM2 and hippocampal FDG uptake. FDG uptake in the hippocampus at the baseline scan predicted mild cognitive impairment conversion to AD. Conclusions We identified the reconfiguration of microglial glucose metabolism in the hippocampus of AD, which could be evaluated by FDG PET as a feasible surrogate imaging biomarker for microglia-mediated inflammation

    Measuring patient acuity and nursing care needs in South Korea: application of a new patient classification system

    Background An accurate and reliable patient classification system (PCS) can help inform decisions regarding adequate assignments for nurse staffing. This study aimed to evaluate the criterion validity of the Asan Patient Classification System (APCS), a new tertiary hospital-specific PCS, by comparing its rating and total scores with those of KPCS-1 and KPCS-GW for measuring patient activity and nursing needs. Methods We performed a retrospective analysis of the medical records of 50,314 inpatients admitted to the general wards of a tertiary teaching hospital in Seoul, South Korea in March, June, September, and December 2019. Spearmanā€™s correlation and Kappa statistics according to quartiles were calculated to examine the criterion validity of the APCS compared with the KPCS-1 and KPCS-GW. Results The average patient classification score was 28.3 points for APCS, 25.7 points for KPCS-1, and 21.6 points for KPCS-GW. The kappa value between APCS and KPCS-1 was 0.91 (95% CI:0.9072, 0.9119) and that between APCS and KPCS-GW was 0.88 (95% CI:0.8757, 0.8810). Additionally, Spearman's correlation coefficients among APCS, KPCS-1, and KPCS-GW showed a very strong correlation. However, 10.8% of the participantsā€™ results were inconsistent, and KPCS-1 tended to classify patients into groups with lower nursing needs compared to APCS. Conclusion This study showed that electronic health record-generated APCS can provide useful information on patientsā€™ severity and nursing activities to measure workload estimation. Additional research is needed to develop and implement a real-world EHR-based PCS system to accommodate for direct and indirect nursing care while considering diverse population and dynamic healthcare system

    Effects of a safflower tea supplement on antioxidative status and bone markers in postmenopausal women

    We conducted this study to examine the effects of safflower seed granular tea containing physiologically active polyphenols on antioxidative activities and bone metabolism. Forty postmenopausal women ages 49 to 64-years were recruited from Daegu and Gyeongbuk and were randomly assigned to either a safflower tea supplement (Saf-tea) group (n = 27) or a placebo group (n = 13). The Saf-tea group received 20 g of safflower seed granule tea per day containing a 13% ethanol extract of defatted safflower seeds, whereas the placebo group received a similar type of tea that lacked the ethanol extract. No significant changes in nutrient intake for either the placebo or Saf-tea groups were observed before or after the study period, except vitamin A intake increased after 6 months in the Saf-tea group. Dietary phytoestrogen intakes were similar in the Saf-tea group (60.3 mg) and placebo group (52.5 mg). Significant increases in plasma genistein and enterolactone were observed in the Saf-tea group. After 6 months of supplementation, serum levels of antioxidant vitamins such as Ī±-tocopherol and ascorbic acid increased significantly, and TBARS levels decreased in the Saf-tea group compared to the placebo group. Serum osteocalcin levels were reduced (P < 0.05) in the Saf-tea group after 6 months, whereas serum osteocalcin did not change in the placebo group. Urinary deoxypyridinoline/creatinine excretion was not different between the two groups at baseline, and did not change in either group after 6 months. Bone mineral density decreased significantly in the placebo group (P < 0.01) but not in the supplemented group. It was concluded that polyphenols (72 mg/day), including serotonin derivatives, in the Saf-tea had both antioxidant and potential bone protecting effects in postmenopausal women without liver toxicity
