It does not tied with certain rules, but still using the fascinating language. Free verse has a structure similar to the prose but can be distinguished by the delivery of language where there are a lot of implicit meaning, because a poem says one thing and means another. The investigated issues in this researches are what kind of symbols associated with the poet's life contained in six poems at Shinshoo Sukecchi Haru to Shura poetry collection by Miyazawa Kenji; how the hermeneutic reading, and what kind of internal structures are found on six poems. The method used in this thesis is Library Research. That is, data acquisition method, data analysis method, and presentation of the result of data analysis method. This research uses the science of semiotics that has been described by Charles Sanders Peirce using triangular trichotomous/triadic that is, representamen (R) - the object (O) - interpretant (I). The second phase describes the hermeneutic interpretation, a poem re-reading or rhyme from the beginning to the end with hermeneutic interpretation/retroactive to understand the entire contents contained in literary works per parts. The third phase seek the intrinsic elements of the internal structure through the structural theory described by Herman J. Waluyo, to understand better the content is and what was written by the poet. The research’s results showes that there are 6 blank symbols, 23 natural symbols, and 11 private symbols. The hermeneutic majority describes the poet’s journey of miserable life and ended with the sincerity of his past. Meanwhile, the internal structure majority describes the poet’s sacrifice, pray, sorrow, destruction, faith, and peace.     Exercise hyperpnea and hypercapnic ventilatory responses in women

    SummaryWe studied the relationship between exercise hyperpnea (i.e., ventilatory dynamics) at the onset of exercise and hypercapnic ventilatory response (HCVR), and their differences between the follicular (FP) and luteal (LP) phases of the menstrual cycle in six healthy females. HCVR was tested under three O2 conditions: hyperoxia (FiO2=1.0), normoxia (0.21), and hypoxia (0.12). HCVR was defined as the relationship between the end-tidal PCO2 and minute ventilation (V˙E) using the regression line of the CO2 slope and a mimetically apneic threshold of CO2. HCVR provocation and measurements were conducted using an inspired CO2 concentration of up to approximately 8mmHg higher than the end-tidal PCO2 level of basal isocapnic the end-tidal PCO2 at each menstrual both the slope and threshold in HCVR showed no statistically significant difference between LP and FP under any inspired FiO2 conditions. In the case of exercise hyperpnea during the onset of submaximal exercise, the mean response time (MRT) in V˙E dynamics showed no significant difference between LP and FP. Consequently, MRT in V˙E response was not related to the slope in HCVR. During steady-state exercise, even though the V˙E/V˙CO2 showed no significance between LP and FP, V˙E/V˙CO2 was significantly related to the slope in HCVR (r=0.59, P<0.05). Exercise ventilation (i.e., V˙E/V˙CO2) would partly be adjusted by the enhancement of the chemoreflex drive to CO2 only during the steady-state exercise

    Lower FEV1 in non-COPD, nonasthmatic subjects: association with smoking, annual decline in FEV1, total IgE levels, and TSLP genotypes

    Few studies have investigated the significance of decreased FEV1 in non-COPD, nonasthmatic healthy subjects. We hypothesized that a lower FEV1 in these subjects is a potential marker of an increased susceptibility to obstructive lung disease such as asthma and COPD. This was a cross-sectional analysis of 1505 Japanese adults. We divided the population of healthy adults with no respiratory diseases whose FEV1/FVC ratio was ≥70% (n = 1369) into 2 groups according to their prebronchodilator FEV1 (% predicted) measurements: <80% (n = 217) and ≥80% (n = 1152). We compared clinical data – including gender, age, smoking habits, total IgE levels, and annual decline of FEV1 – between these 2 groups. In addition, as our group recently found that TSLP variants are associated with asthma and reduced lung function, we assessed whether TSLP single nucleotide polymorphisms (SNPs) were associated with baseline lung function in non-COPD, nonasthmatic healthy subjects (n = 1368). Although about half of the subjects with lower FEV1 had never smoked, smoking was the main risk factor for the decreased FEV1 in non-COPD, nonasthmatic subjects. However, the subjects with lower FEV1 had a significantly higher annual decline in FEV1 independent of smoking status. Airflow obstruction was associated with increased levels of total serum IgE (P = 0.029) and with 2 functional TSLP SNPs (corrected P = 0.027–0.058 for FEV1% predicted, corrected P = 0.015–0.033 for FEV1/FVC). This study highlights the importance of early recognition of a decreased FEV1 in healthy subjects without evident pulmonary diseases because it predicts a rapid decline in FEV1 irrespective of smoking status. Our series of studies identified TSLP variants as a potential susceptibility locus to asthma and to lower lung function in non-COPD, nonasthmatic healthy subjects, which may support the contention that genetic determinants of lung function influence susceptibility to asthma

    Molecular phylogeny of the rotifers with two Indonesian Brachionus lineages

    The rotifer Brachionus plicatilis is an ecologically and commercially important species, and has been studied in various fields such as population dynamics, ecotoxicology and aging. However, recent studies have revealed that the B. plicatilis lineages involve an unknown number of cryptic species, and the group has been regarded as the Brachionus complex. One cause of this complicated taxonomy is the lack of surveys in the tropical zone, which is characterized by enormous species-richness. Accordingly, in this study we collected two Brachionus rotifers from the Sumatra and Sulawesi Islands, Indonesia, and determined their partial nucleotide sequences of mitochondrial DNA cytochrome c oxidase subunit I gene. Subsequently, we constructed molecular phylogenetic trees with fourteen species/lineages from four genera including the two Indonesian rotifers. The two Indonesian Brachionus rotifers were respectively found to be phylogenetically close to B. ibericus and B. rotundiformis. On the other hand, Japanese B. plicatilis was suggested to be phylogenetically closer to B. Manjavacas, which is proposed to be a new species, than to Spanish B. plicatilis. These results imply that the current taxonomy of the Brachionus is problematic, and a major revision is necessary to establish a reliable taxonomy of this group

    Analysis of DOC and Ram for NSCLC

    Background: Current clinical trials demonstrated that combination regimens comprising chemotherapy and immunotherapy lead to better patient outcomes compared to chemotherapy alone as the first line of treatment for non-small cell lung cancer (NSCLC). In addition, the combination therapy of docetaxel (Doc) and ramucirumab (Ram) was considered one of the standard treatments for advanced or relapsed NSCLC patients. However, little is known about the therapeutic responders of this combination therapy among previously treated NSCLC patients. In the present study, we aimed to identify predictive factors for therapeutic response, including programmed death-ligand 1 (PD-L1) expression in tumors, for Doc treatment in combination with Ram. Methods: We retrospectively analyzed a total of 135 advanced or relapsed NSCLC patients who were refractory to platinum-based chemotherapy at eleven institutions in Japan between July 2016 and November 2018. Results: Our observations showed that PD-L1 expression in tumors is not associated with the efficacy of combined therapy of Doc and Ram in previously treated NSCLC patients. Analysis of the patient clinical profiles indicated that prior treatment with immune checkpoint inhibitors (ICIs) is a reliable predictor for the good progression-free survival (PFS) to this combination therapy (P=0.041). Conclusions: Our retrospective study indicated that combination regimens comprising chemotherapy and ICIs followed by Doc and Ram could be an optimal therapeutic option for NSCLC patients regardless of the PD-L1 status of tumors. Further investigations are required to strengthen clinical evidence demonstrating the effectiveness of the combination therapy of Doc plus Ram in previously treated NSCLC patients

    Frequency of CD45RO+ subset in CD4+CD25high regulatory T cells associated with progression of hepatocellular carcinoma

    金沢大学医薬保健研究域医学系The purpose of this study was to assess the properties of CD4+CD25high/low/negative T cell subsets and analyze their relation with dendritic cells (DCs) in patients with hepatocellular carcinoma (HCC). In HCC patients, the prevalence of CD45RO+ cells in CD4+CD25high T cells was increased and associated with higher frequencies of plasmacytoid DCs. Larger proportions of this T cell subset were detected in the patients with larger tumor burdens. These results suggest that increased frequencies of the CD45RO+ subset in CD4+CD25high Tregs in HCC patients may establish the immunosuppressive environment cooperatively with tolerogenic plasmacytoid DCs to promote disease progression of liver cancer. © 2011

    Genetic association of the functional CDHR3 genotype with early-onset adult asthma in Japanese populations

    BackgroundRecent studies have demonstrated that a coding SNP (rs6967330, Cys529→Tyr) in cadherin-related family member 3 (CDHR3), which was previously associated with wheezing illness and hospitalizations in infancy, could support efficient human rhinovirus C (RV-C) entry and replication. Here, we sought to examine the genetic contribution of this variant to the development of adult asthma.MethodsWe performed a candidate gene case–control association study of 2 independent Japanese populations (a total of 3366 adults). The odds ratios (ORs) for association of the A allele at rs6967330 with adult asthma were calculated according to age at onset of asthma. In addition, the effect of the CDHR3 genotype on the development of specific asthma phenotypes was examined.ResultsThe A allele was associated with asthma (OR = 1.56; Mantel–Haenszel p = 0.0040) when the analysis was limited to patients with early-onset adult asthma. In addition, when the analysis was limited to atopic individuals, a stronger association of the CDHR3 variant with early-onset asthma was found, and interaction of the CDHR3 genotype with atopy was demonstrated. Finally, a significant association of this variant was specifically found with a phenotype of asthma characterized by atopy, early-onset, and lower lung function.ConclusionsOur study supports the concept that the CDHR3 variant is an important susceptibility factor for severe adult asthma in individuals who develop the disease in early life. The interaction between the CDHR3 variant and atopy indicates that genetic predisposition to early respiratory viral infection is combined with atopy in promoting asthma

    Adult-onset hereditary pulmonary alveolar proteinosis caused by a single-base deletion in CSF2RB.

    Background Disruption of granulocyte/macrophage colony-stimulating factor (GM-CSF) signalling causes pulmonary alveolar proteinosis (PAP). Rarely, genetic defects in neonatal or infant-onset PAP have been identified in CSF2RA. However, no report has clearly identified any function-associated genetic defect in CSF2RB. Methods and results The patient was diagnosed with PAP at the age of 36 and developed respiratory failure. She was negative for GM-CSF autoantibody and had no underlying disease. Signalling and genetic defects in GM-CSF receptor were screened. GM-CSF-stimulated STAT5 phosphorylation was not observed and GM-CSF-Rβc expression was defective in the patient\u27s blood cells. Genetic screening revealed a homozygous, single-base deletion at nt 631 in exon 6 of CSF2RB on chromosome 22, which caused reductions in GM-CSF dependent signalling and function. Both parents, who were second cousins, showed no pulmonary symptoms, and had normal GM-CSF-signalling, but had a CSF2RB allele with the identical deletion, indicating that the mutant allele may give rise to PAP in an autosomal recessive manner. Conclusions This is the first report identifying a genetic defect in CSF2RB that causes deficiency of GM-CSF-Rβc expression and impaired signalling downstream. These results suggested that GM-CSF signalling was compensated by other signalling pathways, leading to adult-onset PAP
