    Mammary tissue microenvironment determines T cell-dependent breast cancer-associated inflammation

    Although the importance of the host tissue microenvironment in cancer progression and metastasis has been established, the spatiotemporal process establishing a cancer metastasis-prone tissue microenvironment remains unknown. In this study, we aim to understand the immunological character of a metastasis-prone microenvironment in a murine 4T1 breast tumor model, by using the activation of nuclear factor-jb (NF-jB) in cancer cells as a sensor of inflammatory status and by monitoring its activity by bioluminescence imaging. By using a 4T1 breast cancer cell line stably expressing an NF-jB ⁄ Luc2 reporter gene (4T1 NF-jB cells), we observed significantly increased bioluminescence approximately 7 days after metastasis-prone orthotopic mammary fat-pad inoculation but not ectopic s.c. inoculation of 4T1 NF-jB cells. Such in vivo NF-jB activation within the fat-pad 4T1 tumor was diminished in immune-deficient SCID or nude mice, or T celldepleted mice, suggesting the requirement of host T cell-mediated immune responses. Given the fat-pad 4T1 tumor expressed higher inflammatory mediators in a T cell-dependent mechanism compared to the s.c. tumor, our results imply the importance of the surrounding tissue microenvironment for inflaming tumors by collaborating with T cells to instigate metastatic spread of 4T1 breast cancer cells

    Prevalence and safety of robotic surgery for gastrointestinal malignant tumors in Japan

    [Aim] The National Health Insurance system has reimbursed robotic gastrointestinal surgery since April 2018 in Japan. Additionally, strict facility and surgeon standards were established by the government and the academic society. This study aimed to evaluate the prevalence and safety of robotic surgery using a Japanese nationwide web-based database. [Methods] Patients who underwent the following robotic surgeries for malignant tumors in 2018 were included: esophagectomy (RE), total gastrectomy (RTG), distal gastrectomy (RDG), proximal gastrectomy (RPG), low anterior resection (RLAR), and rectal resections other than RLAR (RRR). The number of cases and surgical mortality rates each month were calculated to evaluate the prevalence and safety of robotic procedures. [Results] A total of 3281 patients underwent robotic gastrointestinal surgery. The monthly number of robotic surgeries nearly doubled in April 2018 when they were initially reimbursed by the National Health Insurance system. Operative mortality rates were 0.9%, 0.4%, 0.2%, and 2.8% for RE (n = 330), RTG (n = 239), RDG (n = 1167), and RPG (n = 109), respectively. No mortality was observed in RLAR (n = 1062) or RRR (n = 374). [Conclusion] Robotic surgery for gastrointestinal malignant tumors was safely introduced into daily clinical practice along with rigorous surgeon and facility standards in Japan

    Importance of absorbable surgical sutures for the prevention of stitch abscess after surgery in patients with oral squamous cell carcinoma

    To elucidate the significance of absorbable surgical sutures in the occurrence of stitch abscess after surgery in patients with oral squamous cell carcinoma (SCC). The subjects were 251 patients who underwent excision and/or reconstruction and/or neck dissection for oral SCC using absorbable surgical sutures. Detection rates and characteristics of patients with stitch abscess were retrospectively evaluated by comparing between our present and previous data. There was only one stitch abscess among the 251 patients. A significant difference in the incidence of stitch abscess was found between the present data and our previous data. Of course, no significant correlations were found between the occurrence of stitch abscess using absorbable surgical sutures and the various factors seen in our previous analysis. A complete switch of surgical sutures from silk to absorbable surgical sutures is needed for surgery in patients with oral SCC

    Power System Development of the AGU Remote Innovative CubeSat Alert System -2 – ARICA-2

    We present the power system development of the 2U CubeSat, AGU Remote Innovative CubeSat Alert system -2(ARICA-2). The main goal of the ARICA-2 project is to demonstrate the real-time alert system of transient astronomical sources using commercial satellite network devices. 1U CubeSat ARICA was launched in November 2021. However, we have not been able to send and receive the data at this point. Therefore, we started developing 2U CubeSat ARICA-2, which is an improved version of ARICA, in April 2022. One of the possible causes of the communication problem of ARICA is the power system, such as a negative power budget or a failure in the installation of the inhibit switches. ARICA-2 is upsized from 1U to 2U to ensure a sufficient power generation and is equipped with improved inhibit switches. The calculation of power consumption and simulation of power supply on orbit have been finished. We confirmed the performance of our Electric Power System (EPS) and the health of the installed batteries. We are currently in the EM development phase with the goal of launching in Japanese fiscal year 2024

    Development of the ARICA-2 Satellite Using Spresense as an Onboard Computer

    Gamma-ray bursts (GRBs) are transient astronomical phenomena that emit enormous amounts of energy in electromagnetic waves, mainly in the gamma-ray range, for several seconds to tens of seconds. GRB observations are challenging because of the difficulty in predicting the location and time of occurrence and its extremely short duration. Therefore, it is necessary to notify about the discovery in space and to conduct follow-up observations by researchers. The AGU Remote Innovative CubeSat Alert system-2 (ARICA-2) has been developed to demonstrate a new alert system using commercial satellite network services. ARICA-2 uses SONY’s Spresense as its onboard computer (OBC). We manufactured the special board to attach two Spresenses as a redundancy of the OBC system.     Overview and Status of AGU Remote Innovative Cubesat Alert System-2 on 2023

    We present the overview of the 2U CubeSat, AGU Remote Innovative Cubesat Alert system - 2 (ARICA-2). ARICA-2 was selected as a feasibility study phase of the JAXA-Small Satellite Rush Program (JAXA-SMASH) and the JAXA Innovative Satellite Technology Demonstration-4 project in 2022. The main goal of ARICA-2 is to demonstrate the real-time alert system of transient astronomical sources, such as gamma-ray bursts, using commercial satellite network services. The first 1U CubeSat ARICA, which had the same mission goal as ARICA-2, was successfully launched in 2021 by the JAXA’s Epsilon rocket No.5. However, communication with ARICA has yet to be established due to severe hardware issues. Therefore, ARICA-2 is the re-challenging mission of ARICA. ARICA-2 has several different features compared to ARICA. First, a transceiver using amateur radio frequency is added to the commercial satellite network devices to communicate directly from the ground. Second, ARICA-2 uses Sony’s low-power board Spresense as an onboard computer. Third, the attitude control system using magnetorquer is installed to establish better communication with the commercial network satellites. Fourth, the size of a gamma-ray detector is 70 mm x 70 mm x 10 mm, which is larger by a factor of 200 in volume compared to ARICA, to enhance the detection rate of gamma-ray bursts. We plan to develop the engineering model (EM) in 2023 and perform thermal vacuum and vibration tests on the EM. We report the current status and a prospect of ARICA-2

    Advanced Clinical Usefulness of Ultrasonography for Diseases in Oral and Maxillofacial Regions

    Various kinds of diseases may be found in the oral and maxillofacial regions and various modalities may be applied for their diagnosis, including intra-oral radiography, panoramic radiography, ultrasonography, computed tomography, magnetic resonance imaging, and nuclear medicine methods such as positron emission tomography. Of these modalities, ultrasound imaging is easy to use for the detection of noninvasive and soft tissue-related diseases. Doppler ultrasound images taken in the B-mode can provide vascular information associated with the morphology of soft tissues. Thus, ultrasound imaging plays an important role in confirming the diagnosis of many kinds of diseases in such oral and maxillofacial regions as the tongue, lymph nodes, salivary glands, and masticatory muscles. In the present article, we introduce three new applications of ultrasonography: guided fine-needle aspiration, measurement of tongue cancer thickness, and diagnosis of metastasis to cervical lymph nodes
