Poly[methanoltetrakis[μ3-5-(4-methoxyphenyl)pyrazole-3-carboxylato(2−)]tricopper(II)disodium(I)
The asymmetric unit of the polymeric title complex, [Cu3Na2(C11H8N2O3)4(CH4O)]n, consists of two NaI atoms, three CuII atoms, four 5-(4-methoxyphenyl)pyrazole-3-carboxylate ligands and one methanol molecule. The central CuII atom is coordinated by four N atoms from four pyrazole-3-carboxylate ligands, forming a distorted tetrahedral (CuN4) geometry, while each of the other two CuII atoms is coordinated by two O atoms and two N atoms from the two ligands, forming a slightly distorted square-planar (CuN2O2) geometry. Each of the two NaI atoms is coordinated by five O atoms, forming a distorted octahedral geometry; four O atoms are from the carboxylate groups of the three ligands and the remaining O atom is from the methoxy group of the ligand or from the methanol molecule
Sulforaphane induces adipocyte browning and promotes glucose and lipid utilization
Scope: Obesity is closely related to the imbalance of white adipose tissue storing excess calories, and brown adipose tissue dissipating energy to produce heat in mammals. Recent studies revealed that acquisition of brown characteristics by white adipocytes, termed “browning,” may positively contribute to cellular bioenergetics and metabolism homeostasis. The goal was to investigate the putative effects of natural antioxidant sulforaphane (1-isothiocyanate-4-methyl-sulfonyl butane; SFN) on browning of white adipocytes. Methods and Results: 3T3-L1 mature white adipocytes were treated with SFN for 48 h, and then the mitochondrial content, function, and energy utilization were assessed. SFN was found to induce 3T3-L1 adipocytes browning based on the increased mitochondrial content and activity of respiratory chain enzymes, whereas the mechanism involved the upregulation of nuclear factor E2-related factor 2/ sirtuin1/ peroxisome proliferator-activated receptor gamma coactivator 1 alpha signaling. SFN enhanced uncoupling protein 1 expression, a marker for brown adipocyte, leading to the decrease in cellular ATP. SFN also enhanced glucose uptake and oxidative utilization, lipolysis and fatty acid oxidation in 3T3-L1 adipocytes. Conclusion: SFN-induced browning of white adipocytes enhanced the utilization of cellular fuel, and the application of SFN is a promising strategy to combat obesity and obesity-related metabolic disorder
Higher-order Topological and Nodal Superconductors MS (M = Nb and Ta) Transition-metal Sulfides
Intrinsic topological superconducting materials are exotic and vital to
develop the next-generation topological superconducting devices, topological
quantum calculations, and quantum information technologies. Here, we predict
the topological and nodal superconductivity of MS (M = Nb and Ta)
transition-metal sulfides by using the density functional theory for
superconductors combining with the symmetry indicators. We reveal their
higher-order topology nature with an index of Z4 = 2. These materials have a
higher Tc than the Nb or Ta metal superconductors due to their flat-band and
strong electron-phonon coupling nature. Electron doping and lighter isotopes
can effectively enhance the Tc. Our findings show that the MS (M = Nb and Ta)
systems can be new platforms to study exotic physics in the higher-order
topological superconductors, and provide a theoretical support to utilize them
as the topological superconducting devices in the field of advanced topological
quantum calculations and information technologies.Comment: 5 pages, 3 figure
Hydroxysafflor Yellow A protects spinal cords from ischemia/reperfusion injury in rabbits

Abstract

Background

Hydroxysafflor Yellow A (HSYA), which is one of the most important active ingredients of the Chinese herb <it>Carthamus tinctorius L</it>, is widely used in the treatment of cerebrovascular and cardiovascular diseases. However, the potential protective effect of HSYA in spinal cord ischemia/reperfusion (I/R) injury is still unknown.

Methods

Thirty-nine rabbits were randomly divided into three groups: sham group, I/R group and HSYA group. All animals were sacrificed after neurological evaluation with modified Tarlov criteria at the 48th hour after reperfusion, and the spinal cord segments (L4-6) were harvested for histopathological examination, biochemical analysis and terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL) staining.

Results

Neurological outcomes in HSYA group were slightly improved compared with those in I/R group. Histopathological analysis revealed that HSYA treatment attenuated I/R induced necrosis in spinal cords. Similarly, alleviated oxidative stress was indicated by decreased malondialdehyde (MDA) level and increased superoxide dismutase (SOD) activity after HSYA treatment. Moreover, as seen from TUNEL results, HSYA also protected neurons from I/R-induced apoptosis in rabbits.

Conclusions

These findings suggest that HSYA may protect spinal cords from I/R injury by alleviating oxidative stress and reducing neuronal apoptosis in rabbits.</p

A genomic copy number variant analysis implicates the MBD5 and HNRNPU genes in Chinese children with infantile spasms and expands the clinical spectrum of 2q23.1 deletion
Background: Infantile spasms (IS) is a specific type of epileptic encephalopathy associated with severe developmental disabilities. Genetic factors are strongly implicated in IS, however, the exact genetic defects remain unknown in the majority of cases. Rare mutations in a single gene or in copy number variants (CNVs) have been implicated in IS of children in Western countries. The objective of this study was to dissect the role of copy number variations in Chinese children with infantile spasms. Methods: We used the Agilent Human Genome CGH microarray 180 K for genome-wide detection of CNVs. Real-time qPCR was used to validate the CNVs. We performed genomic and medical annotations for individual CNVs to determine the pathogenicity of CNVs related to IS. Results: We report herein the first genome-wide CNV analysis in children with IS, detecting a total of 14 CNVs in a cohort of 47 Chinese children with IS. Four CNVs (4/47 = 8.5%) (1q21.1 gain; 1q44, 2q31.1, and 17p13 loss) are considered to be pathogenic. The CNV loss at 17p13.3 contains PAFAH1B1 (LIS1), a causative gene for lissencephaly. Although the CNVs at 1q21.1, 1q44, and 2q23.1 have been previously implicated in a wide spectrum of clinical features including autism spectrum disorders (ASD) and generalized seizure, our study is the first report identifying them in individuals with a primary diagnosis of IS. The CNV loss in the 1q44 region contains HNRNPU, a strong candidate gene recently suggested in IS by the whole exome sequencing of children with IS. The CNV loss at 2q23.1 includes MBD5, a methyl-DNA binding protein that is a causative gene of ASD and a candidate gene for epileptic encephalopathy. We also report a distinct clinical presentation of IS, microcephaly, intellectual disability, and absent hallux in a case with the 2q23.1 deletion. Conclusion: Our findings strongly support the role of CNVs in infantile spasms and expand the clinical spectrum associate with 2q23.1 deletion. In particular, our study implicates the HNRNPU and MBD5 genes in Chinese children with IS. Our study also supports that the molecular mechanisms of infantile spasms appear conserved among different ethnic backgrounds
Experimental study of THGEM detector with mini-rim
The gas gain and energy resolution of single and double THGEM detectors
(5{\times}5cm2 effective area) with mini-rims (rim is less than 10{\mu}m) were
studied. The maximum gain can reach 5{\times}103 and 2{\times}105 for single
and double THGEM respectively, while the energy resolution of 5.9 keV X-ray
varied from 18% to 28% for both single and double THGEM detectors of different
hole sizes and thicknesses. All the experiments were investigated in mixture of
noble gases(argon, neon) and small content of other gases(iso-butane,methane) at
atmospheric pressure.Comment: 4pages,6figures, it has been submitted to Chinese Physics
Observation of Majorana fermions with spin selective Andreev reflection in the vortex of topological superconductor
Majorana fermion (MF) whose antiparticle is itself has been predicted in
condensed matter systems. Signatures of the MFs have been reported as zero
energy modes in various systems. More definitive evidences are highly desired
to verify the existence of the MF. Very recently, theory has predicted MFs to
induce spin selective Andreev reflection (SSAR), a novel magnetic property
which can be used to detect the MFs. Here we report the first observation of
the SSAR from MFs inside vortices in Bi2Te3/NbSe2 hetero-structure, in which
topological superconductivity was previously established. By using
spin-polarized scanning tunneling microscopy/spectroscopy (STM/STS), we show
that the zero-bias peak of the tunneling differential conductance at the vortex
center is substantially higher when the tip polarization and the external
magnetic field are parallel than anti-parallel to each other. Such strong spin
dependence of the tunneling is absent away from the vortex center, or in a
conventional superconductor. The observed spin dependent tunneling effect is a
direct evidence for the SSAR from MFs, fully consistent with theoretical
analyses. Our work provides definitive evidences of MFs and will stimulate the
MFs research on their novel physical properties, hence a step towards their
statistics and application in quantum computing.Comment: 4 figures 15 page
Metabolomic and transcriptomic profiling reveals the effect of dietary protein and lipid levels on growth performance in loach (Paramisgurnus dabryanus)
The subject of this study was to explore the optimum requirements of loach (Paramisgurnus dabryanus) regarding dietary proteins and lipids and discuss the underlying mechanism. We designed nine diets to determine the effects of different levels of dietary crude protein (CP: 30%, 35%, and 40%) and ether extract (EE: 6%, 10%, and 14%) on the growth performance and metabolism of P. dabryanus. In total, 2160 healthy P. dabryanus (5.19 ± 0.01 g) were divided into nine groups with four replications at 60 fish per barrel stocking density. The trial lasted for eight weeks. Serum and liver samples were gathered for metabolomic and transcriptomic analyses. The results showed that the specific growth rate of P. dabryanus in the CP40EE10 group was the fastest and notably higher than that in other groups (P< 0.05). Analysis of the metabolome results found that the mTOR signaling pathway, glycerophospholipid metabolism, D-arginine and D-ornithine metabolism were significantly enriched pathways in the CP40EE10 group compared with the other groups (P< 0.05). Moreover, the transcriptomic analysis of differentially expressed genes (DEGs) showed that the expression of ARG (arginase) involved in protein synthesis was significantly upregulated in the CP40EE10 group compared to the slowest growing group (P< 0.05). Additionally, the expression of SPLA2 (secretory phospholipase A2) involved in lipid metabolism and FBP (fructose-1,6-bisphosphatase) involved in glucose metabolism were all significantly downregulated in the CP30EE6 group compared with the CP40EE10 group (P< 0.05). Furthermore, the analysis of differentially expressed metabolites (DEMs) and DEGs co-enriched in the KEGG pathway revealed that the significantly enriched pathways were arginine and proline metabolism, glycerophospholipid metabolism, and glycolysis/gluconeogenesis in CP40EE10 compared with other groups (P< 0.05). We conclude that including 40% CP and 10% EE in the P. dabryanus diet could result in a better growth rate. We hypothesized from metabolomic and transcriptomic analyses that the CP40EE10 diet might promote the growth of P. dabryanus by promoting protein synthesis, lipid metabolism, and energy production
