    In order to depict a mechanism of preschool teachers’ ways of learning at their on-site training session, this study consists of two research projects: project 1 is to clarify preschool teachers and directors overall learning experiences through on-site training session, and project 2 is to examine ways of preschool teachers’ and directors’ learning process at their actual on-site training session. As results, the feel-secured atmosphere of on-site training session facilitates preschool teachers learning process and making practice visible and being aware of its importance through exchanging their minds and thoughts with each other are considered as what they learnt at the session. In order to give on-site training sessions a structure, it is implied that examining its phase, each teachers learning process, and their interweaving mindsets emerged between within inner-self and interaction with others is crucial

    Generation of Tetrafluoroethylene–Propylene Elastomer-Based Microfluidic Devices for Drug Toxicity and Metabolism Studies

Using these devices (PDMS-based devices), the mechanical microenvironment of living tissues, such as pulmonary respiration and intestinal peristalsis, can be reproduced in vitro. However, the use of PDMS-based devices in drug discovery research is limited because of their extensive absorption of drugs. In this study, we investigated the feasibility of the tetrafluoroethylene–propylene (FEPM) elastomer to fabricate a hepatocyte-on-a-chip (FEPM-based hepatocyte chip) with lower drug absorption. The FEPM-based hepatocyte chip expressed drug-metabolizing enzymes, drug-conjugating enzymes, and drug transporters. Also, it could produce human albumin. Although the metabolites of midazolam and bufuralol were hardly detected in the PDMS-based hepatocyte chip, they were detected abundantly in the FEPM-based hepatocyte chip. Finally, coumarin-induced hepatocyte cytotoxicity was less severe in the PDMS-based hepatocyte chip than in the FEPM-based hepatocyte chip, reflecting the different drug absorptions of the two chips. In conclusion, the FEPM-based hepatocyte chip could be a useful tool in drug discovery research, including drug metabolism and toxicity studies

    Effect of edible oils on quercetin, kaempferol and galangin transport and conjugation in the intestinal Caco-2/HT29-MTX co-culture model

    Solubility and matrix play an important role in the gut lumen in delivering bioactive compounds to the absorptive surface of enterocytes. The purpose of this study was to determine the effect of certain commonly consumed lipids, soybean, olive and corn oil, on the transport and conjugation of flavonols (myricetin, quercetin, kaempferol and galangin) using the conjugation-competent co-cultured Caco-2/HT29-MTX intestinal cell monolayer model. To enable identification and quantification of conjugates, each flavonol was enzymatically glucuronidated or sulphated, then analysed by HPLC with triple quadrupole mass spectrometric detection. Quantification showed large differences in mass spectrometric peak area response factors between the aglycones and many of the conjugates, with galangin-sulphate for example ionising ∼15-fold better than galangin. Flavonol aglycones and conjugates were transported to the basolateral side of Caco-2/HT29-MTX co-cultures. The total amount of methyl, sulphate and glucuronide conjugates was in the order: galangin > quercetin > kaempferol > myricetin. All oils inhibited the transport and conjugation of galangin, the most hydrophobic flavonol, whereas they increased the sulphation, and to some extent glucuronidation, of quercetin and kaempferol. The results show that the lipid matrix has the potential to modify both transport and conjugation of dietary flavonols, but that the effect depends upon the structure and hydrophobicity

    Thermodynamic Assessment of the Cu-Pt System

    A CALPHAD assessment of the Cu-Pt system has been carried out. Two and four sublattice models were applied to describe the Gibbs free energies of ordered phases where the contribution of SRO is taken explicitly into account through the reciprocal parameters. The disordered fcc A1 and liquid phases were treated as substitutional solutions. A consistent set of parameters for the phases in the Cu-Pt system as obtained, and those parameters can satisfactorily reproduce the experimental phase equilibria and thermodynamic properties, such as enthalpies, activity of Cu, and long-range order parameters
