KxFe2-ySe2 single crystals: Floating-zone growth, Transport and Structural properties

Single crystals of superconducting KxFe2-ySe2 have been grown with the optical floating-zone technique under application of 8 bar of argon pressure. We found that large and high quality single crystals with dimensions of ~\varnothing6 \times 10 mm could be obtained at the termination of the grown ingot through quenching, while the remaining part of the ingot decomposed. As-grown single crystals commonly represent an intergrowth of two sets of the c-axis characterized by slightly different lattice constants. Single crystal of K0.80Fe1.81Se2 shows a superconducting transition at Tc = 31.6 K, leading to a near 100% expulsion of the external magnetic field in magnetization measurements. On the other hand, neutron-diffraction data indicate that superconductivity in the sample coexists with a iron-vacancy superstructure and static antiferromagnetic order. The anisotropic ratio of the upper critical field Hc2 for both H//c and H//ab configurations is \sim3.46

Enhancer Reprogramming Confers Dependence on Glycolysis and IGF Signaling in KMT2D Mutant Melanoma.

Histone methyltransferase KMT2D harbors frequent loss-of-function somatic point mutations in several tumor types, including melanoma. Here, we identify KMT2D as a potent tumor suppressor in melanoma through an in vivo epigenome-focused pooled RNAi screen and confirm the finding by using a genetically engineered mouse model (GEMM) based on conditional and melanocyte-specific deletion of KMT2D. KMT2D-deficient tumors show substantial reprogramming of key metabolic pathways, including glycolysis. KMT2D deficiency aberrantly upregulates glycolysis enzymes, intermediate metabolites, and glucose consumption rates. Mechanistically, KMT2D loss causes genome-wide reduction of H3K4me1-marked active enhancer chromatin states. Enhancer loss and subsequent repression of IGFBP5 activates IGF1R-AKT to increase glycolysis in KMT2D-deficient cells. Pharmacological inhibition of glycolysis and insulin growth factor (IGF) signaling reduce proliferation and tumorigenesis preferentially in KMT2D-deficient cells. We conclude that KMT2D loss promotes tumorigenesis by facilitating an increased use of the glycolysis pathway for enhanced biomass needs via enhancer reprogramming, thus presenting an opportunity for therapeutic intervention through glycolysis or IGF pathway inhibitors

Metamagnetic transition in EuFe2_2As2_2 single crystals

We report the measurements of anisotropic magnetization and magnetoresistance on single crystals of EuFe2_2As2_2, a parent compound of ferro-arsenide high-temperature superconductor. Apart from the antiferromagnetic (AFM) spin-density-wave transition at 186 K associated with Fe moments, the compound undergoes another magnetic phase transition at 19 K due to AFM ordering of Eu2+^{2+} spins (J=S=7/2J=S=7/2). The latter AFM state exhibits metamagnetic transition under magnetic fields. Upon applying magnetic field with H∥cH\parallel c at 2 K, the magnetization increases linearly to 7.0 μB\mu_{B}/f.u. at μ0H\mu_{0}H=1.7 T, then keeps at this value of saturated Eu2+^{2+} moments under higher fields. In the case of H∥abH\parallel ab, the magnetization increases step-like to 6.6 μB\mu_{B}/f.u. with small magnetic hysteresis. A metamagnetic phase was identified with the saturated moments of 4.4 μB\mu_{B}/f.u. The metamagnetic transition accompanies with negative in-plane magnetoresistance, reflecting the influence of Eu2+^{2+} moments ordering on the electrical conduction of FeAs layers. The results were explained in terms of spin-reorientation and spin-reversal based on an AA-type AFM structure for Eu2+^{2+} spins. The magnetic phase diagram has been established.Comment: 10 pages, 8 figures. accepted for publication in New Journal of Physics as a special issue articl

BACKGROUND: A recent genome-wide scan has identified two genetic variants in the HLA-DP region strongly associated with hepatitis B infection in Japanese. This study evaluates the effects of these risk variants in Chinese, where the HBV infection is the most popular in the world. METHODS AND FINDINGS: We have assessed the relationship between these two single nucleotide polymorphisms (rs3077 and rs9277535) and chronic hepatitis B infection in two independent case-control studies. The first population in Chinese Han included 736 patients and 782 spontaneously recovered controls. The second set was established in Chinese Zhuang minority of 177 patients and 208 controls. Both A alleles of rs3077 and rs9277535 significantly deceased the risk to CHB in Chinese Han (OR = 0.540, 95%CI: 0.464-0.628, P = 4.068×10(-16) and OR = 0.696, 95%CI: 0.601-0.806, P = 1.062×10(-6), respectively). Conceivably, rs9277535 was found to be associated with decreased risk of the disease in Chinese Zhuang, with an OR of 0.606 (95%CI, 0.441-0.833, P = 0.002). CONCLUSION: Chronic hepatitis B susceptibility loci in HLA-DP region (rs3077 and rs9277535) identified by genome-wide scan in Japanese population were validated in Chinese population. These findings might provide clues to develop screening and surveillance strategies

Using a data sample of 4.481×1084.481\times 10^8 ψ(3686)\psi(3686) events collected with the BESIII detector, we report the first observation of the four-lepton-decays J/ψ→e+e−e+e−J/\psi\to e^+e^-e^+e^- and J/ψ→e+e−μ+μ−J/\psi\to e^+e^-\mu^+\mu^- utilizing the process ψ(3686)→π+π−J/ψ\psi(3686)\to \pi^+\pi^- J/\psi. The branching fractions are determined to be [4.32±0.26 (stat)±0.19 (syst)]×10−5[4.32\pm0.26~(\rm stat)\pm0.19~(\rm syst)]\times 10^{-5} and [2.45 ±0.21 (stat)±0.10 (syst)]×10−5[2.45~\pm0.21~(\rm stat)\pm0.10~(\rm syst)]\times 10^{-5}, respectively. The results deviate from theoretical predictions, by 2.8 and 5.2 σ\sigma, respectively. No significant signal is observed for J/ψ→μ+μ−μ+μ−J/\psi\to \mu^+\mu^-\mu^+\mu^-, and an upper limit on the branching fraction is set at 1.6×10−61.6\times 10^{-6} at the 90%\% confidence level. A CP asymmetry observable is constructed for the first two channels, which is measured to be (−0.019±0.067±0.025)(-0.019\pm0.067\pm0.025) and (−0.016±0.081±0.003)(-0.016\pm0.081\pm0.003), respectively. No evidence for CP violation is observed in this process.Comment: 9 pages, 1 figur

BACKGROUND: The programmed cell death-1 receptor/programmed cell death-1 ligand (PD-1/PD-L1) pathway plays a crucial role in tumor evasion from host immunity. This study was designed to evaluate the association between circulating PD-L1/PD-1 and prognosis after cryoablation in patients with HBV-related hepatocellular carcinoma (HCC). METHODOLOGY/PRINCIPAL FINDINGS: In the present study, 141 HBV-related HCC patients were enrolled and of those 109 patients received cryoablation. Circulating PD-L1/PD-1 expression was tested by flow cytometry, and 23 patients were simultaneously evaluated for intratumoral PD-L1 expression by immunohistochemical staining. Circulating PD-1/PD-L1 expression was associated with severity of diseases in patients with HCC, and the circulating PD-L1 expression was closely correlated with intratumoral PD-L1 expression. Of the clinical parameters, PD-1/PD-L1 expression was associated with tumor size, blood vessel invasion and BCLC staging. Moreover, PD-1/PD-L1 expression dropped after cryoablation while being elevated at the time of tumor recurrence. Patients with higher expression of circulating PD-L1, as well as circulating PD-1, had a significantly shorter overall survival and tumor-free survival than those with lower expression. Multivariate analysis confirmed that circulating PD-L1 could serve as an independent predictor of overall survival and tumor-recurrence survival in HCC patients after cryoablation. CONCLUSIONS/SIGNIFICANCE: Upregulation of circulating PD-L1/PD-1 is associated with poor post-cryoablation prognosis in patients with HBV-related hepatocellular carcinoma
