    SES Health Gradients during the Epidemiological Transition: The Case of China

    The epidemiological transition, which has already passed the developed world, is still progressing in many developing countries. A particular problem associated with this transition is the under-diagnosis and lack of treatment of chronic diseases, and these may exhibit SES gradients and exacerbate social inequality. Using hypertension as an example and data from China (CHNS), we find that the prevalence of hypertension in China is already close to levels in developed countries, under-diagnosis is pervasive, treatment is rare, and failure to control is widespread. Consistent with the literature, we find no income and education gradients in the prevalence of hypertension. However, there are strong education gradients in diagnosis and treatment in urban areas. The income gradients in all aspects of hypertension are relatively weak and sometimes nonexistent. Interestingly, we find that access to health care does not contribute to the diagnosis of hypertension, nor does it aid much in the treatment and control of hypertension. Our results suggest that the epidemiological transition has indeed occurred, but both the Chinese public and its health care system are ill-prepared. There is an urgent need to educate the public on chronic illnesses, and to raise the quality of health care so that patients receive proper diagnoses and guidance on how to treat and control those chronic illnesses.epidemiological transition, under-diagnosis, SES health gradients

    Engineering of biofuel crops for improving alternative energy production [abstract]

Teaming up with three other laboratories (Drs. Gary Stacey, Xu Dong, and Monty Kerley) at MU and several other institutions in Missouri, we are developing an engineering approach to improve biofuel production as an alternative source of energy. A two-year funded project with a total of $355,635 has been awarded to our MU team. One of the most important crops in this project is switchgrass (Panicum virgatum). To be successful in this project employing engineering approach, it is essential to develop an efficient Agrobacterium-mediated transformation process in switchgrass. In spite of previous reports, Agrobacterium-mediated transformation of this crop has been proven to be very difficult. Therefore, since our project started we have optimized a number of critical conditions affecting switchgrass transformation. These conditions included the switchgrass genotypes, cocultivation temperatures and medium salt concentrations, Agrobacterium strains, transformation vectors, selection system and selective agents. We also plan to examine the impact of types of promoter driving selectable markers on transformation. These works will lay a good foundation for efficient transformation of switchgrass via Agrobacterium. Some of the significant progresses will be presented

    Performance and Reliability Evaluation for DSRC Vehicular Safety Communication

    <p>Inter-Vehicle Communication (IVC) is a vital part of Intelligent Transportation System (ITS), which has been extensively researched in recent years. Dedicated Short Range Communication (DSRC) is being seriously considered by automotive industry and government agencies as a promising wireless technology for enhancing transportation safety and efficiency of road utilization. In the DSRC based vehicular ad hoc networks (VANETs), the transportation safety is one of the most crucial features that needs to be addressed. Safety applications usually demand direct vehicle-to-vehicle ad hoc communication due to a highly dynamic network topology and strict delay requirements. Such direct safety communication will involve a broadcast service because safety information can be beneficial to all vehicles around a sender. Broadcasting safety messages is one of the fundamental services in DSRC. In order to provide satisfactory quality of services (QoS) for various safety applications, safety messages need to be delivered both timely and reliably. To support the stringent delay and reliability requirements of broadcasting safety messages, researchers have been seeking to test proposed DSRC protocols and suggesting improvements. A major hurdle in the development of VANET for safety-critical services is the lack of methods that enable one to determine the effectiveness of VANET design mechanism for predictable QoS and allow one to evaluate the tradeoff between network parameters. Computer simulations are extensively used for this purpose. A few analytic models and experiments have been developed to study the performance and reliability of IEEE 802.11p for safety-related applications. In this thesis, we propose to develop detailed analytic models to capture various safety message dissemination features such as channel contention, backoff behavior, concurrent transmissions, hidden terminal problems, channel fading with path loss, multi-channel operations, multi-hop dissemination in 1-Dimentional or 2-Dimentional traffic scenarios. MAC-level and application-level performance metrics are derived to evaluate the performance and reliability of message broadcasting, which provide insights on network parameter settings. Extensive simulations in either Matlab or NS2 are conducted to validate the accuracy of our proposed models.</p>Dissertatio

A pebble p(i) that is initially placed at the vertex i has destination pi(i) for each i is an element of [n]. At each step, we choose a matching and swap the two pebbles on each of the edges. Let rt(G, pi), the routing number for pi, be the minimum number of steps necessary for the pebbles to reach their destinations. Li, Lu and Yang proved that rt(C-n, pi) = 5, if rt(C-n, pi) = n-1, then pi = 23 ... n1 or its inverse. By a computer search, they showed that the conjecture holds for n \u3c 8. We prove in this paper that the conjecture holds for all even n \u3e= 6

    Oscillatory Ca2+ Signaling in the Isolated Caenorhabditis elegans Intestine: Role of the Inositol-1,4,5-trisphosphate Receptor and Phospholipases C β and γ

    Defecation in the nematode Caenorhabditis elegans is a readily observable ultradian behavioral rhythm that occurs once every 45–50 s and is mediated in part by posterior body wall muscle contraction (pBoc). pBoc is not regulated by neural input but instead is likely controlled by rhythmic Ca2+ oscillations in the intestinal epithelium. We developed an isolated nematode intestine preparation that allows combined physiological, genetic, and molecular characterization of oscillatory Ca2+ signaling. Isolated intestines loaded with fluo-4 AM exhibit spontaneous rhythmic Ca2+ oscillations with a period of ∼50 s. Oscillations were only detected in the apical cell pole of the intestinal epithelium and occur as a posterior-to-anterior moving intercellular Ca2+ wave. Loss-of-function mutations in the inositol-1,4,5-trisphosphate (IP3) receptor ITR-1 reduce pBoc and Ca2+ oscillation frequency and intercellular Ca2+ wave velocity. In contrast, gain-of-function mutations in the IP3 binding and regulatory domains of ITR-1 have no effect on pBoc or Ca2+ oscillation frequency but dramatically increase the speed of the intercellular Ca2+ wave. Systemic RNA interference (RNAi) screening of the six C. elegans phospholipase C (PLC)–encoding genes demonstrated that pBoc and Ca2+ oscillations require the combined function of PLC-γ and PLC-β homologues. Disruption of PLC-γ and PLC-β activity by mutation or RNAi induced arrhythmia in pBoc and intestinal Ca2+ oscillations. The function of the two enzymes is additive. Epistasis analysis suggests that PLC-γ functions primarily to generate IP3 that controls ITR-1 activity. In contrast, IP3 generated by PLC-β appears to play little or no direct role in ITR-1 regulation. PLC-β may function instead to control PIP2 levels and/or G protein signaling events. Our findings provide new insights into intestinal cell Ca2+ signaling mechanisms and establish C. elegans as a powerful model system for defining the gene networks and molecular mechanisms that underlie the generation and regulation of Ca2+ oscillations and intercellular Ca2+ waves in nonexcitable cells

    Validation of a guideline-based decision support system for the diagnosis of primary headache disorders based on ICHD-3 beta

    BACKGROUND: China may have the largest population of headache sufferers and therefore the most serious burden of disease worldwide. However, the rate of diagnosis for headache disorders is extremely low, possibly due to the relative complexity of headache subtypes and diagnostic criteria. The use of computerized clinical decision support systems (CDSS) seems to be a better choice to solve this problem. METHODS: We developed a headache CDSS based on ICHD-3 beta and validated it in a prospective study that included 543 headache patients from the International Headache Center at the Chinese PLA General hospital, Beijing, China. RESULTS: We found that the CDSS correctly recognized 159/160 (99.4%) of migraine without aura, 36/36 (100%) of migraine with aura, 20/21 (95.2%) of chronic migraine, and 37/59 (62.7%) of probable migraine. This system also correctly identified 157/180 (87.2%) of patients with tension-type headache (TTH), of which infrequent episodic TTH was diagnosed in 12/13 (92.3%), frequent episodic TTH was diagnosed in 99/101 (98.0%), chronic TTH in 18/20 (90.0%), and probable TTH in 28/46 (60.9%). The correct diagnostic rates of cluster headache and new daily persistent headache (NDPH) were 90.0% and 100%, respectively. In addition, the system recognized 32/32 (100%) of patients with medication overuse headache. CONCLUSIONS: With high diagnostic accuracy for most of the primary and some types of secondary headaches, this system can be expected to help general practitioners at primary hospitals improve diagnostic accuracy and thereby reduce the burden of headache in China

    Hypomethylation of IL10 and IL13 Promoters in CD4+ T Cells of Patients with Systemic Lupus Erythematosus

    Interleukin- (IL-)10 and IL-13 play important roles in Th2 cell differentiation and production of autoantibodies in patients with (SLE). However, the mechanisms leading to IL10 and IL13 overexpression in SLE patients are not well understood. In this study, we confirm that the levels of both IL10 and IL13 mRNA in CD4+ T cells and of serum IL10 and IL13 proteins are increased in SLE patients. We show that the DNA methylation levels within IL10 and IL13 gene regulatory domains are reduced in SLE CD4+ T cells relative to healthy controls and negatively correlate with IL10 and IL13 mRNA expression. Moreover, treating healthy CD4+ T cells with the demethylating agent 5-azacytidine (5-azaC) increased IL10 and IL13 mRNA transcription. Together, our results show that promoter methylation is a determinant of IL10 and IL13 expression in CD4+ T cells, and we propose that DNA hypomethylation leads to IL10 and IL13 overexpression in SLE patients
