    Fruit quality assessment based on mineral elements and juice properties in nine citrus cultivars

    IntroductionCitrus fruit is considered a superfood due to its multiple nutritional functions and health benefits. Quantitative analysis of the numerous quality characteristics of citrus fruit is required to promote its sustainable production and industrial utilization. However, little information is available on the comprehensive quality assessment of various fruit quality indicators in different citrus cultivars.MethodsA total of nine different fresh citrus fruits containing seeds were collected as the experimental materials. The objectives of this study were: (i) to determine the morphological and juice properties of citrus fruits, (ii) to measure the mineral elements in the peel, pulp, and seeds, and (iii) to evaluate the fruit quality index (FQI) using the integrated quality index (IQI) and the Nemoro quality index (NQI) methods.ResultsThere were significant differences in fruit quality characteristics, including morphological, mineral, and juice quality, among the investigated citrus cultivars. The proportion of pulp biomass was the highest, followed by that of peel and seeds. N and Cu had the highest and lowest concentrations, respectively, among the measured elements across all citrus fruits, and the amounts of N, P, Mg, Cu, and Zn in seeds, K and Al in pulp, and Ca, Fe, and Mn in peel were the highest, dramatically affecting the accumulation of minerals in the whole fruit and their distribution in various fruit parts. Additionally, Ningmeng fruits had the highest vitamin C and titratable acidity (TA) but the lowest total soluble solids (TSS) and total phenolic (TP) contents, resulting in the lowest TSS/TA and pH values. In contrast, Jinju fruits had the highest TSS and TP contents. Based on the mineral element and juice quality parameters, principal component analysis showed that the citrus fruits were well separated into four groups, and the dendrogram also showed four clusters with different distances. The FQI range based on the IQI method (FQIIQI) and NQI method (FQINQI) was 0.382-0.590 and 0.106-0.245, respectively, and a positive relationship between FQIIQI and FQINQI was observed.ConclusionOur results highlight the great differences in mineral and juice characteristics among fruit parts, which mediated fruit quality. The strategy of fruit quality assessment using the FQI can be expanded for targeted utilization in the citrus industry

    Retrospective evaluation of whole exome and genome mutation calls in 746 cancer samples

    Funder: NCI U24CA211006Abstract: The Cancer Genome Atlas (TCGA) and International Cancer Genome Consortium (ICGC) curated consensus somatic mutation calls using whole exome sequencing (WES) and whole genome sequencing (WGS), respectively. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium, which aggregated whole genome sequencing data from 2,658 cancers across 38 tumour types, we compare WES and WGS side-by-side from 746 TCGA samples, finding that ~80% of mutations overlap in covered exonic regions. We estimate that low variant allele fraction (VAF < 15%) and clonal heterogeneity contribute up to 68% of private WGS mutations and 71% of private WES mutations. We observe that ~30% of private WGS mutations trace to mutations identified by a single variant caller in WES consensus efforts. WGS captures both ~50% more variation in exonic regions and un-observed mutations in loci with variable GC-content. Together, our analysis highlights technological divergences between two reproducible somatic variant detection efforts

    Bleeding due to successive duodenal and rectal ulcers in an 81-year-old patient with severe COVID-19: a case report

    Abstract Background In the early stages of the coronavirus disease 2019 (COVID-19) outbreak, the most widely recognised symptoms of the disease were fever, cough, shortness of breath, myalgia, and fatigue. However, in addition to these symptoms, COVID-19 can cause systemic symptoms outside the lungs. Older patients with severe COVID-19 often require admission to the intensive care unit (ICU). Acute rectal ulcer bleeding, characterised by painless, profuse haematochezia, caused by solitary or multiple rectal ulcers, is one of the main causes of severe haematochezia in patients with COVID-19 in the ICU. However, recurrent duodenal ulcer bleeding followed by rectal ulcer bleeding has not previously been reported in older patients during ICU treatment for severe COVID-19. Cases presentation Herein, we report the case of an 81-year-old woman admitted to the emergency department due to severe COVID-19 and transferred to the ICU 2 days later for treatment. During treatment in the ICU, the patient developed recurrent duodenal ulcer bleeding and underwent endoscopic electrocoagulation haemostasis and gastroduodenal artery embolisation. However, the night after the final haemostatic operation, due to rectal ulcer bleeding, the patient discharged bloody stools intermittently, which was effectively controlled using endoscopic electrocoagulation, topical medication, blood transfusion, and haemostatic drugs. Conclusions To the best of our knowledge, this is the first report of duodenal ulcer bleeding followed by rectal ulcer bleeding in an older patient with severe COVID-19 infection. This report creates awareness for clinicians about the multiple and complex gastrointestinal symptoms that may occur during COVID-19 treatment

    Deep characteristics analysis on travel time of emergency traffic

    Owing to the rapid development of emergency rescue transportation in cities and the frequent emergencies, demand for emergency rescue is increasing drastically. How to select an emergency rescue route quickly and shorten the rescue travel time under the condition of limited urban road resources is of great significance. Based on the characteristics analysis of emergency rescue, this paper classifies priority levels of different emergency traffic, moreover, the travel times are also analysed with three scenarios: 1) emergency rescue vehicles encountering no queues; 2) encountered queues but lanes available; 3) encountered queues with no available lanes. Related case study shows that model in this paper can effectively shorten travel time of emergency traffic in the route and improve its efficiency.Accepted Author ManuscriptTransport and Plannin

    Supplementation of 5,6-Dimethylbenzimidazole and Cobalt in High-Concentrate Diet Improves the Ruminal Vitamin B<sub>12</sub> Synthesis and Fermentation of Sheep

    The objective of this experiment was to investigate the effects of 5,6-dimethylbenzimidazole (5,6-DMB) and cobalt (Co) on the ruminal vitamin B12 synthesis and fermentation parameters of sheep under high concentrate conditions. Twenty-four Kazakh rams (body weight = 39.23 ± 2.61 kg and 8 months old) fitted with permanent ruminal fistulas were randomly divided into four groups with six rams in each group. The control (CON) group was fed a basal ration with a concentrate-to-roughage ratio of 70:30, and the experimental groups (T60, T75 and T90) were fed a basal diet with 60 mg 5,6-DMB+ 0.25 mg Co, 75 mg 5,6-DMB+ 0.5 mg Co and 90 mg 5,6-DMB+ 0.75 mg Co supplied to each kilogram of basal diet, respectively. The experiment lasted for 26 days, with the first 14 days being an adaptation period to allow the sheep to adapt to the diet type and environment, and the second 12 days being a sample period. On 0, 7 and 12 d of the sample period, all sheep were weighed before the morning feed. Rumen fluid samples were collected from all sheep on the last 4 days of the sample period. The results showed that the ruminal vitamin B12 content was higher in trial groups than that of the CON group at 3 h after feeding (p p p p p 12, propionate, ammonia-N and microbial protein contents and pH in the rumen of sheep, and the best results were obtained by the amounts of 75 mg/kg 5,6-DMB and 0.5 mg/kg Co, respectively

    Data based violated behavior analysis of taxi driver in metropolis in China

    Violation probability of taxi drivers in metropolis is far more than that of normal drivers because they are labor-intensive, overconfident of self-driving skill, and always searching potential customers, sometimes even picking up or dropping off passengers randomly. In this paper, four types of violated behavior of taxi drivers in metropolis were first summarized, based on which corresponding scale table was initial designed with social statistical method. Furthermore, with certain samples, relative item analysis, exploratory factor analysis, validity analysis and reliability analysis were conducted to verify validity of the initial scale table, based on which some improvements were made, and we can see that the modified scale table in the paper has high fitness degree, good reliability and validity to detect violated behavior of taxi driver accurately. Finally, large area survey data of taxi driver questionnaire from Shanghai was collected with the modified scale table above, the analysis results showed that among four types of violated behavior of taxi drivers in metropolis, the probability over-speed is top to 89.57%, in which probabilities of behaviors of “driving over-speed at mid-night” and “accelerating to across the intersection during the yellow signal” are top to 64.2% and 58.2% respectively, which is meaningful for the improvement of taxi drivers’ behaviors specification and traffic safety regulation.Transport and Plannin

    Biodegradation and Mineralization of Polystyrene by Plastic-Eating Mealworms: Part 2. Role of Gut Microorganisms

    The role of gut bacteria of mealworms (the larvae of Tenebrio molitor Linnaeus) in polystyrene (PS) degradation was investigated. Gentamicin was the most effective inhibitor of gut bacteria among six antibiotics tested. Gut bacterial activities were essentially suppressed by feeding gentamicin food (30 mg/g) for 10 days. Gentamicin-feeding mealworms lost the ability to depolymerize PS and mineralize PS into CO<sub>2</sub>, as determined by characterizing worm fecula and feeding with <sup>13</sup>C-labeled PS. A PS-degrading bacterial strain was isolated from the guts of the mealworms, Exiguobacterium sp. strain YT2, which could form biofilm on PS film over a 28 day incubation period and made obvious pits and cavities (0.2–0.3 mm in width) on PS film surfaces associated with decreases in hydrophobicity and the formation of C–O polar groups. A suspension culture of strain YT2 (10<sup>8</sup> cells/mL) was able to degrade 7.4 ± 0.4% of the PS pieces (2500 mg/L) over a 60 day incubation period. The molecular weight of the residual PS pieces was lower, and the release of water-soluble daughter products was detected. The results indicated the essential role of gut bacteria in PS biodegradation and mineralization, confirmed the presence of PS-degrading gut bacteria, and demonstrated the biodegradation of PS by mealworms
