    The Genome Sequence of 'Mycobacterium massiliense' Strain CIP 108297 Suggests the Independent Taxonomic Status of the Mycobacterium abscessus Complex at the Subspecies Level

    Members of the Mycabacterium abscessus complex are rapidly growing mycobacteria that are emerging as human pathogens. The M. abscassus complex was previously composed of three species, namely M. abscessus sensu strict, 'M. massiliense', and M. bolletii', In 2011, 'M. massiliense' and 'M. bolletre' were united and reclassified as a single subspecies within M. abscessus: M. abscessus subsp. bolletii. However, the placement of 'M. massiliense' Within the boundary of M. abscessus subsp. balletii remains highly controversial with regard to clinical aspects. In this study, we revisited the taxonomic status of members of the M. abscessus complex based on comparative analysis of he whole-genome sequences of 53 strains, The genome sequence of the previous type strain of 'Mycobacterium massiliense' (CIP 108297) was determined using next-generation sequencing. The genome tree based on average nucleotide identity (AN I) values supported the differentiation of M. bolletii' and M. massiliense' at the subspecies level. The genome tree also clearly illustrated that 'M. bolletil' and 'M. massiliense' form a distinct phylogenetic clade within the radiation of the M. abscessus complex. The genomic distances observed in this study suggest that the current M. abscessus subsp. bolletii taxon should be divided into two subspecies, M. abscessus subsp. massiliense subsp. nov. and M. abscessus subsp. bolletii, to correspondingly accommodate the previously known 'M. assiliense' and 'M. bolletii' strains.

    Integration of Genetic Programming and TABU Search Mechanism for Automatic Detection of Magnetic Resonance Imaging in Cervical Spondylosis

    Cervical spondylosis is a kind of degenerative disease which not only occurs in elder patients. The age distribution of patients is unfortunately decreasing gradually. Magnetic Resonance Imaging (MRI) is the best tool to confirm the cervical spondylosis severity but it requires radiologist to spend a lot of time for image check and interpretation. In this study, we proposed a prediction model to evaluate the cervical spine condition of patients by using MRI data. Furthermore, to ensure the computing efficiency of the proposed model, we adopted a heuristic programming, genetic programming (GP), to build the core of refereeing engine by combining the TABU search (TS) with the evolutionary GP. Finally, to validate the accuracy of the proposed model, we implemented experiments and compared our prediction results with radiologistā€™s diagnosis to the same MRI image. The experiment found that using clinical indicators to optimize the TABU list in GP+TABU got better fitness than the other two methods and the accuracy rate of our proposed model can achieve 88% on average. We expected the proposed model can help radiologists reduce the interpretation effort and improve the relationship between doctors and patients

    Kinetics of cytokine expression in cirrhotic rats

    AbstractBackgroundCytokines are involved in liver injury and cirrhosis and systemic and hepatic cytokine levels may help predict cirrhosis evolution. However, the relevant survey has not been performed.MethodsMale Sprague-Dawley rats (240ā€“270g) received either common bile duct ligation (BDL, animal model of cholestatic liver injury) or sham operation (control). Five rats were sacrificed and liver and serum were collected from each in weeks 1, 2, 4, 6, 8 and 10 after surgery. Hepatic expression of interferon-Ī³ (IFN-Ī³), tumor necrosis factor-Ī± (TNF-Ī±), interleukin-10 (IL-10) and transforming growth factor-Ī² (TGF-Ī²) were analyzed by immunohistochemial staining. The corresponding serum levels were measured by ELISA.ResultsCompared to the corresponding sham groups, hepatic expression of these cytokines in BDL rats was significantly and progressively enhanced during cirrhosis development. However, serum IFN-Ī³ levels of BDL rats did not change significantly. Serum TNF-Ī± of BDL rats increased gradually and reached a peak in week 6. Serum TGF-Ī² level was elevated up to week 8, whereas IL-10 level decreased progressively until week 6.ConclusionCirrhosis development in BDL rats is associated with progressively enhanced expression of hepatic pro-inflammatory and anti-inflammatory cytokines, which is not in accord with the corresponding serum concentration. The circulating cytokine concentration may not totally reflect the hepatic expression level throughout the development of cirrhosis

    Duplex-specific nuclease efficiently removes rRNA for prokaryotic RNA-seq

    Next-generation sequencing has great potential for application in bacterial transcriptomics. However, unlike eukaryotes, bacteria have no clear mechanism to select mRNAs over rRNAs; therefore, rRNA removal is a critical step in sequencing-based transcriptomics. Duplex-specific nuclease (DSN) is an enzyme that, at high temperatures, degrades duplex DNA in preference to single-stranded DNA. DSN treatment has been successfully used to normalize the relative transcript abundance in mRNA-enriched cDNA libraries from eukaryotic organisms. In this study, we demonstrate the utility of this method to remove rRNA from prokaryotic total RNA. We evaluated the efficacy of DSN to remove rRNA by comparing it with the conventional subtractive hybridization (Hyb) method. Illumina deep sequencing was performed to obtain transcriptomes from Escherichia coli grown under four growth conditions. The results clearly showed that our DSN treatment was more efficient at removing rRNA than the Hyb method was, while preserving the original relative abundance of mRNA species in bacterial cells. Therefore, we propose that, for bacterial mRNA-seq experiments, DSN treatment should be preferred to Hyb-based methods.

    Nanofibrous insulin/vildagliptin core-shell PLGA scaffold promotes diabetic wound healing

    Introduction: Slow wound repair in diabetes is a serious adverse event that often results in loss of a limb or disability. An advanced and encouraging vehicle is wanted to enhance clinically applicable diabetic wound care. Nanofibrous insulin/vildagliptin core-shell biodegradable poly (lactic-co-glycolic acid) (PLGA) scaffolds to prolong the effective drug delivery of vildagliptin and insulin for the repair of diabetic wounds were prepared.Methods: To fabricate core-shell nanofibrous membranes, vildagliptin mixture with PLGA, and insulin solution were pumped via separate pumps into two differently sized capillary tubes that were coaxially electrospun.Results and Discussion: Nanofibrous core-shell scaffolds slowly released effective vildagliptin and insulin over 2Ā weeks in vitro migration assay and in vivo wound-healing models. Water contact angle (68.3 Ā± 8.5Ā° vs. 121.4 Ā± 2.0Ā°, p = 0.006) and peaked water absorbent capacity (376% Ā± 9% vs. 283% Ā± 24%, p = 0.003) of the insulin/vildagliptin core-shell nanofibrous membranes remarkably exceeded those of a control group. The insulin/vildagliptin-loaded core-shell nanofibers improved endothelial progenitor cells migration in vitro (762 Ā± 77Ā cells/mm2 vs. 424.4 Ā± 23Ā cells/mm2, p < 0.001), reduced the Ī±-smooth muscle actin content in vivo (0.72 Ā± 0.23 vs. 2.07 Ā± 0.37, p < 0.001), and increased diabetic would recovery (1.9 Ā± 0.3Ā mm2 vs. 8.0 Ā± 1.4Ā mm2, p = 0.002). Core-shell insulin/vildagliptin-loaded nanofibers extend the drug delivery of insulin and vildagliptin and accelerate the repair of wounds associated with diabetes

    Near-Infrared Properties of Metal-poor Globular Clusters in the Galactic Bulge Direction

    Aims. J, H, and K' images obtained from the near-infrared imager CFHTIR on the Canada-France-Hawaii Telescope are used to derive the morphological parameters of the red giant branch (RGB) in the near-infrared color-magnitude diagrams for 12 metal-poor globular clusters in the Galactic bulge direction. Using the compiled data set of the RGB parameters for the observed 12 clusters, in addition to the previously studied 5 clusters, we discuss the properties of the RGB morphology for the clusters and compare them with the calibration relations for the metal-rich bulge clusters and the metal-poor halo clusters. Methods. The photometric RGB shape indices such as colors at fixed magnitudes of MK = MH = (-5.5, -5, -4, and -3), magnitudes at fixed colors of (J - K)o = (J - H)o = 0.7, and the RGB slope are measured from the fiducial normal points defined in the near- infrared color-magnitude diagrams for each cluster. The magnitudes of RGB bump and tip are also estimated from the differential and cumulative luminosity functions of the selected RGB stars. The derived RGB parameters have been used to examine the overall behaviors of the RGB morphology as a function of cluster metallicity. Results. The correlations between the near-infrared photometric RGB shape indices and the cluster metallicity for the programme clusters compare favorably with the previous observational calibration relations for metal-rich clusters in the Galactic bulge and the metal-poor halo clusters. The observed near-infrared magnitudes of the RGB bump and tip for the investigated clusters are also in accordance with the previous calibration relations for the Galactic bulge clusters.Comment: 12 pages, 9 figures, accepted for publication in Astronomy & Astrophysic

    Identifying central symptom clusters and correlates in children with acute leukemia undergoing chemotherapy: a network analysis

    BackgroundPrevious studies have examined symptom clusters in children with acute leukemia, yet a knowledge gap persists regarding central symptom clusters and their influencing factors. By identifying these central clusters and associated factors, healthcare providers can enhance their understanding and effective management of symptoms. Our study seeks to address this gap by identifying symptom clusters, exploring central clusters, and investigating the demographic and health-related factors associated with these clusters in children with acute leukemia undergoing chemotherapy.MethodsA total of 586 children with acute leukemia from January 2021 to April 2023 were recruited from China. They were investigated using Memorial Symptom Assessment Scale 10-18 during chemotherapy. The principal component analysis was used to identify the symptom clusters. An association network was conducted to describe the relationships among symptoms and clusters. A multiple linear model was used to investigate the associated factors for the severity of overall symptoms and each symptom cluster.ResultsFive clusters were identified, including oral and skin cluster, somatic cluster, self-image disorder cluster, gastrointestinal cluster and psychological cluster. Gastrointestinal cluster was the most central symptom cluster. Age, sex, clinical classification, number of having chemotherapy and education degree and marital status of the primary caregiver are associated with the severity of these five symptom clusters.ConclusionOur study highlights the importance of evaluating symptom clusters in children with acute leukemia during chemotherapy. Specifically, addressing gastrointestinal symptoms is crucial for effective symptom management and overall care

    Acute Physiology and Chronic Health Evaluation II and Simplified Acute Physiology Score II in Predicting Hospital Mortality of Neurosurgical Intensive Care Unit Patients

    We study the predictive power of Acute Physiology and Chronic Health Evaluation II (APACHE II) and Simplified Acute Physiology Score II (SAPS II) in neurosurgical intensive care unit (ICU) patients. Retrospective investigation was conducted on 672 consecutive ICU patients during the last 2 yr. Data were collected during the first 24 hours of admission and analyzed to calculate predicted mortality. Mortality predicted by two systems was compared and, multivariate analyses were then performed for subarachnoid hemorrhage (SAH) and traumatic brain injury (TBI) patients. Observed mortality was 24.8% whereas predicted mortalities were 37.7% and 38.4%, according to APACHE II and SAPS II. Calibration curve was close to the line of perfect prediction. SAPS II was not statistically significant according to a Lemeshow-Hosmer test, but slightly favored by area under the curve (AUC). In SAH patients, SAPS II was an independent predictor for mortality. In TBI patients, both systems had independent prognostic implications. Scoring systems are useful in predicting mortality and measuring performance in neurosurgical ICU setting. TBI patients are more affected by systemic insults than SAH patients, and this discrepancy of predicting mortality in each neurosurgical disease prompts us to develop a more specific scoring system targeted to cerebral dysfunction
