    Anaphylaxis after jellyfish ingestion with no history of stings: a pediatric case report

    Background Jellyfish stings are known to induce allergic skin reactions; however, case reports of anaphylaxis after jellyfish ingestion have been increasing, especially in Asian countries. Some cases of anaphylaxis after jellyfish ingestion have been reported in patients with a previous history of frequent jellyfish stings. Herein, we report a pediatric patient with anaphylaxis after jellyfish ingestion with no history of jellyfish stings. Case presentation A 14-year-old girl developed two episodes of anaphylaxis, and her diet diaries revealed that edible jellyfish was common to the meals in both the anaphylaxis events. A skin prick test using five types of edible jellyfish products revealed a positive reaction to some jellyfish, and anaphylaxis was observed after the ingestion of jellyfish in an oral food challenge test. She had no history of jellyfish stings or frequent swimming in the ocean. The basophil activation test showed positive results on stimulation with extracts from various types of edible jellyfish. We observed serum immunoglobulin E (IgE) reactivity to purified jellyfish collagen and jellyfish acid-soluble extracts. Moreover, immunoblotting analysis showed IgE reactivity to two bands at approximately 40 and 70 kDa using purified jellyfish collagen, which may be a causative antigen. Conclusions Edible salted jellyfish can be one of the causative foods of anaphylaxis. Clinicians should be aware of the possibility of anaphylactic reactions due to jellyfish ingestion even without a history of jellyfish stings

    水耕栽培によって,生育温度と培地の栄養レベルがカ一ネーションの生育に及ぼす影饗を調査した. ‘レナ’の発根苗を15,20,25,30℃の温度で栽培し,各温度区ごとに栄養レベルを全窒素50-150ppm,りん酸10-60ppm,カリ20-120ppmの範囲とした区を設けた. 結果を要約するとつぎのとうりである. 1.生育温度と栄養レベルは生育に大きく影響した. 生育温度が15℃から20℃の間での好適栄養レベルは全窒素75-125ppm,りん酸30-50ppm,カリ60-120ppmの範囲にあるものと思われた. 2.もっともよい生育を示したのは生育温度を20℃,栄養レベルを全窒素75ppm,りん酸30ppm,カリ60ppmとした区であった. 3.上記の結果とは対照的に,生育温度を30℃とした区においては栄養レベルの生育に及ぼす影響が明らかでなく、茎長や乾物重あるいは肥料の吸収量なども減少した. 4.節数は,生育温度が高いほど増加した

    Multiple myeloma continues to be a lethal malignancy despite the development of treatments such as high-dose chemotherapy combined with stem cell transplantation. Multiple myeloma arises through an accumulation of multiple genetic anges, including immunoglobulin gene rearrangements involved in Cyclin D. The main difficulties in multiple myeloma treatments are drug-resistance. DNA methylation of the5\u27 CpG islands of genes is often found in multiple myeloma. To screen for he genes involved in tumorigenesis of multiple myeloma, which are silenced by DNA methylation, we performed cDNA microarray analysis using multiple myeloma cell lines treated with demethylating agent5-aza-2-deoxycytidine (DAC), and entified RASD1, a dexamethasone (Dex)-inducible gene, as one of the targets of epigenetic changes. Inactivation of RASD1 was found to correlate with resistance to Dex, and treatment of multiple myeloma cells with DAC restored sensitivity to Dex. These findings suggest the involvement of epigenetic gene silencing in multiple myeloma progression and drug-resistance, and the usefulness of demethylation therapy for multiple myeloma treatment. Crystal Growth and Design 23, 4285 (2023); https://doi.org/10.1021/acs.cgd.3c00108.In this study, we investigated calcium oxalate (CaOx) kidney stones and showed direct evidence of the solution-mediated phase transition of calcium oxalate dihydrate (COD; the metastable phase) to calcium oxalate monohydrate (COM; the stable phase). We examined the crystal phases, crystal textures, and protein distributions within thin sections of calcium oxalate kidney stones. Observation with a polarized-light microscope showed that the outline of the mosaic texture, in which COM crystals are assembled in a mosaic pattern, roughly coincides with COD’s crystallographically stable face angles. Microfocus X-ray CT measurement captured the intermediate process of the phase transition, starting inside the COD single crystal and gradually transforming to COM crystals. In addition, the distribution of osteopontin and prothrombin fragment-1, common proteins contained in urine and visualized by multicolor fluorescence immunostaining, showed no apparent striations inside the COM single crystals with the mosaic texture, although the striation is apparent inside the COD single crystals. This is probably because the phase transition of mosaic-like COM occurred in a semiclosed system inside the COD single crystal, so the effect of periodic (day-night, seasonal, etc.) urinary protein concentration changes was small. On the other hand, striations were visible in concentrically laminated COM. This indicated that concentrically laminated COM formed in response to the changes in urinary protein concentrations. From the above, we conclude that the COD single crystals and the concentrically laminated COM seen in CaOx stones are primary structures, and the mosaic COM is a secondary structure that is a pseudomorph formed by the solution-mediated phase transition from COD single crystals

    Photoreactions of Sc3N@C80 with Disilirane, Silirane, and Digermirane: A Photochemical Method to Separate Ih and D5h Isomers

    Under photoirradiation, Sc3N@Ih-C80 reacted readily with disilirane 1, silirane 4, and digermirane 7 to afford the corresponding 1:1 adducts, whereas Sc3N@D5h-C80 was recovered without producing those adducts. Based on these results, we described a novel method for the exclusive separation of Ih and D5h isomers of Sc3N@C80. The method includes three procedures: selective derivatization of Sc3N@Ih-C80 using 1, 4, and 7, facile HPLC separation of pristine Sc3N@D5h-C80 and Sc3N@Ih-C80 derivatives, and thermolysis of Sc3N@Ih-C80 derivatives to collect pristine Sc3N@Ih-C80. In addition, laser flash photolysis experiments were conducted to elucidate the reaction mechanism. Decay of the transient absorption of 3Sc3N@Ih-C80* was observed to be enhanced in the presence of 1, indicating the quenching process. When Sc3N@D5h-C80 was used, the transient absorption was much less intensive. Therefore, the quenching of 3Sc3N@D5h-C80* by 1 could not be confirmed. Furthermore, we applied time-dependent density functional theory (TD-DFT) calculations of the photoexcited states of Sc3N@C80 to obtain insights into the reaction mechanism

    Effect of soy protein on the muscle in human

    Background : In recent years, the number of bedridden people is rapidly increasing due to aging or lack of exercise in Japan. This problem is becoming more serious, since there is no countermeasure against it. In the present study, we designed to investigate whether dietary proteins, especially soy, had beneficial effects on skeletal muscle in 59 volunteers with various physical activities. Methods : We subjected 59 volunteers with various physical activities to meal intervention examination. Persons with low and high physical activities were divided into two dietary groups, the casein diet group and the soy diet group. They ate daily meals supplemented with 7.8 g of powdered casein or soy protein isolate every day for 30 days. Bedridden patients in hospitals were further divided into three dietary groups : the no supplementation diet group, the casein diet group and the soy diet group. They were also subjected to a blood test, a urinalysis, magnetic resonance imaging analysis and muscle strength test of the knee before and after the meal intervention study. Results : Thirty-day soy protein supplementation significantly increased skeletal muscle volume in participants with low physical activity, compared with 30-day casein protein supplementation. Both casein and soy protein supplementation increased the volume of quadriceps femoris muscle in bedridden patients. Consistently, soy protein significantly increased their extension power of the knee, compared with casein protein. Although casein protein increased skeletal muscle volume more than soy protein in bedridden patients, their muscle strength changes by soy protein supplementation were bigger than those by casein protein supplementation. Conclusions : The supplementation of soy protein would be one of the effective foods which prevent the skeletal muscle atrophy caused by immobilization or unloading

    An attenuated vaccinia vaccine encoding the severe acute respiratory syndrome coronavirus-2 spike protein elicits broad and durable immune responses, and protects cynomolgus macaques and human angiotensin-converting enzyme 2 transgenic mice from severe acute respiratory syndrome coronavirus-2 and its variants

    As long as the coronavirus disease-2019 (COVID-19) pandemic continues, new variants of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) with altered antigenicity will emerge. The development of vaccines that elicit robust, broad, and durable protection against SARS-CoV-2 variants is urgently required. We have developed a vaccine consisting of the attenuated vaccinia virus Dairen-I (DIs) strain platform carrying the SARS-CoV-2 S gene (rDIs-S). rDIs-S induced neutralizing antibody and T-lymphocyte responses in cynomolgus macaques and human angiotensin-converting enzyme 2 (hACE2) transgenic mice, and the mouse model showed broad protection against SARS-CoV-2 isolates ranging from the early-pandemic strain (WK-521) to the recent Omicron BA.1 variant (TY38-873). Using a tandem mass tag (TMT)-based quantitative proteomic analysis of lung homogenates from hACE2 transgenic mice, we found that, among mice subjected to challenge infection with WK-521, vaccination with rDIs-S prevented protein expression related to the severe pathogenic effects of SARS-CoV-2 infection (tissue destruction, inflammation, coagulation, fibrosis, and angiogenesis) and restored protein expression related to immune responses (antigen presentation and cellular response to stress). Furthermore, long-term studies in mice showed that vaccination with rDIs-S maintains S protein-specific antibody titers for at least 6 months after a first vaccination. Thus, rDIs-S appears to provide broad and durable protective immunity against SARS-CoV-2, including current variants such as Omicron BA.1 and possibly future variants
