    Force transduction by Triton cytoskeletons

    Force-initiated signal transduction can occur either via membrane-based ionic mechanisms or through changes in cytoskeletal–matrix linkages. We report here the stretch-dependent binding of cytoplasmic proteins to Triton X-100 cytoskeletons of L-929 cells grown on collagen-coated silicone. Triton X-100–insoluble cytoskeletons were stretched by 10% and incubated with biotinylated cytoplasmic proteins. Analysis with two-dimensional gel electrophoresis showed stretch-dependent binding of more than 10 cytoplasmic protein spots. Bound cytoplasmic proteins were purified by a photocleavable biotin tag and stretch-dependent binding of paxillin, focal adhesion kinase, and p130Cas was found, whereas the binding of vinculin was unchanged and actin binding decreased with stretch. Paxillin binding upon stretch was morphologically and biochemically similar in vitro and in vivo, that is, enhanced in the periphery and inhibited by the tyrosine phosphatase inhibitor, phenylarsine oxide. Thus, we suggest that transduction of matrix forces occurs through force-dependent conformation changes in the integrated cytoskeleton

    A Complete Survey of the Central Molecular Zone in NH3

    We present a map of the major part of the central molecular zone (CMZ) of simultaneous observations in the NH3 (J,K) = (1,1) and (2,2) lines using the Kagoshima 6-m telescope. The mapped area is -1.000 < l < 1.625 deg, -0.375 < b < +0.250 deg. The kinetic temperatures derived from the (2,2) to (1,1) intensity ratios are 20--80 K or exceed 80 K. The gases corresponding to temperature of 20--80 K and > 80 K contain 75% and 25% of the total NH3 flux, respectively. These temperatures indicate that the dense molecular gas in the CMZ is dominated by gas that is warmer than the majority of the dust present there. A comparison with the CO survey by Sawada et al. (2001) shows that the NH3 emitting region is surrounded by a high pressure region on the l-v plane. Although NH3 emission traces dense gas, it is not extended into a high pressure region. Therefore, the high pressure region is less dense and has to be hotter. This indicates that the molecular cloud complex in the Galactic center region has a ``core'' of dense and warm clouds which are traced by the NH3 emission, and an ``envelope'' of less dense and hotter gas clouds. Besides heating by ambipolar diffusion, the hot plasma gas emitting the X-ray emission may heat the hot ``envelope''.Comment: 21 pages, 14 figures, accepted for PAS

    NH3 in the Galactic Center is formed in Cool Conditions

    It is an open question why the temperature of molecular gas in the Galactic center region is higher than that of dust. To address this problem, we made simultaneous observations in the NH_3 (J,K) = (1,1), (2,2), and (3,3) lines of the central molecular zone (CMZ) using the Kagoshima 6 m telescope. The ortho-to-para ratio of NH_3 molecules in the CMZ is 1.5--3.5 at most observed area. This ratio is higher than the statistical equilibrium value, and suggests that the formation temperature of NH_3 is 11--20 K. This temperature is similar to the dust temperature estimated from the submillimeter and infrared continuum. This result suggests that the NH_3 molecules in the CMZ were produced on dust grains with the currently observed temperature (11--20 K), and they were released into the gas phase by supernova shocks or collisions of dust particles. The discrepancy between warm molecular gas and cold dust can be explained by the transient heating of the interstellar media in the CMZ approximately 10^5 years ago when the NH_3 molecules were released from the dust.Comment: 19 pages, 12 figures, accepted for PAS

    Increases in Nonspecific Immunoglobulin E and Eosinophils after H. pylori Eradication

    Helicobacter pylori infection has been reported to be inversely associated with allergic disorders. We by chance experienced a patient with atrophic gastritis who presented marked elevations of both nonspecific serum immunoglobulin E and eosinophil counts after H. pylori eradication. A 49-year-old Japanese man received eradication of H. pylori using lansoprazole 60 mg/day, amoxicillin 1,500 mg/day, and clarithromycin 400 mg/day for 7 days. Serum immunoglobulin E increased to more than four times its pretreatment level, 306 → 485 → 1,325 U/ml, and peripheral eosinophil counts increased to more than three times, 99 → 139 → 298 per μl. Deducing from the current case, H. pylori eradication might develop allergic disorders in some patients

    A New Candidate Supporting Drug, Rikkunshito, for the QOL in Advanced Esophageal Cancer Patients with Chemotherapy Using Docetaxel/5-FU/CDDP

    Purpose. Docetaxel/5-FU/CDDP (DFP) therapy is a useful treatment for advanced esophageal cancer. However, adverse reactions such as chemotherapy-induced nausea and vomiting (CINV) interfere often with continuation of the chemotherapy. We investigated the efficacy of rikkunshito (TJ-43) on CINV. Methods. Nineteen patients who were going to undergo DFP therapy were enrolled. They were assigned to the following two groups: a TJ-43-treated group and -nontreated group. The following parameters were compared between the 2 groups: (1) the frequency of symptoms occurred, (2) vomiting, nausea, and anorexia score, and (3) QOL score. Results. The incidence of symptoms was lower in the TJ-43-treated group than that in the control group. The nausea score of the TJ-43-treated group was significantly lower than that of the control group. In the QOL score, the mood score and the ADL score decreased significantly in the control group. Conclusion. We recommend TJ-43 administration in patients undergoing DFP chemotherapy

    Isolation of cDNA Encoding 7H6-Reactive Polypeptide Defines a New Class of Protein with alpha-Helical Coiled-Coil Structure and DA-Box Similar to Yeast Chromosomal Segregation Proteins

    7H6 monoclonal antibody was recently developed in our laboratory by im-munizing mice with a bile canaliculus-rich fraction of the rat liver. The anti-body reacted with a novel 155 Kd polypeptide designated 7H6 antigen that specifically localizes at tight junctions of various epithelia. Correlations of the paracellular barrier function of the tight junction with expression of the 7H6 antigen at the cell border have suggested important roles of this polypeptide for the maintenance of tight junctional functions. As the first step for the analysis of the antigen at the molecular level, we isolated a series of cDNA clones encod-ing 7H6-reactive polypeptides. Five clones were isolated by immunoscreening. Among them a clone designated RL5.3 which carries the largest 5.3Kb insert was characterized in this study. Both plaque screening and immunoblotting of the fusion protein produced by the RL5.3 clone with lysogen confirmed that the pro-tein specifically reacts with the 7H6 monoclonal antibody. Using DNA fragmentsof the RL5.3 clone, 21 clones were further identified. Studies with restriction enzymes and probe hybridization revealed that all the cDNA clones were derived from a single class of transcripts. A partial sequence identified one open reading frame with an α-helical coiled-coil structure and highly conserved aspartate (D)- alanine (A) residues with a helix-loop-helix structure corresponding to DA- box. Since this domain has been specifically found in yeast chromosomal segregation proteins (SMC1, CUT3 and CUT14), the polypeptide encoded by the RL5.3 clone provides the first rodent counterpart of these protein family. Yeast is known to be lethal when SMC and CUT proteins are deleted, suggesting essential roles of these proteins for cell cycle progression as a regulator for chromosomal segregation. Identification of a mammalian counterpart of this pro-tein family may give us some clues for a better understanding of fundamental regulatory mechanisms in the function of tigh junctions

    The geomorphology, color, and thermal properties of Ryugu: Implications for parent-body processes

    The near-Earth carbonaceous asteroid 162173 Ryugu is thought to have been produced from a parent body that contained water ice and organic molecules. The Hayabusa2 spacecraft has obtained global multi-color images of Ryugu. Geomorphological features present include a circum-equatorial ridge, east/west dichotomy, high boulder abundances across the entire surface, and impact craters. Age estimates from the craters indicate a resurfacing age of ≲ 106 years for the top 1-meter layer. Ryugu is among the darkest known bodies in the Solar System. The high abundance and spectral properties of boulders are consistent with moderately dehydrated materials, analogous to thermally metamorphosed meteorites found on Earth.
