    Understanding cellular internalization pathways of silicon nanowires

    BACKGROUND: Understanding how cells interact with nanomaterials is important for rational design of nanomaterials for nanomedicine and transforming them for clinical applications. Particularly, the mechanism for one-dimensional (1D) nanomaterials with high aspect ratios still remains unclear. RESULTS: In this work, we present amine-functionalized silicon nanowires (SiNW-NH2) entering CHO-β cells via a physical membrane wrapping mechanism. By utilizing optical microscopy, transmission electron microscopy, and confocal fluorescence microscopy, we successfully visualized the key steps of internalization of SiNW-NH2 into cells. CONCLUSION: Our results provide insight into the interaction between 1D nanomaterials and confirm that these materials can be used for understanding membrane mechanics through physical stress exerted on the membrane

    Theory of Electron-Phonon Dynamics in Insulating Nanoparticles

    We discuss the rich vibrational dynamics of nanometer-scale semiconducting and insulating crystals as probed by localized electronic impurity states, with an emphasis on nanoparticles that are only weakly coupled to their environment. Two principal regimes of electron-phonon dynamics are distinguished, and a brief survey of vibrational-mode broadening mechanisms is presented.     MicroRNA-155 expression is independently predictive of outcome in chordoma.

    BackgroundChordoma pathogenesis remains poorly understood. In this study, we aimed to evaluate the relationships between microRNA-155 (miR-155) expression and the clinicopathological features of chordoma patients, and to evaluate the functional role of miR-155 in chordoma.MethodsThe miRNA expression profiles were analyzed using miRNA microarray assays. Regulatory activity of miR-155 was assessed using bioinformatic tools. miR-155 expression levels were validated by reverse transcription-polymerase chain reaction. The relationships between miR-155 expression and the clinicopathological features of chordoma patients were analyzed. Proliferative, migratory and invasive activities were assessed by MTT, wound healing, and Matrigel invasion assays, respectively.ResultsThe miRNA microarray assay revealed miR-155 to be highly expressed and biologically active in chordoma. miR-155 expression in chordoma tissues was significantly elevated, and this expression correlated significantly with disease stage (p = 0.036) and the presence of metastasis (p = 0.035). miR-155 expression also correlated significantly with poor outcomes for chordoma patients (hazard ratio, 5.32; p = 0.045). Inhibition of miR-155 expression suppressed proliferation, and the migratory and invasive activities of chordoma cells.ConclusionsWe have shown miR-155 expression to independently affect prognosis in chordoma. These results collectively indicate that miR-155 expression may serve not only as a prognostic marker, but also as a potential therapeutic target in chordoma

    Examining Incoming Credit Differences Between First-Year and Transfer Students

    In this study, the difference in the number of initial credits between incoming transfer and first-year students entering a land grant university in a professional education program was examined (N = 488). A multivariate analysis of variance revealed that transfer students transferred significantly more total credits and more credits that counted toward degree programs than did first-year students. Undergraduates who had graduated from small high schools transferred more credits and more credits that counted than did those from large high schools. However, first-year students transferred a significantly higher percentage of total credits that counted toward the degree programs than did transfer students. Implications for advisors, institutions, and policy makers are discussed

    Handling linkage disequilibrium in linkage analysis using dense single-nucleotide polymorphisms

    The presence of linkage disequilibrium violates the underlying assumption of linkage equilibrium in most traditional multipoint linkage approaches. Studies have shown that such violation leads to bias in qualitative trait linkage analysis when parental genotypes are unavailable. Appropriate handling of marker linkage disequilibrium can avoid such false positive evidence. Using the rheumatoid arthritis simulated data from Genetic Analysis Workshop 15, we examined and compared the following three approaches to handle linkage disequilibrium among dense markers in both qualitative and quantitative trait linkage analyses: a simple algorithm; SNPLINK, methods for marker selection; and MERLIN-LD, a method for modeling linkage disequilibrium by creating marker clusters. In analysis ignoring linkage disequilibrium between markers, we observed LOD score inflation only in the affected sib-pair linkage analysis without parental genotypes; no such inflation was present in the quantitative trait locus linkage analysis with severity as our phenotype with or without parental genotypes. Using methods to model or adjust for linkage disequilibrium, we found a substantial reduction of inflation of LOD score in affected sib-pair linkage analysis. Greater LOD score reduction was observed by decreasing the amount of tolerable linkage disequilibrium among markers selected or marker clusters using MERLIN-LD; the latter approach showed most reduction. SNPLINK performed better with selected markers based on the D' measure of linkage disequilibrium as opposed to the r2 measure and outperformed the simple algorithm. Our findings reiterate the necessity of properly handling dense markers in linkage analysis, especially when parental genotypes are unavailable

    Applying the Rasch Model to Evaluate the Self-Directed Online Learning Scale (SDOLS) for Graduate Students

    With the rapid growth of online learning and the increased attention paid to student attrition in online programs, much research has been aimed at studying the effectiveness of online education to improve students’ online learning experience and student retention. Utilizing the online learning literature as a multi-faceted theoretical framework, the study developed and employed a new survey instrument. The Self-Directed Online Learning Scale (SDOLS) was used to examine graduate student perceptions of effectiveness of online learning environments as demonstrated by their ability to take charge of their own learning, and to identify key factors in instructional design for effective improvements. The study applied the Rasch rating scale model to evaluate and validate SDOLS through a psychometric lens to establish the reliability and validity of SDOLS. Results from Rasch analysis addressed two research questions. First, evidence was found to generally support the new instrument as being psychometrically sound but three problematic items were also identified as grounds for future improvement of SDOLS. Second, the study assessed the importance of various factors as measured by the SDOLS items in contributing to students’ ability to self-manage their own online learning. Finally, the new instrument is expected to contribute to the work of various stakeholders in online education and can serve to improve students’ online learning experience and effectiveness, increase online retention rates, and reduce online dropouts

    Modelling Heat Transfer of Carbon Nanotubes

    Modelling heat transfer of carbon nanotubes is important for the thermal management of nanotube-based composites and nanoelectronic device. By using a finite element method for three-dimensional anisotropic heat transfer, we have simulated the heat conduction and temperature variations of a single nanotube, a nanotube array and a part of nanotube-based composite surface with heat generation. The thermal conductivity used is obtained from the upscaled value from the molecular simulations or experiments. Simulations show that nanotube arrays have unique cooling characteristics due to its anisotropic thermal conductivity.Comment: 10 pages, 4 figure
