    Curcumin combining with si-MALAT1 inhibits the invasion and migration of colon cancer SW480 cells

    To study the effect of small interfering RNA targeting metastasis-associated lung adenocarcinoma transcript1 (si-MALAT1) combining with curcumin on the invasion and migration abilities of human colon cancer SW480 cells, and to explore the involved molecular mechanism. The recombinant lentiviral vector expressing si-MALAT1 was constructed, and its titer was determined by gradient dilution method. The colon cancer SW480 cells with stable expression of si-MALAT1 was established, followed by treatment with curcumin at different concentrations. The effect of curcumin or si-MALAT1 alone and the combination of the two on the cell activity was detected by MTT assay. The cell invasion and migration abilities were detected by transwell and scratch-wound assay. The relative expression level of MALAT1 was detected by RT-qPCR. The protein expression was determined by Western blot analysis. The IC50 of curcumin alone was 77.69 µmol/L, which was 51.17 mol/Lwhen combined with curcumin and random sequence. The IC50 of curcumin was 30.02 µmol/L when combined with si-MALAT1. The increased susceptibility multiples was 2.58. The wound healing rates were 30.9% and 67.5% after treatment with si-MALAT1 combined with curcumin for 24 hrs and 48 hrs, respectively. The numbers of invasion cells were 200±12, 162±13, 66±8, 53±4 and 16±3 after treatment with si-MALAT1 combined with curcumin for 48 hrs. The relative expression level of lncRNA-MALAT1 in the curcumin group was 68%, and the relative expression level of lncRNA-MALAT1 in si-MALAT1group was 56%, and that for the combination treatment group was about 21%. The protein expression levels of β- catenin, c-myc and cyclinD1 were significantly down-regulated upon treatment with certain concentration of si-MALAT1 alone or combined with curcumin.si-MALAT1 could significantly inhibit the invasion and migration of SW480 cells by enhancing the sensitivity of SW480 cells to curcumin. The mechanism involved mignt be related to the down-regulation of β-catenin, c-myc and cyclinD1 proteins

    AbstractBased on the combustion, explosions and air dynamics and related theory etc, this paper describes the mathematical model of gas explosion in detail, combined with the gas explosion transmission mechanism, make a research on two wave-three area structure of gas explosion and the energy change rule of the array face of precursor wave and the array face of flame wave, with the fluid dynamics analysis Fluent software, this paper makes a numerical simulation and analysis on the overpressure transmission rule when gas explosion takes place in different types roadways. The results of the study show that: Fluent software can be used to accurately simulate gas explosion condition, when explosion wave spreads in the roadway turns, the bigger of the overpressure value in corner, the stronger of the destructive power; when tunnel has bifurcation, the overpressure will release in bifurcation, but explosions wave with flame wave will produce more powerful destruction effect. The research results can be used as a certain reference for how to prevent and control the gas explosion, and how to reduce the power of the gas explosion etc

PC12 cell is a pheochromocytoma cell line from RattusNorvegicus, because it has some characters of nerve cells and easy to cultivate and passage, these cells have been proved to be a useful cell model to study nervous physiology and pharmacology. There are several of PC12 cells, American type culture collection supply two kinds of PC12 cell named PC12 cell and PC12Adh. There are high differentiation, low differentiation and undifferentiating in domestic. Although they are very similar, there are still some differences, and not every PC12 cell is effective for every experimental model. After compared, we hold that PC12Adh cell line is more suitable for neurite outgrowth studies under ROCK inhibitor than the PC12 cell line, PC12 cells that induced by NGF and high differentiated PC12 cell are similar to cerebral cortical neurons, they are suitable for various physiological and pathological study of nervous system. Undifferentiated PC12 cells due to low levels of dopamine, therefore it is not suitable for study on neural cells

    Genome-wide search for the genes accountable for the induced resistance to HIV-1 infection in activated CD4+ T cells: apparent transcriptional signatures, co-expression networks and possible cellular processes

    BACKGROUND: Upon co-stimulation with CD3/CD28 antibodies, activated CD4 + T cells were found to lose their susceptibility to HIV-1 infection, exhibiting an induced resistant phenotype. This rather unexpected phenomenon has been repeatedly confirmed but the underlying cell and molecular mechanisms are still unknown. METHODS: We first replicated the reported system using the specified Dynal beads with PHA/IL-2-stimulated and un-stimulated cells as controls. Genome-wide expression and analysis were then performed by using Agilent whole genome microarrays and established bioinformatics tools. RESULTS: We showed that following CD3/CD28 co-stimulation, a homogeneous population emerged with uniform expression of activation markers CD25 and CD69 as well as a memory marker CD45RO at high levels. These cells differentially expressed 7,824 genes when compared with the controls on microarrays. Series-Cluster analysis identified 6 distinct expression profiles containing 1,345 genes as the representative signatures in the permissive and resistant cells. Of them, 245 (101 potentially permissive and 144 potentially resistant) were significant in gene ontology categories related to immune response, cell adhesion and metabolism. Co-expression networks analysis identified 137 “key regulatory” genes (84 potentially permissive and 53 potentially resistant), holding hub positions in the gene interactions. By mapping these genes on KEGG pathways, the predominance of actin cytoskeleton functions, proteasomes, and cell cycle arrest in induced resistance emerged. We also revealed an entire set of previously unreported novel genes for further mining and functional validation. CONCLUSIONS: This initial microarray study will stimulate renewed interest in exploring this system and open new avenues for research into HIV-1 susceptibility and its reversal in target cells, serving as a foundation for the development of novel therapeutic and clinical treatments

    A multiplex polymerase chain reaction (PCR) was designed for the simultaneous detection of four viruses involved in postweaning multisystemic wasting syndrome (PMWS) in pigs: porcine circovirus type 2 (PCV-2), type 2 porcine reproductive and respiratory syndrome virus (PRRSV), porcine parvovirus (PPV) and pseudorabies virus (PRV). Each of the four pairs of primers exclusively amplified the targeted fragment of the specific viruses. The sensitivity of the multiplex PCR, using purified plasmid constructs containing the specific viral target fragments, was 4.0×103, 4.5×103, 3.0×102 and 5.0×102 copies for PCV-2, type 2 PRRSV, PPV, PRV. Among 82 clinical samples, coinfection by PCV-2 and type 2 PRRSV was the most common. When compared with the virus isolation method commonly used to detect viruses, the multiplex PCR assay was found to be more sensitive and rapid and, as such, may prove to be a good alternative method for the detection of and differentiation in PMWS in pigs.Višestruka lančana reakcija polimerazom (PCR) razvijena je za istodobni dokaz četiriju virusa koji se nalaze kod sindroma kržljavosti odbijene prasadi: svinjskog cirkovirusa tip 2, virusa reprodukcijskog i respiratornog sindroma tip 2, parvovirusa i virusa bolesti Aujeszkoga. Svaki od četiri para početnica bio je specifičan isključivo za umnožavanje specifičnog fragmenta određenog virusa. Osjetljivost višestrukog PCR-a upotrebom pročišćenih konstrukata plazmida koji sadrže specifične virusne fragmente bila je na razini 4,0×103 kopija za svinjski cirkovirus tip 2, 4,5×103 kopija za virus reprodukcijskog i respiratornog sindroma, 3,0×102 za svinjski parvovirus i 5,0×102 kopija za virus bolesti Aujeszkoga. Među 82 klinička uzorka često je dokazana koinfekcija svinjskim cirkovirusom tip 2 i virusom respiratornog i reprodukcijskog sindroma. U usporedbi s izolacijom virusa, višestruka lančana reakcija polimerazom pokazala se osjetljivijom i bržom te se kao takva može rabiti kao alternativna metoda za dokaz i razlikovanje uzročnika sindroma kržljavosti prasadi

    A New Energy-Aware Flexible Job Shop Scheduling Method Using Modified Biogeography-Based Optimization

    Industry consumes approximately half of the total worldwide energy usage. With the increasingly rising energy costs in recent years, it is critically important to consider one of the most widely used energies, electricity, during the production planning process. We propose a new mathematical model that can determine efficient scheduling to minimize the makespan and electricity consumption cost (ECC) for the flexible job shop scheduling problem (FJSSP) under a time-of-use (TOU) policy. In addition to the traditional two subtasks in FJSSP, a new subtask called speed selection, which represents the selection of variable operating speeds, is added. Then, a modified biogeography-based optimization (MBBO) algorithm combined with variable neighborhood search (VNS) is proposed to solve the biobjective problem. Experiments are performed to verify the effectiveness of the proposed MBBO algorithm for obtaining an improved scheduling solution compared to the basic biogeography-based optimization (BBO) algorithm, genetic algorithm (GA), and harmony search (HS)

    Preparation of a Nanosized As2O3/Mn0.5Zn0.5Fe2O4 Complex and Its Anti-Tumor Effect on Hepatocellular Carcinoma Cells

    Manganese-zinc-ferrite nanoparticles (Mn0.5Zn0.5Fe2O4, MZF-NPs) prepared by an improved co-precipitation method and were characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD) and energy dispersive spectrometry (EDS). Then thermodynamic testing of various doses of MZF-NPs was performed in vitro. The cytotoxicity of the Mn0.5Zn0.5Fe2O4 nanoparticles in vitro was tested by the MTT assay. A nanosized As2O3/Mn0.5Zn0.5Fe2O4 complex was made by an impregnation process. The complex’s shape, component, envelop rate and release rate of As2O3 were measured by SEM, EDS and atom fluorescence spectrometry, respectively. The therapeutic effect of nanosized As2O3/Mn0.5Zn0.5Fe2O4 complex combined with magnetic fluid hyperthermia (MFH) on human hepatocelluar cells were evaluated in vitro by an MTT assay and flow cytometry. The results indicated that Mn0.5Zn0.5Fe2O4 and nanosized As2O3/Mn0.5Zn0.5Fe2O4 complex were both prepared successfully. The Mn0.5Zn0.5Fe2O4 nanoparticles had powerful absorption capabilities in a high-frequency alternating electromagnetic field, and had strong magnetic responsiveness. Moreover, Mn0.5Zn0.5Fe2O4 didn’t show cytotoxicity in vitro. The therapeutic result reveals that the nanosized As2O3/Mn0.5Zn0.5Fe2O4 complex can significantly inhibit the growth of hepatoma carcinoma cells
