Estimation of microphytobenthic resuspension fluxes in a shallow lagoon in Hokkaido, Japan
We conducted field sampling in a subarctic shallow lagoon (Hichirippu Lagoon) in the eastern part of Hokkaido, Japan. We investigated the chemical composition of the water column, sediment, and sinking particles collected by the sediment trap. The standing stock of chlorophyll a (Chl-a) in the water column and surface sediment were 0.4 to 9.3 and 35.9 to 184 mg m−2, respectively. Using stable isotope analysis, the contribution rate of microphytobenthos to the sinking particles was found to range from 63 to 74%. The average Chl-a content in the sinking particles was significantly lower than that of the water column. Our results suggest that the contribution of phytoplankton present in the water column to the Chl-a collected in the traps is likely to be negligible. We assumed that the Chl-a fluxes obtained in this study were microphytobenthic resuspension fluxes. The daily flux of Chl-a accounted for 47.0 to 1,270% of the total standing stock of Chl-a in the water column. The mean relative percentage of daily Chl-a flux divided by the standing stock of Chl-a in the sediment was 6.5%, which indicates that approximately 7% of the microphytobenthos present in the sediment was resuspended, and 93% of the total succeeded at escaping the winnowing action. Although the resuspension phenomenon had little effect on the population of microphytobenthos, the resuspended microphytobenthos had a major impact on the total micro algal biomass in the water column. This is the first direct estimate of microphytobenthic resuspension flux in shallow estuaries
Reduction of T-Box 15 gene expression in tumor tissue is a prognostic biomarker for patients with hepatocellular carcinoma
Genome-wide analysis is widely applied to detect molecular alterations during oncogenesis and tumor progression. We analyzed DNA methylation profiles of hepatocellular carcinoma (HCC), and investigated the clinical role of most heypermethylated of tumor, encodes T-box 15 (TBX15), which was originally involved in mesodermal differentiation. We conducted a genome-wide analysis of DNA methylation of tumor and non-tumor tissue of 15 patients with HCC, and revealed TBX15 was the most hypermethylated gene of tumor (Beta-value in tumor tissue = 0.52 compared with non-tumor tissue). Another validation set, which comprised 58 HCC with radical resection, was analyzed to investigate the relationships between tumor phenotype and TBX15 mRNA expression. TBX15 mRNA levels in tumor tissues were significantly lower compared with those of nontumor tissues (p < 0.0001). When we assigned a cutoff value = 0.5-fold, the overall survival 5-year survival rates of the low-expression group (n = 17) were significantly shorter compared with those of the high-expression group (n = 41) (43.3% vs. 86.2%, p = 0.001). Multivariate analysis identified low TBX15 expression as an independent prognostic factor for overall and disease-free survival. Therefore, genome-wide DNA methylation profiling indicates that hypermethylation and reduced expression of TBX15 in tumor tissue represents a potential biomarker for predicting poor survival of patients with HCC
Muscle mass, quality, and strength; physical function and activity; and metabolic status in cachectic patients with head and neck cancer
Background & aims: Cancer cachexia is commonly associated with poor prognosis in patients with head and neck cancer (HNC). However, its pathophysiology and treatment are not well established. The current study aimed to assess the muscle mass/quality/strength, physical function and activity, resting energy expenditure (REE), and respiratory quotient (RQ) in cachectic patients with HNC.
Methods: This prospective cross-sectional study analyzed 64 patients with HNC. Body composition was measured via direct segmental multifrequency bioelectrical impedance analysis, and muscle quality was assessed using echo intensity on ultrasonography images. Muscle strength was investigated utilizing handgrip strength and isometric knee extension force (IKEF). Physical function was evaluated using the 10-mwalking speed test and the five times sit-to-stand (5-STS) test. Physical activity was examined using a wearable triaxial accelerometer. REE and RQ were measured via indirect calorimetry. These parameters were compared between the cachectic and noncachectic groups.
Results: In total, 23 (36%) patients were diagnosed with cachexia. The cachectic group had a significantly lower muscle mass than the noncachectic group. Nevertheless, there was no significant difference in terms of fat between the two groups. The cachectic group had a higher quadriceps echo intensity and a lower handgrip strength and IKEF than the noncachectic group. Moreover, they had a significantly slower normal and maximum walking speed and 5 STS speed. The number of steps, total activity time, and time of activity (<3 Mets) did not significantly differ between the two groups. The cachectic group had a shorter time of activity (≥3 Mets) than the noncachectic group. Furthermore, the cachectic group had a significantly higher REE/body weight and REE/fat free mass and a significantly lower RQ than the noncachectic group.
Conclusions: The cachectic group had a lower muscle mass/quality/strength and physical function and activity and a higher REE than the noncachectic group. Thus, REE and physical activity should be evaluated to determine energy requirements. The RQ was lower in the cachectic group than that in the noncachectic group, indicating changes in energy substrate. Further studies must be conducted to examine effective nutritional and exercise interventions for patients with cancer cachexia
Visceral fat obesity is the key risk factor for the development of reflux erosive esophagitis in 40–69-years subjects
[Background] Visceral fat obesity can be defined quantitatively by abdominal computed tomography, however, the usefulness of measuring visceral fat area to assess the etiology of gastrointestinal reflux disease has not been fully elucidated. [Methods] A total of 433 healthy subjects aged 40–69 years (234 men, 199 women) were included in the study. The relationship between obesity-related factors (total fat area, visceral fat area, subcutaneous fat area, waist circumference, and body mass index) and the incidence of reflux erosive esophagitis was investigated. Lifestyle factors and stomach conditions relevant to the onset of erosive esophagitis were also analyzed. [Results] The prevalence of reflux erosive esophagitis was 27.2% (118/433; 106 men, 12 women). Visceral fat area was higher in subjects with erosive esophagitis than in those without (116.6 cm2 vs. 64.9 cm2, respectively). The incidence of erosive esophagitis was higher in subjects with visceral fat obesity (visceral fat area ≥ 100 cm2) than in those without (61.2% vs. 12.8%, respectively). Visceral fat obesity had the highest odds ratio (OR) among obesity-related factors. Multivariate analysis showed that visceral fat area was associated with the incidence of erosive esophagitis (OR = 2.18), indicating that it is an independent risk factor for erosive esophagitis. In addition, daily alcohol intake (OR = 1.54), gastric atrophy open type (OR = 0.29), and never-smoking history (OR = 0.49) were also independently associated with the development of erosive esophagitis. [Conclusions] Visceral fat obesity is the key risk factor for the development of reflux erosive esophagitis in subjects aged 40–69 years
Survey of Period Variations of Superhumps in SU UMa-Type Dwarf Novae. VIII: The Eighth Year (2015-2016)
Continuing the project described by Kato et al. (2009, arXiv:0905.1757), we
collected times of superhump maxima for 128 SU UMa-type dwarf novae observed
mainly during the 2015-2016 season and characterized these objects. The data
have improved the distribution of orbital periods, the relation between the
orbital period and the variation of superhumps, the relation between period
variations and the rebrightening type in WZ Sge-type objects. Coupled with new
measurements of mass ratios using growing stages of superhumps, we now have a
clearer and statistically greatly improved evolutionary path near the terminal
stage of evolution of cataclysmic variables. Three objects (V452 Cas, KK Tel,
ASASSN-15cl) appear to have slowly growing superhumps, which is proposed to
reflect the slow growth of the 3:1 resonance near the stability border.
ASASSN-15sl, ASASSN-15ux, SDSS J074859.55+312512.6 and CRTS J200331.3-284941
are newly identified eclipsing SU UMa-type (or WZ Sge-type) dwarf novae.
ASASSN-15cy has a short (~0.050 d) superhump period and appears to belong to EI
Psc-type objects with compact secondaries having an evolved core. ASASSN-15gn,
ASASSN-15hn, ASASSN-15kh and ASASSN-16bu are candidate period bouncers with
superhump periods longer than 0.06 d. We have newly obtained superhump periods
for 79 objects and 13 orbital periods, including periods from early superhumps.
In order that the future observations will be more astrophysically beneficial
and rewarding to observers, we propose guidelines how to organize observations
of various superoutbursts.Comment: 123 pages, 162 figures, 119 tables, accepted for publication in PASJ

Survey of Period Variations of Superhumps in SU UMa-Type Dwarf Novae
We systematically surveyed period variations of superhumps in SU UMa-type
dwarf novae based on newly obtained data and past publications. In many
systems, the evolution of superhump period are found to be composed of three
distinct stages: early evolutionary stage with a longer superhump period,
middle stage with systematically varying periods, final stage with a shorter,
stable superhump period. During the middle stage, many systems with superhump
periods less than 0.08 d show positive period derivatives. Contrary to the
earlier claim, we found no clear evidence for variation of period derivatives
between superoutburst of the same object. We present an interpretation that the
lengthening of the superhump period is a result of outward propagation of the
eccentricity wave and is limited by the radius near the tidal truncation. We
interpret that late stage superhumps are rejuvenized excitation of 3:1
resonance when the superhumps in the outer disk is effectively quenched. Many
of WZ Sge-type dwarf novae showed long-enduring superhumps during the
post-superoutburst stage having periods longer than those during the main
superoutburst. The period derivatives in WZ Sge-type dwarf novae are found to
be strongly correlated with the fractional superhump excess, or consequently,
mass ratio. WZ Sge-type dwarf novae with a long-lasting rebrightening or with
multiple rebrightenings tend to have smaller period derivatives and are
excellent candidate for the systems around or after the period minimum of
evolution of cataclysmic variables (abridged).Comment: 239 pages, 225 figures, PASJ accepte
A liver-derived secretory protein, selenoprotein P, causes insulin resistance
金沢大学医薬保健研究域医学系The liver may regulate glucose homeostasis by modulating the sensitivity/resistance of peripheral tissues to insulin, by way of the production of secretory proteins, termed hepatokines. Here, we demonstrate that selenoprotein P (SeP), a liver-derived secretory protein, causes insulin resistance. Using serial analysis of gene expression (SAGE) and DNA chip methods, we found that hepatic SeP mRNA levels correlated with insulin resistance in humans. Administration of purified SeP impaired insulin signaling and dysregulated glucose metabolism in both hepatocytes and myocytes. Conversely, both genetic deletion and RNA interference-mediated knockdown of SeP improved systemic insulin sensitivity and glucose tolerance in mice. The metabolic actions of SeP were mediated, at least partly, by inactivation of adenosine monophosphate-activated protein kinase (AMPK). In summary, these results demonstrate a role of SeP in the regulation of glucose metabolism and insulin sensitivity and suggest that SeP may be a therapeutic target for type 2 diabetes. © 2010 Elsevier Inc
