    Identification of StBEL5 RNA as a long-distance mobile signal in short-day facilitated tuber formation

    In potato, the BEL1-like transcription factor, StBEL5 and its protein partner POTH1, regulate tuber formation by mediating hormone levels in the stolon tip. Heterografting experiments show that StBEL5 mRNA can move across the graft union to localize in stolon tips and enhance tuber formation. Over-expression of StBEL5 full-length transcripts including the untranslated regions (UTRs) endows transgenic lines with the capacity to overcome the long-day inhibitory effects on tuber formation.;In this study, the precise localization of endogenous StBEL5 mRNA and other gene specific transcripts in the vascular tissues were determined by laser microdissection coupled to laser pressure catapulting (LMPC) and following RT-PCR. The results demonstrate the presence of StBEL5 mRNA in phloem cells which is consistent with its role as a mobile RNA.;StBEL5 full length transcripts exhibited better mobility compared to its UTR-truncated form and StBEL14 transcripts in agroinfiltration experiments. No translation enhancement was observed for full length StBEL5 transcripts compared to its UTR-truncated transcripts by in vitro translation assay. This indicates involvement of StBEL5 UTRs in facilitating its long-distance movement. Further studies of applying EMSA and northwestern blotting to identify StBEL5 mRNA binding proteins may provide pivotal cues to understand the mechanism of phloem delivered StBEL5 RNA in this long-distance signaling pathway during tuber formation

    Crop residue management and fertilization effects on soil organic matter and associated biological properties

    Returning crop residue may result in nutrient reduction in soil in the first few years. A two-year field experiment was conducted to assess whether this negative effect is alleviated by improved crop residue management (CRM). Nine treatments (3 CRM and 3 N fertilizer rates) were used. The CRM treatments were (1) R0: 100 % of the N using mineral fertilizer with no crop residues return; (2) R: crop residue plus mineral fertilizer as for the R0; and (3) Rc: crop residue plus 83 % of the N using mineral and 17 % manure fertilizer. Each CRM received N fertilizer rates at 270, 360, and 450 kg N ha−1 year−1. At the end of the experiment, soil NO3-N was reduced by 33 % from the R relative to the R0 treatment, while the Rc treatment resulted in a 21 to 44 % increase in occluded particulate organic C and N, and 80 °C extracted dissolved organic N, 19 to 32 % increase in microbial biomass C and protease activity, and higher monounsaturated phospholipid fatty acid (PLFA):saturated PLFA ratio from stimulating growth of indigenous bacteria when compared with the R treatment. Principal component analysis showed that the Biolog and PLFA profiles in the three CRM treatments were different from each other. Overall, these properties were not influenced by the used N fertilizer rates. Our results indicated that application of 17 % of the total N using manure in a field with crop residues return was effective for improving potential plant N availability and labile soil organic matter, primarily due to a shift in the dominant microorganisms

    Stratospheric PULSE–continental cold air outbreak coupling relationships: Interannual and interdecadal changes

    Stratospheric processes and their role in weather and climate have attracted increasing interests. The correspondence between the occurrence of pulse-like, stronger stratospheric poleward warm airmass transport (PULSE) events and the continental-scale cold air outbreak (CAO) events in northern hemispheric winter is found to be unstable from year to year. This increases the difficulties in utilizing the more predictable stratospheric variability in the sub-seasonal forecasts of CAOs, which can cause cold hazards. Using the ERA5 reanalysis data covering 37 winters (November–March) in the period 1979–2015, this study categorizes the CAO events over mid-latitudes of Eurasia (CAO_EA) and those over North America (CAO_NA) into two groups: those coupled with and those decoupled with the PULSE events. The coupled CAOs are further categorized into events that are, respectively, lead-coupled and lag-coupled with PULSEs. The intensity and affected area of extremely cold temperatures tend to be larger during CAOs that are coupled with PULSEs, particularly during the CAO_NA events that are lag-coupled with PULSEs and the CAO_EA events that are lead-coupled with PULSEs. Remarkable interannual and interdecadal variations are observed in the percentage of CAOs that are coupled with PULSEs for each winter, which is an important reference for determining the window of opportunity for skillful sub-seasonal forecasts of CAO by using the stratospheric signals. At both interdecadal and interannual timescales, a warm phase of the El Niño–Southern Oscillation (ENSO) in winter is favorable for the higher lag-coupling rate of CAO_NA and the lead-coupling rate of CAO_EA, and vice versa. The ENSO signals related to the interdecadal changes of the CAO coupling rate in winter can be traced back to the previous winter, while an ENSO phase transition from the previous winter to the current winter is closely related to the interannual changes of the CAO coupling rate

    Evaluation of oral Lanzhou lamb rotavirus vaccine via passive transfusion with CD4+/CD8+ T lymphocytes

    AbstractLanzhou Lamb derived Rotavirus (RV) Vaccine (namely LLR) for children is only used in China. Since there were no reports on evaluation of LLR, even the data of phase IV clinical trial, we proceed the evaluation of LLR through focusing on T-cell to investigate whether LLR could induce the potential function involving in protection as a vaccine. Four groups of nude mice were transfused with CD4+/CD8+ T-cells isolated from LLR-immunized (primed) and LLR-unimmunized (naïve) mice via intraperitonea (i.p.) respectively. Consequently, the adoption mice were challenged with mice-origin wild rotavirus EDIM (Epizootic Diarrhea of Infant Mice) by intragastric administration. Series of fecal/serum samples were collected and viral shedding, then serum IgA/IgG and secreted IgA were assayed. Compared to the mice transfused with T lymphocytes from naïve mice, the nude mice transfused with CD4+ T lymphocytes from primed mice induce fecal and serum IgA increasing more rapidly, and have a shorter duration of virus shedding too. Whereas, no significant difference in virus clearance was found between the mice transfused with CD8+ T lymphocytes isolated from primed and naïve mice. Therefore, we cleared the distinct roles of transfused CD4+/CD8+ T lymphocytes for rotavirus clearance in nude mice, that the viral clearance conducted by CD4+ T lymphocytes. Meanwhile, it has ability to help induction of LLR specific immunogenicity. Comparing with the transfusion of cell from primed and naïve mice, LLR can induce CD4+ T lymphocytes memory which is a potential index to reflect the immunogenicity and protection, while CD8+ T lymphocytes remove rotavirus by CTL with little memory ability

    The ‘credibility paradox’ in China’s science communication: Views from scientific practitioners

    In contrast to increasing debates on China’s rising status as a global scientific power, issues of China’s science communication remain under-explored. Based on 21 in-depth interviews in three cities, this article examines Chinese scientists’ accounts of the entangled web of influence which conditions the process of how scientific knowledge achieves (or fails to achieve) its civic authority. A main finding of this study is a ‘credibility paradox’ as a result of the over-politicisation of science and science communication in China. Respondents report that an absence of visible institutional endorsements renders them more public credibility and better communication outcomes. Thus, instead of exploiting formal channels of science communication, scientists interviewed were more keen to act as ‘informal risk communicators’ in grassroots and private events. Chinese scientists’ perspectives on how to earn public support of their research sheds light on the nature and impact of a ‘civic epistemology’ in an authoritarian state

    Association between apathy in patients with maintenance dialysis and hospitalization or mortality: a prospective cohort study

    BackgroundPatients receiving maintenance dialysis experience increased rates of hospitalization and mortality. Apathy is associated with reduced quality of life and increased hospitalization, institutionalization, and death. Whether apathy contributes to poor outcomes in population undergoing maintenance dialysis remain unknown.MethodsWe conducted a prospective cohort study of maintenance dialysis population who were consecutively recruited at the Dialysis Center of Shanghai General Hospital between July 2017 and August 2018 and were followed up for 3 year. Apathy status was measured by the Apathy Evaluation Scale. The study outcomes were the occurrence of death and first hospitalization.ResultsA total of 647 participants included in this study, 274 (42.3%) had a current apathy and 373 (57.7%) were not. During the follow-up period, 394 (60.9%) were hospitalized, and 169 (26.1%) died. Kaplan–Meier analysis showed that the risks of hospitalization and mortality were significantly higher in individuals with apathy than in those without apathy (both p < 0.001). Apathy at baseline was associated with hospitalization and death both in univariate analysis and in all multivariable models (all p < 0.001).ConclusionApathy was highly prevalent and independently correlated with an increased risk of poor outcomes in patients with maintenance dialysis
