    Synthesis and characterization of interspecific trigenomic hybrids and allohexaploids between three cultivated Brassica allotetraploids and wild species Brassica fruticulosa

    Interspecific reciprocal crosses between three cultivated Brassica allotetraploids and one wild species Brassica fruticulosa (FF, 2n =16) were made and the trigenomic hybrids were produced only with embryo rescue. From the crosses with Brassica juncea (AABB, 2n = 36) and Brassica napus (AACC, 2n = 38), hybrids (F.AB, 2n = 26; F.AC, 2n = 27) were obtained only with B. fruticulosa as female, but the reciprocal crosses with Brassica carinata (BBCC, 2n = 34) gave rise to hybrids (F.BC/BC.F, 2n = 25). These hybrids showed an intermediate morphology and were sterile for male and female except those with B. carinata. All the triploids showed high frequency of bivalents during diakinesis and metaphase I (MI) of meiosis. The allohexaploids (FF.AABB, 2n = 52; FF.AACC, 2n = 54; BBCC.FF, 2n = 50) were synthesized by colchicine treatments of respective hybrids in vitro, which showed growth vigor and had larger stature than the hybrids, but were male sterile except one (FF.AABB). They produced progenies with the expected chromosome complements (F.AABB, 2n = 44; F.AACC, 2n = 46; BBCC.F, 2n = 42) after pollination by respective Brassica allotetraploids. In these allohexaploids, chromosomes were mainly paired as bivalents at diakinesis and segregated equally at anaphase I (AI) during meiosis of pollen mother cells (PMCs).     Health Disparities and Comparison of Psychiatric Medication Use before and after the COVID-19 Pandemic Lockdown among General Practitioner Practices in the North East of England.

    BackgroundPsychiatric medications play a vital role in the management of mental health disorders. However, the COVID-19 pandemic and subsequent lockdown limited access to primary care services, leading to an increase in remote assessment and treatment options to maintain social distancing. This study aimed to investigate the impact of the COVID-19 pandemic lockdown on the use of psychiatric medication in primary care settings.MethodsWe conducted a retrospective claims-based analysis of anonymized monthly aggregate practice-level data on anxiolytics and hypnotics use from 322 general practitioner (GP) practices in the North East of England, where health disparities are known to be higher. Participants were all residents who took anxiolytics and hypnotics from primary care facilities for two financial years, from 2019/20 to 2020/21. The primary outcome was the volume of Anxiolytics and Hypnotics used as the standardized, average daily quantities (ADQs) per 1000 patients. Based on the OpenPrescribing database, a random-effect model was applied to quantify the change in the level and trend of anxiolytics and hypnotics use after the UK national lockdown in March 2020. Practice characteristics extracted from the Fingertips data were assessed for their association with a reduction in medication use following the lockdown.ResultsThis study in the North East of England found that GP practices in higher health disparate regions had a lower workload than those in less health disparate areas, potentially due to disparities in healthcare utilization and socioeconomic status. Patients in the region reported higher levels of satisfaction with healthcare services compared to the England average, but there were differences between patients living in higher versus less health disparate areas. The study highlights the need for targeted interventions to address health disparities, particularly in higher health disparate areas. The study also found that psychiatric medication use was significantly more common in residents living in higher health disparate areas. Daily anxiolytics and hypnotics use decreased by 14 items per 1000 patients between the financial years 2019/20 and 2020/21. A further nine items per 1000 decreased for higher health disparate areas during the UK national lockdown.ConclusionsPeople during the COVID-19 lockdown were associated with an increased risk of unmet psychiatric medication demand, especially for higher health disparate areas that had low-socioeconomic status

    Relaxed Current Matching Requirements in Highly Luminescent Perovskite Tandem Solar Cells and Their Fundamental Efficiency Limits

    Perovskite-based tandem solar cells are of increasing interest as they approach commercialization. Here we use experimental parameters from optical spectroscopy measurements to calculate the limiting efficiency of perovskite–silicon and all-perovskite two-terminal tandems, employing currently available bandgap materials, as 42.0% and 40.8%, respectively. We show luminescence coupling between subcells (the optical transfer of photons from the high-bandgap to low-bandgap subcell) relaxes current matching when the high-bandgap subcell is a luminescent perovskite. We calculate that luminescence coupling becomes important at charge trapping rates (≤106 s–1) already being achieved in relevant halide perovskites. Luminescence coupling increases flexibility in subcell thicknesses and tolerance to different spectral conditions. For maximal benefit, the high-bandgap subcell should have the higher short-circuit current under average spectral conditions. This can be achieved by reducing the bandgap of the high-bandgap subcell, allowing wider, unstable bandgap compositions to be avoided.     Resurrection of an ancestral 5S rRNA

    <p>Abstract</p> <p>Background</p> <p>In addition to providing phylogenetic relationships, tree making procedures such as parsimony and maximum likelihood can make specific predictions of actual historical sequences. Resurrection of such sequences can be used to understand early events in evolution. In the case of RNA, the nature of parsimony is such that when applied to multiple RNA sequences it typically predicts ancestral sequences that satisfy the base pairing constraints associated with secondary structure. The case for such sequences being actual ancestors is greatly improved, if they can be shown to be biologically functional.</p> <p>Results</p> <p>A unique common ancestral sequence of 28 <it>Vibrio </it>5S ribosomal RNA sequences predicted by parsimony was resurrected and found to be functional in the context of the <it>E. coli </it>cellular environment. The functionality of various point variants and intermediates that were constructed as part of the resurrection were examined in detail. When separately introduced the changes at single stranded positions and individual double variants at base-paired positions were also viable. An additional double variant was examined at a different base-paired position and it was also valid.</p> <p>Conclusions</p> <p>The results show that at least in the case of the 5S rRNAs considered here, ancestors predicted by parsimony are likely to be realistic when the prediction is not overly influenced by single outliers. It is especially noteworthy that the phenotype of the predicted ancestors could be anticipated as a cumulative consequence of the phenotypes of the individual variants that comprised them. Thus, point mutation data is potentially useful in evaluating the reasonableness of ancestral sequences predicted by parsimony or other methods. The results also suggest that in the absence of significant tertiary structure constraints double variants that preserve pairing in stem regions will typically be accepted. Overall, the results suggest that it will be feasible to resurrect additional meaningful 5S rRNA ancestors as well as ancestral sequences of many different types of RNA.</p

    Alterations in vascular function in primary aldosteronism - a cardiovascular magnetic resonance imaging study

    Introduction: Excess aldosterone is associated with increased cardiovascular risk. Aldosterone has a permissive effect on vascular fibrosis. Cardiovascular magnetic resonance imaging (CMR) allows study of vascular function by measuring aortic distensibility. We compared aortic distensibility in primary aldosteronism (PA), essential hypertension (EH) and normal controls and explored the relationship between aortic distensibility and pulse wave velocity (PWV).&lt;p&gt;&lt;/p&gt; Methods: We studied PA (n=14) and EH (n=33) subjects and age-matched healthy controls (n=17) with CMR, including measurement of aortic distensibility, and measured PWV using applanation tonometry. At recruitment, PA and EH patients had similar blood pressure and left ventricular mass.&lt;p&gt;&lt;/p&gt; Results: Subjects with PA had significantly lower aortic distensibilty and higher PWV compared to EH and healthy controls. These changes were independent of other factors associated with reduced aortic distensibility, including aging. There was a significant relationship between increasing aortic stiffness and age in keeping with physical and vascular aging. As expected, aortic distensibility and PWV were closely correlated.&lt;p&gt;&lt;/p&gt; Conclusion: These results demonstrate that PA patients display increased arterial stiffness compared to EH, independent of vascular aging. The implication is that aldosterone invokes functional impairment of arterial function. The long-term implications of arterial stiffening in aldosterone excess require further study.&lt;p&gt;&lt;/p&gt

    Synthesis and characterisation of coating polyurethane cationomers containing fluorine built-in hard urethane segments

    Polyurethane cationomers were synthesised in the reaction of 4,4’-methylenebis(phenyl isocyanate) with polyoxyethylene glycol (M = 2,000) or poly(tetrafluoroethyleneoxide-co-difluoromethylene oxide) α,ω-diisocyanate and N-methyl diethanolamine. Amine segments were built-in to the urethane-isocyanate prepolymer in the reaction with 1-bromobutane or formic acid, and then they were converted to alkylammonium cations. The obtained isocyanate prepolymers were then extended in the aqueous medium that yielded stable aqueous dispersions which were applied on the surfaces of test poly(tetrafluoroethylene) plates. After evaporation of water, the dispersions formed thin polymer coatings. 1H, 13C NMR and IR spectral methods were employed to confirm chemical structures of synthesised cationomers. Based on 1H NMR and IR spectra, the factors κ and α were calculated, which represented the polarity level of the obtained cationomers. The DSC, wide angle X-ray scattering and atom force microscopy methods were employed for the microstructural assessment of the obtained materials. Changes were discussed in the surface free energy and its components, as calculated independently according to the method suggested by van Oss–Good, in relation to chemical and physical structures of cationomers as well as morphology of coating surfaces obtained from those cationomers. Fluorine incorporated into cationomers (about 30%) contributed to lower surface free energy values, down to about 15 mJ/m2. That was caused by gradual weakening of long-range interactions within which the highest share is taken by dispersion interactions

    ANALYSIS OF SPONTANEOUS IMBIBITION IN FRACTAL TREE-LIKE NETWORK SYSTEM

    Spontaneous imbibition in porous media is common in nature, imbibition potential is very important for understanding the imbibition ability, or the ability to keep high imbibition rate for a long time. Structure parameters have influence on imbibition potential. This work investigates the process of spontaneous imbibition of liquid into a fractal tree-like network, taking fractal structure parameters into consideration. The analytical expression for dimensionless imbibition rate with this fractal tree-like network is derived. The influence of structure parameters on imbibition potential is discussed. It is found that optimal diameter ratio beta is important for networks to have imbibition potential. Moreover, with liquid imbibed in more sub-branches, some structures of parameter combinations will show the characteristic of imbibition potential gradually. Finally, a parameter plane is made to visualize the percentage of good parameter in all possible combinations and to evaluate the imbibition potential of a specific network system more directly. It is also helpful to design and to optimize a fractal network with good imbibition potential

    Water imbibition of shale and its potential influence on shale gas recovery-a comparative study of marine and continental shale formations

    A large volume of fracturing fluid is pumped into a well to stimulate shale formation. The water is imbibed into the reservoir during this procedure. The effect of the imbibed water on gas recovery is still in debate. In this work, we study the spontaneous imbibition of water into marine shale samples from the Sichuan Basin and continental shale samples from Erdos Basin to explore the fluid imbibition characteristics and permeability change during water imbibition. Comparison of imbibition experiments shows that shale has stronger water imbibition and diffusion capacity than relatively higher permeability sandstone. Once the imbibition stops, water in shale has stronger ability to diffuse into deeper matrix, the water content in the main flow path decreases. Experiments in this study show that marine shale has stronger water imbibition capacity than continental shale. The permeability of continental shale decreases significantly with increasing imbibition water volume; however, the permeability of marine shale decreases at first and increases after a certain imbibition time. The induced fracture is obvious in the marine shale. SEM analysis shows that the relationship between the clay mineral and organic matter of continental shale is much more complex than that of marine shale, which may be the key factor restricting the water imbibition because the flow path is trapped by swelled clay minerals. Through this study, we concluded that whether gas recovery benefits from water imbibition depends on three aspects: 1) the diffusion ability of liquid into matrix; 2) the new cracks introduced by imbibed water; and 3) the formation sensibility.     A pilot randomised double blind controlled trial of the efficacy of purified fatty acids for the treatment of women with endometriosis-associated pain (PurFECT):study protocol

    Abstract Background Endometriosis affects 6–10% of women and is associated with debilitating pelvic pain. It costs the UK > £2.8 billion per year in loss of productivity. Endometriosis can be managed by surgical excision or medically by ovarian suppression. However, ~ 75% symptoms recur after surgery and available medical treatments have undesirable side effects and are contraceptive. Omega-3 purified fatty acids (PUFA) have been shown in animal models to reduce factors that are thought to lead to endometriosis-associated pain, have minimal side effects, and no effects on fertility. This paper presents a protocol for a two-arm, pilot parallel randomised controlled trial (RCT) which aims to inform the planning of a future multicentre trial to evaluate the efficacy of Omega-3 PUFA in the management of endometriosis-associated pain in women. Methods The study will recruit women with endometriosis over a 12-month period in the National Health Service (NHS) Lothian, UK, and randomise them to 8 weeks of treatment with Omega-3 PUFA or comparator (olive oil). The primary objective is to assess recruitment and retention rates. The secondary objectives are to determine the effectiveness/acceptability to participants of the proposed methods of recruitment/randomisation/treatments/questionnaires, to inform the sample size calculation and to refine the research methodology for a future large randomised controlled trial. Response to treatment will be monitored by pain scores and questionnaires assessing physical and emotional function compared at baseline and 8 weeks. Discussion We recognise that there may be potential difficulties in mounting a large randomised controlled trial for endometriosis to assess Omega-3 PUFA because they are a dietary supplement readily available over the counter and already used by women with endometriosis. We have therefore designed this pilot study to assess practical feasibility and following the ‘Initiative on Methods, Measurement, and Pain Assessment in Clinical Trials’ recommendations for the design of chronic pain trials.
