Effect of external beam radiation therapy versus transcatheter arterial chemoembolization for non-diffuse hepatocellular carcinoma (â„ 5Â cm): a multicenter experience over a ten-year period
BackgroundThe optimal local treatment for HCC with tumor diameter â„ 5Â cm is not well established. This research evaluated the effectiveness of external beam radiation therapy (EBRT) versus transcatheter arterial chemoembolization (TACE) for HCC with tumor diameter â„ 5Â cm.MethodsA total of 1210 HCC patients were enrolled in this study, including 302 and 908 patients that received EBRT and TACE, respectively. Propensity score matching (PSM) was used to identify patient pairs with similar baseline characteristics. Overall survival (OS) was the primary study endpoint.ResultsWe identified 428 patients using 1:1 PSM for survival comparison. Compared with the TACE group, the EBRT group had a significantly longer median OS (mOS) before (14.9 vs. 12.3 months, p = 0.0085) and after (16.8 vs. 11.4 months, p = 0.0026) matching. In the subgroup analysis, compared with the TACE group, the EBRT group had a significantly longer mOS for HCC with tumor diameters of 5-7Â cm (34.1 vs. 14.3 months, p = 0.04) and 7-10Â cm (34.4 vs. 10 months, p = 0.00065), whereas for HCC with tumor diameters â„ 10Â cm, no significant difference in mOS was observed (11.2 vs. 11.2 months, p = 0.83). In addition, the multivariable Cox analysis showed that Child-A, alkaline phosphatase < 125 U/L, and EBRT were independent prognostic indicators for longer survival.ConclusionEBRT is more effective than TACE as the primary local treatment for HCC with tumor diameter â„ 5Â cm, especially for HCC with tumor diameter of 5-10Â cm
Development and validation of an ultra?performance liquid chromatography quadrupole time of flight mass spectrometry method for rapid quantification of free amino acids in human urine
An ultra-performance liquid chromatography quadrupole time of flight mass spectrometry (UPLC-qTOFMS)method using hydrophilic interaction liquid chromatography was developed and validated for simultaneous quantification of 18 free amino acids in urine with a total acquisition time including the column re-equilibration of less than 18 min per sample. This method involves simple sample preparation steps which consisted of 15 times dilution with acetonitrile to give a final composition of 25 % aqueous and 75 % acetonitrile without the need of any derivatization. The dynamic range for our calibration curve is approximately two orders of magnitude (120-fold from the lowest calibration curve point) with good linearity (r2 ? 0.995 for all amino acids). Good separation of all amino acids as well as good intra- and inter-day accuracy (<15 %) and precision (<15 %) were observed using three quality control samples at a concentration of low, medium and high range of the calibration curve. The limits of detection (LOD) and lower limit of quantification of our method were ranging from approximately 1â300 nM and 0.01â0.5 ”M, respectively. The stability of amino acids in the prepared urine samples was found to be stable for 72 h at 4 °C, after one freeze thaw cycle and for up to 4 weeks at ?80 °C. We have applied this method to quantify the content of 18 free amino acids in 646 urine samples from a dietary intervention study. We were able to quantify all 18 free amino acids in these urine samples, if they were present at a level above the LOD. We found our method to be reproducible (accuracy and precision were typically <10 % for QCL, QCM and QCH) and the relatively high sample throughput nature of this method potentially makes it a suitable alternative for the analysis of urine samples in clinical setting

Evaluation of Virus-Free Chrysanthemum âHangjuâ Productivity and Response to Virus Reinfection in the Field: Molecular Insights into VirusâHost Interactions
The shoot apical meristem culture has been used widely to produce virus-free plantlets which have the advantages of strong disease resistance, high yield, and prosperous growth potential. However, this virus-free plant will be naturally reinfected in the field. The physiological and metabolic responses in the reinfected plant are still unknown. The flower of chrysanthemum âHangjuâ is a traditional medicine which is unique to China. In this study, we found that the virus-free âHangjuâ (VFH) was reinfected with chrysanthemum virus B/R in the field. However, the reinfected VFH (RVFH) exhibited an increased yield and medicinal components compared with virus-infected âHangjuâ (VIH). Comparative analysis of transcriptomes was performed to explore the molecular response mechanisms of the RVFH to CVB infection. A total of 6223 differentially expressed genes (DEGs) were identified in the RVFH vs. the VIH. KEGG enrichment and physiological analyses indicated that treatment with the virus-free technology significantly mitigated the plantsâ lipid and galactose metabolic stress responses in the RVFH. Furthermore, GO enrichment showed that plant viral diseases affected salicylic acid (SA)-related processes in the RVFH. Specifically, we found that phenylalanine ammonia-lyase (PAL) genes played a major role in defense-related SA biosynthesis in âHangjuâ. These findings provided new insights into the molecular mechanisms underlying plant virusâhost interactions and have implications for developing strategies to improve plant resistance against viruses
Influence of Crystallization Behavior of the Metal/Glass - Ceramic Composite Coating on the High Temperature Oxidation Resistance of the Coating
According to the harsh environment faced by the core thermal parts of aero - engines and the excellent high - temperature protection performance of the glass - ceramic coating, the flow - coating method was used to prepare nickel - containing B2O3 - Al2O3 - BaO - CeO2 metal/glass - ceramic composite coating (Ni/BABC) on the surface of the nickel - based alloy GH4586 for aero - engines. Differential thermal analysis (DTA), X - ray diffraction (XRD), scanning electron microscope/energy dispersive analysis (SEM/EDS), weight gain method and other test methods were used to study the effects of different nucleating agents and different heat treatment processes on the crystallization behavior of glass - ceramic with a constitution of B2O3 - Al2O3 - BaO - CeO2 and the effects of crystallization behavior on the high temperature oxidation resistance of the composite coating. Results showed that 4% (mass fraction) ZrO2 could achieve the best nucleation effect as a nucleating agent, and the precipitated crystal phase was BaAl2B2O7. The best crystallization process for the glass - ceramic was an annealing at 650 â for 30 h. The longer the annealing time, the higher the crystallization rate. Additionally, after 30 h, the crystallization rate tended to be stable at about 46%. The glass - ceramic composite coating had a significant protective effect against the high - temperature oxidation of the substrate. The coating sample with a crystallization rate of 46% had the lowest oxidation weight gain rate, gaining a weight of only 1.129 0 mg/cm2 after oxidation at 900 â for 30 h. Furthermore, it had the most uniform and dense structure after oxidation, its surface was smooth, and its high temperature oxidation resistance was excellent
<i>Nigrospora oryzae</i> Causing Leaf Spot Disease on <i>Chrysanthemum</i> Ă <i>morifolium</i> Ramat and Screening of Its Potential Antagonistic Bacteria
Chrysanthemum Ă morifolium Ramat. is a famous perennial herb with medicinal, edible, and ornamental purposes, but the occurrence of plant diseases can reduce its value. A serious disease that caused leaf spots in C. morifolium appeared in 2022 in Tongxiang City, Zhejiang Province, China. The C. morifolium leaves with brown spots were collected and used for pathogen isolation. By completing Kochâs postulates, it was proven that the isolate had pathogenicity to infect C. morifolium. It was determined that the pathogen isolated from chrysanthemum leaves was Nigrospora oryzae, through morphology and a multilocus sequence analysis method using a combination of the internal transcribed spacer gene (ITS), beta-tubulin gene (TUB2), and translation elongation factor 1-alpha gene (TEF1-α). This is the first report of C. morifolium disease caused by N. oryzae in the world. Through dual culture assay on PDA plates, 12 strains of bacteria with antagonistic effects were selected from 231 strains from the C. morifolium phyllosphere, among which Bacillus siamensis D65 had the best inhibitory effect on N. oryzae growth. In addition, the components of a strain D65 fermentation broth were profiled by SPME-GC-Q-TOF analysis, providing a foundation for further application and research of biological control
Double-Sided Nano-ZnO: Superior Antibacterial Properties and Induced Hepatotoxicity in Zebrafish Embryos
Zinc oxide nanoparticles (Nano-ZnO) have been widely used in the food, cosmetics, and biomedical fields due to their excellent antibacterial and antioxidant properties. However, with the widespread application of Nano-ZnO, Nano-ZnO inevitably enters the environment and living organisms, causing harm to human health and ecosystem safety. Therefore, the biosafety and toxicological issues of Nano-ZnO are gradually being emphasized. Our study found that Nano-ZnO has superior antibacterial properties compared to ofloxacin in the fight against Staphylococcus aureus (S. aureus). Given that ofloxacin can inhibit bacterial-induced inflammation, we constructed a model of bacterial inflammation using S. aureus in zebrafish. We found that Nano-ZnO inhibited the NF-ÎșB-mediated inflammatory signaling pathway. However, in the process, we found that Nano-ZnO caused hepatic steatosis in zebrafish. This suggested that Nano-ZnO had a certain hepatotoxicity, but did not affect liver development. Subsequently, we investigated the mechanism of hepatotoxicity produced by Nano-ZnO. Nano-ZnO triggered oxidative stress in the liver by generating ROS, which then induced endoplasmic reticulum stress to occur. It further activated srebp and its downstream genes fasn and acc1, which promoted the accumulation of fatty acid synthesis and the development of steatosis, leading to the development of nonalcoholic fatty liver disease (NAFLD). To address the hepatotoxicity of Nano-ZnO, we added carbon dots for the treatment of NAFLD. The carbon dots were found to normalize the steatotic liver. This provided a new strategy to address the hepatotoxicity caused by Nano-ZnO. In this work, we systematically analyzed the antibacterial advantages of Nano-ZnO in vivo and in vitro, explored the mechanism of Nano-ZnO hepatotoxicity, and proposed a new method to treat Nano-ZnO hepatotoxicity
