    Isolation and Characterization of 89K Pathogenicity Island-Positive ST-7 Strains of Streptococcus suis Serotype 2 from Healthy Pigs, Northeast China

    Streptococcus suis is a swine pathogen which can also cause severe infection, such as meningitis, and streptococcal-like toxic shock syndrome (STSS), in humans. In China, most of the S. suis infections in humans were reported in the southern areas with warm and humid climates, but little attention had been paid to the northern areas. Data presented here showed that the virulent serotypes 1, 2, 7, and 9 of S. suis could be steadily isolated from the healthy pigs in the pig farms in all the three provinces of Northeast China. Notably, a majority of the serotype 2 isolates belonged to the 89K pathogenicity island-positive ST-7 clone that had historically caused the human STSS outbreaks in the Sichuan and Jiangsu provinces of China, although the human STSS case caused by S. suis had never been reported in northern areas of China. Data presented here indicated that the survey of S. suis should be expanded to or reinforced in the northern areas of China

    Tensile, Creep, and Fatigue Behaviors of 3D-Printed Acrylonitrile Butadiene Styrene

    Acrylonitrile butadiene styrene (ABS) is a widely used thermoplastics in 3D printing. However, there is a lack of thorough investigation of the mechanical properties of 3D-printed ABS components, including orientation-dependent tensile strength and creep fatigue properties. In this work, a systematic characterization is conducted on the mechanical properties of 3D-printed ABS components. Specifically, the effect of printing orientation on the tensile and creep properties is investigated. The results show that, in tensile tests, the 0° printing orientation has the highest Young’s modulus of 1.81 GPa, and ultimate strength of 224 MPa. In the creep test, the 90° printing orientation has the lowest k value of 0.2 in the plastics creep model, suggesting 90° is the most creep resistant direction. In the fatigue test, the average cycle number under load of 30 N is 3796 cycles. The average cycle number decreases to 128 cycles when the load is 60 N.     Two Rapid Power Iterative DOA Estimators for UAV Emitter Using Massive/Ultra-massive Receive Array

    To provide rapid direction finding (DF) for unmanned aerial vehicle (UAV) emitter in future wireless networks, a low-complexity direction of arrival (DOA) estimation architecture for massive multiple input multiple output (MIMO) receiver arrays is constructed. In this paper, we propose two strategies to address the extremely high complexity caused by eigenvalue decomposition of the received signal covariance matrix. Firstly, a rapid power-iterative rotational invariance (RPI-RI) method is proposed, which adopts the signal subspace generated by power iteration to gets the final direction estimation through rotational invariance between subarrays. RPI-RI makes a significant complexity reduction at the cost of a substantial performance loss. In order to further reduce the complexity and provide a good directional measurement result, a rapid power-iterative Polynomial rooting (RPI-PR) method is proposed, which utilizes the noise subspace combined with polynomial solution method to get the optimal direction estimation. In addition, the influence of initial vector selection on convergence in the power iteration is analyzed, especially when the initial vector is orthogonal to the incident wave. Simulation results show that the two proposed methods outperform the conventional DOA estimation methods in terms of computational complexity. In particular, the RPIPR method achieves more than two orders of magnitude lower complexity than conventional methods and achieves performance close to CRLB. Moreover, it is verified that the initial vector and the relative error have a significant impact on the performance of the computational complexity

    Analysis of the vp2 gene sequence of a new mutated mink enteritis parvovirus strain in PR China

    <p>Abstract</p> <p>Background</p> <p>Mink enteritis virus (MEV) causes a highly contagious viral disease of mink with a worldwide distribution. MEV has a linear, single-stranded, negative-sense DNA with a genome length of approximately 5,000 bp. The VP2 protein is the major structural protein of the parvovirus encoded by the <it>vp</it>2 gene. VP2 is highly antigenic and plays important roles in determining viral host ranges and tissue tropisms. This study describes the bionomics and <it>vp</it>2 gene analysis of a mutated strain, MEV-DL, which was isolated recently in China and outlines its homologous relationships with other selected strains registered in Genbank.</p> <p>Results</p> <p>The MEV-DL strain can infect F81 cells with cytopathic effects. Pig erythrocytes were agglutinated by the MEV-DL strain. The generation of MEV-DL in F81 cells could infect mink within three months and cause a disease that was similar to that caused by wild-type MEV. A comparative analysis of the <it>vp</it>2 gene nucleotide (nt) sequence of MEV-DL showed that this was more than 99% homologous with other mink enteritis parvoviruses in Genbank. However, the nucleotide residues at positions 1,065 and 1,238 in the MEV-DL strain of the <it>vp</it>2 gene differed from those of all the other MEV strains described previously. It is noteworthy that the mutation at the nucleotide residues position 1,238 led to Asp/Gly replacement. This may lead to structural changes. A phylogenetic tree and sequence distance table were obtained, which showed that the MEV-DL and ZYL-1 strains had the closest inheritance distance.</p> <p>Conclusions</p> <p>A new variation of the <it>vp</it>2 gene exists in the MEV-DL strain, which may lead to structural changes of the VP2 protein. Phylogenetic analysis showed that MEV-DL may originate from the ZYL-1 strain in DaLian.</p

    Enhanced Stem Cell Osteogenic Differentiation by Bioactive Glass Functionalized Graphene Oxide Substrates

    An unmet need in engineered bone regeneration is to develop scaffolds capable of manipulating stem cells osteogenesis. Graphene oxide (GO) has been widely used as a biomaterial for various biomedical applications. However, it remains challenging to functionalize GO as ideal platform for specifically directing stem cell osteogenesis. Herein, we report facile functionalization of GO with dopamine and subsequent bioactive glass (BG) to enhance stem cell adhesion, spreading, and osteogenic differentiation. On the basis of graphene, we obtained dopamine functionalized graphene oxide/bioactive glass (DGO/BG) hybrid scaffolds containing different content of DGO by loading BG nanoparticles on graphene oxide surface using sol-gel method. To enhance the dispersion stability and facilitate subsequent nucleation of BG in GO, firstly, dopamine (DA) was used to modify GO. Then, the modified GO was functionalized with bioactive glass (BG) using sol-gel method. The adhesion, spreading, and osteoinductive effects of DGO/BG scaffold on rat bone marrow mesenchymal stem cells (rBMSCs) were evaluated. DGO/BG hybrid scaffolds with different content of DGO could influence rBMSCs’ behavior. The highest expression level of osteogenic markers suggests that the DGO/BG hybrid scaffolds have great potential or elicit desired bone reparative outcome

    Perturbation of Thymocyte Development Underlies the PRRS Pandemic: A Testable Hypothesis

    Porcine reproductive and respiratory syndrome virus (PRRSV) causes immune dysregulation during the Critical Window of Immunological Development. We hypothesize that thymocyte development is altered by infected thymic antigen presenting cells (TAPCs) in the fetal/neonatal thymus that interact with double-positive thymocytes causing an acute deficiency of T cells that produces “holes” in the T cell repertoire allowing for poor recognition of PRRSV and other neonatal pathogens. The deficiency may be the result of random elimination of PRRSV-specific T cells or the generation of T cells that accept PRRSV epitopes as self-antigens. Loss of helper T cells for virus neutralizing (VN) epitopes can result in the failure of selection for B cells in lymph node germinal centers capable of producing high affinity VN antibodies. Generation of cytotoxic and regulatory T cells may also be impaired. Similar to infections with LDV, LCMV, MCMV, HIV-1 and trypanosomes, the host responds to the deficiency of pathogen-specific T cells and perhaps regulatory T cells, by “last ditch” polyclonal B cell activation. In colostrum-deprived PRRSV-infected isolator piglets, this results in hypergammaglobulinemia, which we believe to be a “red herring” that detracts attention from the thymic atrophy story, but leads to our second independent hypothesis. Since hypergammaglobulinemia has not been reported in PRRSV-infected conventionally-reared piglets, we hypothesize that this is due to the down-regulatory effect of passive maternal IgG and cytokines in porcine colostrum, especially TGFβ which stimulates development of regulatory T cells (Tregs)

    Secondary infection with Streptococcus suis serotype 7 increases the virulence of highly pathogenic porcine reproductive and respiratory syndrome virus in pigs

    <p>Abstract</p> <p>Background</p> <p>Porcine reproductive and respiratory syndrome virus (PRRSV) and <it>Streptococcus suis </it>are common pathogens in pigs. In samples collected during the porcine high fever syndrome (PHFS) outbreak in many parts of China, PRRSV and <it>S. suis </it>serotype 7 (SS7) have always been isolated together. To determine whether PRRSV-SS7 coinfection was the cause of the PHFS outbreak, we evaluated the pathogenicity of PRRSV and/or SS7 in a pig model of single and mixed infection.</p> <p>Results</p> <p>Respiratory disease, diarrhea, and anorexia were observed in all infected pigs. Signs of central nervous system (CNS) disease were observed in the highly pathogenic PRRSV (HP-PRRSV)-infected pigs (4/12) and the coinfected pigs (8/10); however, the symptoms of the coinfected pigs were clearly more severe than those of the HP-PRRSV-infected pigs. The mortality rate was significantly higher in the coinfected pigs (8/10) than in the HP-PRRSV- (2/12) and SS7-infected pigs (0/10). The deceased pigs of the coinfected group had symptoms typical of PHFS, such as high fever, anorexia, and red coloration of the ears and the body. The isolation rates of HP-PRRSV and SS7 were higher and the lesion severity was greater in the coinfected pigs than in monoinfected pigs.</p> <p>Conclusion</p> <p>HP-PRRSV infection increased susceptibility to SS7 infection, and coinfection of HP-PRRSV with SS7 significantly increased the pathogenicity of SS7 to pigs.</p

    Marketing is an important aspect in running fishing business because it is an economic activity that influences the fluctuation of fishermen\u27s income. The production can be useless if the price is low, thus, marketing has to be good and efficient. This research is about the efficiency of the marketing system of fresh fish layang (Decapterus russeli) in the fish auction place. Specifically, this research is to know : 1) the marketing system of fresh fish layang in Tegal City. 2) the marketing margin of fresh fish layang in Tegal City. 3) the distribution flow of fresh fish layang in Tegal City. 4) the reason fishermen sell their products in the Fish Auction Place. The method use in this research is descriptive analysis method, The registration data and literature study. Based on the analysis, it is known that the marketing system of fresh in the Fish Auction Place, seen from the marketing cost calculation, purchasing price, selling price and profit is < 1, which means efficient. And if seen from the marketing margin, the most efficient flow is channel 4 (the 4th channel). Meanwhile, the reason why fishermen sell their Fish Auction Place is because of the guarantee that their product will be sold. Fish is a product that is easily broken and rotten. Therefore, the guarantee that the product will be sold, can minimize loss risk for fisherman
