    Hubungan IL-10 Dengan Serum Kreatinin Dan Terjadinya Komplikasi Pada Preeklampsia Perawatan Konservatif

    Tujuan: mengetahui hubungan penurunan IL-10 dengan terjadinya peningkatan serum kreatinin (SK) dan terjadinya komplikasi preeklampsia.Bahan dan Metode: penelitian ini merupakan analitik observasional yang dilakukan pada 30 wanita preeklampsia berat tipe dini yang dilakukan perawatan konservatif. Dilakukan pemeriksaan IL-10 pada serum darah dengan metode ELISA sebelum perawatan konservatif dan kemudian dinilai luaran maternal (SK dan komplikasi preeklampsia)Hasil: didapatkan rerata kadar IL-10 pada preeklampsia tipe dini 0,71 ± 0,66 pg/mL, rerata luaran SK 0,83 ± 0,29, terjadinya komplikasi 9 kasus (edema paru, impending eklampsia, sindroma HELLP). Tidak didapatkan hubungan antara IL-10 dengan peningkatan serum kreatinin (p = 0,483) dan komplikasi preeklampsia (p = 0,828).Simpulan: IL-10 bukan merupakan faktor prediktif untuk luaran maternal pada preeklampsia perawatan konservatif

    Short-term clinical outcomes and five-year survival analysis of laparoscopic-assisted transanal natural orifice specimen extraction versus conventional laparoscopic surgery for sigmoid and rectal cancer: a single-center retrospective study

    BackgroundThe cosmetic benefits of natural orifice specimen extraction (NOSE) are easily noticeable, but its principles of aseptic and tumor-free procedure have caused controversy.MethodsWe conducted a retrospective analysis of the clinical data of patients who underwent laparoscopic-assisted transanal NOSE or conventional laparoscopic surgery (CLS) for sigmoid and rectal cancer at our hospital between January 2018 and December 2018. The study aimed to compare the general characteristics, perioperative indicators, postoperative complications, and five-year follow-up results between the two groups.ResultsA total of 121 eligible patients were enrolled, with 52 underwent laparoscopic-assisted transanal NOSE and 69 underwent CLS. There were no significant differences observed between the two groups in terms of gender, age, body mass index (BMI), TNM stage, etc. (P > 0.05). However, the NOSE group exhibited significantly shorter total incision length and longer operation time compared to the CLS group (P < 0.05). There were no statistically significant differences observed between the two groups in terms of positive rate of bacterial culture, incidence rates of intraabdominal infections or anastomotic leakage (P > 0.05). Furthermore, during follow-up period there was no statistically significant difference observed between these two groups concerning overall survival rate and disease-free survival outcomes (P > 0.05).ConclusionsThe management of surgical complications in CLS is exemplary, with NOSE presenting a sole advantage in terms of incision length albeit at the cost of prolonged operative time. Therefore, NOSE may be deemed appropriate for patients who place high emphasis on postoperative cosmetic outcomes

    Identification of LncRNA Linc00513 Containing Lupus-Associated Genetic Variants as a Novel Regulator of Interferon Signaling Pathway

    Systemic lupus erythematosus (SLE) is a complex autoimmune disease characterized by augmented type I interferon signaling. High-throughput technologies have identified plenty of SLE susceptibility single-nucleotide polymorphisms (SNPs) yet the exact roles of most of them are still unknown. Functional studies are principally focused on SNPs in the coding regions, with limited attention paid to the SNPs in non-coding regions. Long non-coding RNAs (lncRNAs) are important players in shaping the immune response and show relationship to autoimmune diseases. In order to reveal the role of SNPs located near SLE related lncRNAs, we performed a transcriptome profiling of SLE patients and identified linc00513 as a significantly over expressed lncRNA containing functional SLE susceptibility loci in the promoter region. The risk-associated G allele of rs205764 and A allele of rs547311 enhanced linc00513 promoter activity and related to increased expression of linc00513 in SLE. We also identified linc00513 to be a novel positive regulator of type I interferon pathway by promoting the phosphorylation of STAT1 and STAT2. Elevated linc00513 expression positively correlated with IFN score in SLE patients. Linc00513 expression was higher in active disease patients than those inactive ones. In conclusion, our data identify two functional promoter variants of linc00513 that contribute to increased level of linc00513 and confer susceptibility on SLE. The study provides new insights into the genetics of SLE and extends the role of lncRNAs in the pathogenesis of SLE

    Identification of Renal Long Non-coding RNA RP11-2B6.2 as a Positive Regulator of Type I Interferon Signaling Pathway in Lupus Nephritis

    Objective: Lupus nephritis (LN) is one of the most serious complications of systemic lupus erythematosus (SLE). Type I interferon (IFN-I) is associated with the pathogenesis of LN. Long non-coding RNAs (lncRNAs) have been implicated in the pathogenesis of SLE, however, the roles of lncRNAs in LN are still poorly understood. Here, we identified and investigated the function of LN-associated lncRNA RP11-2B6.2 in regulating IFN-I signaling pathway.Methods: RNA sequencing was used to analyze the expression of lncRNAs in kidney biopsies from LN patients and controls. Antisense oligonucleotides and CRISPRi system or overexpression plasmids and CRISPRa system were used to perform loss or gain of function experiments. In situ hybridization, imaging flow cytometry, dual-luciferase reporter assay, and ATAC sequencing were used to study the functions of lncRNA RP11-2B6.2. RT-qPCR, ELISA, and western blotting were done to detect RNA and protein levels of specific genes.Results: Elevated lncRNA RP11-2B6.2 was observed in kidney biopsies from LN patients and positively correlated with disease activity and IFN scores. Knockdown of lncRNA RP11-2B6.2 in renal cells inhibited the expression of IFN stimulated genes (ISGs), while overexpression of lncRNA RP11-2B6.2 enhanced ISG expression. Knockdown of LncRNA RP11-2B6.2 inhibited the phosphorylation of JAK1, TYK2, and STAT1 in IFN-I pathway, while promoted the chromatin accessibility and the transcription of SOCS1.Conclusion: The expression of lncRNAs is abnormal in the kidney of LN. LncRNA RP11-2B6.2 is a novel positive regulator of IFN-I pathway through epigenetic inhibition of SOCS1, which provides a new therapeutic target to alleviate over-activated IFN-I signaling in LN

    Re-examination of the effect of ownership structure on financial reporting: Evidence from share pledges in China

    In this paper, we present evidence that firms with concentrated ownership manage earnings when their large shareholders have an incentive to do so. The large shareholders of Chinese public firms often pledge their shares for loans. Before the split share reform in 2006, loan terms were based on the book value of the firm. Since then, the share price has become critical for share pledged loans. We postulate that the reform triggered large shareholders’ incentive to influence financial reports. Using a sample of non-state-owned enterprises, we test the effect of share pledges on earnings smoothing and how this effect changes after the reform. Our results suggest that share pledging firms smooth their earnings more than other firms, but these results are only found after the split share reform. Accordingly, our results provide more direct evidence on the effect of ownership concentration on financial reporting

    Aiming at the problem of serious shutdowns of conveyors caused by abnormal coal flow of scraper conveyors, a coal flow monitoring method based on speckle structured light is proposed. The point cloud data of the coal body on the scraper conveyor is collected through the speckle structured light acquisition system. Then, the proposed PDS-Algorithm (Planar Density Simplification Algorithm) is used to complete the simplification and differentiation of the collected point cloud data, which provides a basis for constructing geometric characteristics of coal flow lineament. This paper uses the processed point cloud data to calculate the volume of the coal mass and monitor the coal flow of the scraper conveyor. Finally, this method is used in the detection of abnormal coal flow of a coal mine scraper conveyor, and the results show that the proposed abnormal flow monitoring method can meet the accuracy and real-time requirements of coal mine abnormal alarms

    Preliminary characterization of exopolysaccharides produced by Abortiporus biennis in submerged fermentation

    Abortiporus biennis is a rare edible and medicinal mushroom that is valuable in traditional Chinese medicine. Exopolysaccharides (EPSs) as valuable metabolites have been harvested from fermentation broths of A. biennis. However, studies on preliminary characterisations and bioactivities of A. biennis EPSs are lacking. In this study, therefore, the production, characteristics and bioactivity of EPSs produced by A. biennis in submerged fermentation were investigated. An EPS-1 was isolated and purified from the culture broth of A. biennis using a Sepharose CL-6B column chromatography. The molecular weight, monosaccharide composition, structural characteristics, chain conformation and thermal property of EPS-1 were determined by gel-filtration chromatography, gas chromatography-mass spectroscopy, Fourier-transform infrared spectroscopy, Congo red test and thermogravimetric analysis, respectively. The in vitro antioxidant and immunoregulation activities of the EPS-1 were evaluated by adopting DPPH and hydroxyl radical-scavenging assays, and analysing NO production in macrophage RAW 264.7 cells, respectively. Results showed that the culture medium containing 30 g/L lactose and 3.0 g/L tryptone had the most suitable carbon and nitrogen sources, respectively, for EPS production in A. biennis. The maximum EPS production (18.29 g/L) was obtained after 6 days of cultivation in a 5 L stirred-tank reactor under the most suitable culture medium. After purification, EPS-1 with 85.4% yield and 96.1% carbohydrate content was obtained from the culture broth of A. biennis. EPS-1 was characterised as a neutral heteropolysaccharide with a molecular weight of 22.07 kDa and is composed of glucose, mannose and galactose at a molar ratio of 3.3:2.0:1.0. EPS-1 with a relatively high thermal stability existed as a random coil conformation in the aqueous medium and possessed prominent radical-scavenging ability and macrophage stimulation activity in vitro. Therefore, EPS-1 could be explored as a functional ingredient with potential applications in food, medical and cosmetic industries
