    The mechanisms of NLRP3 inflammasome/pyroptosis activation and their role in diabetic retinopathy

    In the working-age population worldwide, diabetic retinopathy (DR), a prevalent complication of diabetes, is the main cause of vision impairment. Chronic low-grade inflammation plays an essential role in DR development. Recently, concerning the pathogenesis of DR, the Nod-Like Receptor Family Pyrin Domain Containing 3 (NLRP3) inflammasome in retinal cells has been determined as a causal factor. In the diabetic eye, the NLRP3 inflammasome is activated by several pathways (such as ROS and ATP). The activation of NPRP3 leads to the secretion of inflammatory cytokines interleukin-1Ī² (IL-1Ī²) and interleukin-18 (IL-18), and leads to pyroptosis, a rapid inflammatory form of lytic programmed cell death (PCD). Cells that undergo pyroptosis swell and rapture, releasing more inflammatory factors and accelerating DR progression. This review focuses on the mechanisms that activate NLRP3 inflammasome and pyroptosis leading to DR. The present research highlighted some inhibitors of NLRP3/pyroptosis pathways and novel therapeutic measures concerning DR treatment

    COMPATIBILITY EVALUATION OF BZ25-1 CRUDE OILS IN BOHAI BAY, CHINA

    ABSTRACT BZ25-1 oilfield is located in the southeast of Bohai bay which geographically lies between 119 o 00ā€²to 119 o 15ā€²east longitude and 38 o 10ā€²to 38 o 20ā€²north latitude. It has two oil blocks, including Shahejie (SHJ) waxy oil and Minghuazhen (MHZ) heavy oil, with six wellhead platforms WHPA~WHPF and six submarine pipelines. Therein, the WHPC-WHPB and WHPB-SPM (Single Point Mooring) pipelines transport the mixture of the two produced crude oils. However, the mixing of the two oils will certainly bring out a change in their components and properties, which directly affects the safe operation of the submarine pipelines and offshore production facilities. Therefore, this paper compounds three kinds of MHZ/SHJ mixed oils with blending ratios of 1:1, 3:1 and 9:1, mainly studies how the components, rheological and thermophysical properties of the oil mixtures change with the blending ratio. The major objective of this study is to evaluate the compatibility of the two crude oils and provide a theoretical basis for the production optimization and risk elusion of the oilfield. The results of the study show that the components and properties of SHJ crude oil are quite different from those of MHZ oil, the flow behavior of SHJ oil is more sensitive to temperature. As MHZ oil in the compounds increases, the contents of asphaltene, resin, sulfur and carbon residue will increase except wax contents, their viscosities, densities and flash points will also increase, but their pour points, yield stresses, calorific values and other major thermophysical parameters will decrease. A blending ratio of 2~7:1 for MHZ to SHJ crude oil can be concluded to make the properties of the compounds meet the safe and economic requirements of the subsea pipeline and offshore facility operations and ensure the compatibility of the mixed oils. In actuality, the field operations have confirmed that the recommended blending ratio is reasonable and practicable

    Self-leadership as an attribute of service leadership: Its relationship to well-being among university students in Hong Kong

    IntroductionIn the scientific literature, although conceptual models and empirical evidence have shown that leadership attributes are intimately linked to the well-being of followers, there is a lack of studies focusing on leadership in the service economy. According to the Service Leadership Theory, service leadership is a process that satisfies the needs of self, others, and systems (teams, organizations, communities, and societies) in ethical ways that is characterized by leadership competence, character, and care. With specific reference to self-leadership emphasized in service leadership, higher levels of service leadership attributes should promote personal well-being. However, the relationships between ā€œservice leadership attributesā€ and ā€œwell-beingā€ in leaders at the intrapersonal level in leadership education among Chinese university students are rarely examined.MethodsIn this study, we collected data from 198 students to understand the linkages between ā€œservice leadership attributesā€ and ā€œwell-beingā€ in university students taking a course on service leadership. For tracking changes in students, we collected both pretest and posttest data on validated measures of ā€œservice leadership attributesā€ (i.e., ā€œknowledge,ā€ ā€œattitude,ā€ and ā€œbehaviorā€) and ā€œwell-beingā€ (i.e., ā€œpositive youth development attributesā€ and ā€œlife satisfactionā€).ResultsResults showed that the posttest scores on all three domains of ā€œservice leadership attributesā€ as well as two dimensions of ā€œwell-beingā€ encompassing life satisfaction and positive youth development attributes were higher than the respective pretest scores, suggesting that students experienced a shift in a positive direction after taking the course. Cross-lagged analyses showed that pretest service leadership attitude and behavior predicted posttest positive youth development attributes; pretest service leadership behavior predicted posttest life satisfaction. Pretest life satisfaction also predicted posttest service leadership behavior.DiscussionFindings suggest that there is an intimate relationship between ā€œservice leadership attributesā€ and ā€œwell-beingā€ in the ā€œpre-workā€ context among university students

    Does Exposure to Foreign Culture Influence Creativity? Maybe It's Not Only Due to Concept Expansion

    Multicultural experience refers to those experiences gained through individuals' contact with other cultures. This study focused on exploring whether knowledge of different cultures can improve creative performance-and also how multicultural experiences influenced this performance through changes in individual's physiological mechanisms. Study 1 explored the influence of different cultural priming on creative story-writing tasks. Eighty-nine Chinese college students were randomly assigned to 4 conditions: sole American culture, dual cultures, sole Chinese culture or control condition, and made to watch 45 min slides with cultural elementsā€”including pictures, music and videos,ā€”and then they were asked to complete the creative story-writing task. The results showed that American culture priming group's score was significantly higher than the control condition with regards to the uniqueness and novelty of the creative story-writing task. Study 2 was aimed at exploring the relationship between physiological arousal levels induced by different cultural and creative performance. We divided the whole experiment into five stages,ā€”including the baseline, picture, listening to music, watching video, and completing creative tasks. Through Biofeedback measurement, we recorded the physiological indexes of participants in different groups in every stage, including skin conductance, thermal, electroencephalographic, and heart rate. The results showed that contacting with foreign cultures would increase individuals' physiological arousal level and brain activity, which contributed to the following creative task

    Evaluating the role of serum uric acid in the risk stratification and therapeutic response of patients with pulmonary arterial hypertension associated with congenital heart disease (PAH-CHD)

    Background: Pulmonary arterial hypertension (PAH) is a malignant pulmonary vascular disease that negatively impacts quality of life, exercise capacity, and mortality. This study sought to investigate the relationship between serum uric acid (UA) level and the disease severity and treatment response of patients with PAH and congenital heart disease (PAH-CHD).Methods: This study included 225 CHD patients and 40 healthy subjects. Serum UA was measured in all patients, and UA levels and haemodynamic parameters were re-evaluated in 20 patients who had received PAH-specific drug treatment for at least 7 Ā± 1Ā month.Results: Serum UA levels were significantly higher in PAH-CHD patients than in CHD patients with a normal pulmonary artery pressure and normal subjects (347.7 Ā± 105.7Ā Ī¼mol/L vs. 278.3 Ā± 84.6Ā Ī¼mol/L; 347.7 Ā± 105.7Ā Ī¼mol/L vs. 255.7 Ā± 44.5Ā Ī¼mol/L, p < 0.05). UA levels in the intermediate and high risk groups were significantly higher than those in the low-risk group (365.6 Ā± 107.8Ā Ī¼mol/L vs. 311.2 Ā± 82.8Ā Ī¼mol/L; 451.6 Ā± 117.6Ā Ī¼mol/L vs. 311.2 Ā± 82.8Ā Ī¼mol/L, p < 0.05). Serum UA levels positively correlated with mean pulmonary arterial pressure, WHO functional class, pulmonary vascular resistance, and NT-proBNP (r = 0.343, 0.357, 0.406, 0.398; p < 0.001), and negatively with mixed venous oxygen saturation (SvO2) and arterial oxygen saturation (SaO2) (r = āˆ’0.293, āˆ’0.329; p < 0.001). UA significantly decreased from 352.7 Ā± 97.5 to 294.4 Ā± 56.8Ā Ī¼mol/L (p = 0.001) after PAH-specific drug treatment for at least 6Ā months, along with significant decreases in mean pulmonary arterial pressure and pulmonary vascular resistance and increases in cardiac index and mixed SvO2.Conclusion: Serum UA can be used as a practical and economic biomarker for risk stratification and the evaluation of PAH-specific drug treatment effects for patients with PAH-CHD

    Optimizing interplanar spacing, oxygen vacancies and micromorphology via lithium-ion pre-insertion into ammonium vanadate nanosheets for advanced cathodes in aqueous zinc-ion batteries

    Ammonium vanadates, featuring an Nā”€HĀ·Ā·Ā·O hydrogen bond network structure between NH4+ and Vā”€O layers, have become popular cathode materials for aqueous zinc-ion batteries (AZIBs). Their appeal lies in their multi-electron transfer, high specific capacity, and facile synthesis. However, a major drawback arises as Zn2+ ions tend to form bonds with electronegative oxygen atoms between Vā”€O layers during cycling, leading to irreversible structural collapse. Herein, Li+ pre-insertion into the intermediate layer of NH4V4O10 is proposed to enhance the electrochemical activity of ammonium vanadate cathodes for AZIBs, which extends the interlayer distance of NH4V4O10 to 9.8 ƅ and offers large interlaminar channels for Zn2+ (de)intercalation. Moreover, Li+ intercalation weakens the crystallinity, transforms the micromorphology from non-nanostructured strips to ultrathin nanosheets, and increases the level of oxygen defects, thus exposing more active sites for ion and electron transport, facilitating electrolyte penetration, and improving electrochemical kinetics of electrode. In addition, the introduction of Li+ significantly reduces the bandgap by 0.18 eV, enhancing electron transfer in redox reactions. Leveraging these unique advantages, the Li+ pre-intercalated NH4V4O10 cathode exhibits a high reversible capacity of 486.1 mAh gāˆ’1 at 0.5 A gāˆ’1 and an impressive capacity retention rate of 72% after 5,000 cycles at 5 A gāˆ’1

    Foxm1 transcription factor is required for maintenance of pluripotency of P19 embryonal carcinoma cells

    Transcription factor Foxm1 plays a critical role during embryonic development and its expression is repressed during retinoic acid (RA)-induced differentiation of pluripotent P19 embryonal carcinoma cells at the early stage, correlated with downregulation of expression of pluripotency markers. To study whether Foxm1 participates in the maintenance of pluripotency of stem cells, we knock down Foxm1 expression in P19 cells and identify that Oct4 are regulated directly by Foxm1. Knockdown of Foxm1 also results in spontaneous differentiation of P19 cells to mesodermal derivatives, such as muscle and adipose tissues. Maintaining Foxm1 expression prevents the downregulation of pluripotency-related transcription factors such as Oct4 and Nanog during P19 cell differentiation. Furthermore, overexpression of FOXM1 alone in RA-differentiated P19 cells (4 days) or human newborn fibroblasts restarts the expression of pluripotent genes Oct4, Nanog and Sox2. Together, our results suggest a critical involvement of Foxm1 in maintenance of stem cell pluripotency
