    A novel culture system for modulating single cell geometry in 3D

    Dedifferentiation of chondrocytes during in vitro expansion remains an unsolved challenge for repairing serious articular cartilage defects. In this study, a novel culture system was developed to modulate single cell geometry in 3D and investigate its effects on the chondrocyte phenotype. The approach uses 2D micropatterns followed by in situ hydrogel formation to constrain single cell shape and spreading. This enables independent control of cell geometry and extracellular matrix. Using collagen I matrix, we demonstrated the formation of a biomimetic collagenous “basket” enveloping individual chondrocytes cells. By quantitatively monitoring the production by single cells of chondrogenic matrix (e.g. collagen II and aggrecan) during 21-day cultures, we found that if the cell’s volume decreases, then so does its cell resistance to dedifferentiation (even if the cells remain spherical). Conversely, if the volume of spherical cells remains constant (after an initial decrease), then not only do the cells retain their differentiated status, but previously de-differentiated redifferentiate and regain a chondrocyte phenotype. The approach described here can be readily applied to pluripotent cells, offering a versatile platform in the search for niches toward either self-renewal or targeted differentiation

    Paclitaxel liposomes exert radio-sensitization effect on breast cancer cell line SK-BR-3 by regulating expressions of apoptotic proteins

    Purpose: To study the radio-sanitization effect of paclitaxel liposomes on breast cancer cells, SK-BR-3.Methods: Breast cancer cell line SK-BR-3 was cultured and made into a cell suspension. Four groups of cells were used: control and radiotherapy groups, paclitaxel group, and paclitaxel liposome + radiotherapy group (combination group). The growth inhibitory effects of the different treatments on SKBR-3 cells were determined with CCK-8 method. Apoptosis in each group was evaluated with flow cytometry, while Western blotting was employed to assay Bcl-2, Caspase-3 and Bax protein levels.Results: There were marked inhibitory effect on growth of SK-BR-3 cells in drug, radiotherapy and combination groups, relative to control, while apoptosis was greater in combination group than in drug and radiotherapy groups (p < 0.05). The Bcl-2 protein level was higher in radiotherapy, drug and combination groups than in control group, while Caspase-3 and Bax proteins were markedly higher thancontrol values (p < 0.05).Conclusion: Paclitaxel liposomes exert radio-sensitization effect on SK-BR-3 by regulating the levels of apoptotic proteins.     Single-cell microfluidics to study the effects of genome deletion on bacterial growth behavior

    By directly monitoring single cell growth in a microfluidic platform, we interrogated genome-deletion effects in Escherichia coli strains. We compared the growth dynamics of a wild type strain with a clean genome strain, and their derived mutants at the single-cell level. A decreased average growth rate and extended average lag time were found for the clean genome strain, compared to those of the wild type strain. Direct correlation between the growth rate and lag time of individual cells showed that the clean genome population was more heterogeneous. Cell culturability (the ratio of growing cells to the sum of growing and nongrowing cells) of the clean genome population was also lower. Interestingly, after the random mutations induced by a glucose starvation treatment, for the clean genome population mutants that had survived the competition of chemostat culture, each parameter markedly improved (i.e., the average growth rate and cell culturability increased, and the lag time and heterogeneity decreased). However, this effect was not seen in the wild type strain; the wild type mutants cultured in a chemostat retained a high diversity of growth phenotypes. These results suggest that quasi-essential genes that were deleted in the clean genome might be required to retain a diversity of growth characteristics at the individual cell level under environmental stress. These observations highlight that single-cell microfluidics can reveal subtle individual cellular responses, enabling in-depth understanding of the population

    A Compact Monopole Antenna With Filtering Response for WLAN Applications

    A novel compact monopole antenna with filtering response for WLAN applications is presented in this paper. The antenna is composed of a capacity-loaded matching patch, two resonators, and two end-coupled monopoles. The resonators consist of wide rectangular patches and narrow shorted lines to form the resonance, and the resonators are printed on the different layers to increase the design flexibility. Two meandering monopoles are located at the edges of the top layer with capacitive coupling at each other's end. The mutual couplings between the meander monopoles are utilized to produce two radiation nulls at the two band-edges. Based on the design method, the prototype of the proposed antenna was designed, fabricated and measured. The measured results show that the antenna has a broad bandwidth of 16% for S 11 <; -10 dB. Also, ideal omnidirectional radiation patterns, and steep band-edge selectivity with two radiation nulls are achieved for the proposed antenna

    A 3D hydrodynamic flow-focusing device for cell sorting

    Optical-based microfluidic cell sorting has become increasingly attractive for applications in life and environmental sciences due to its ability of sophisticated cell handling in flow. The majority of these microfluidic cell sorting devices employ two-dimensional fluid flow control strategies, which lack the ability to manipulate the position of cells arbitrarily for precise optical detection, therefore resulting in reduced sorting accuracy and purity. Although three-dimensional (3D) hydrodynamic devices have better flow-focusing characteristics, most lack the flexibility to arbitrarily position the sample flow in each direction. Thus, there have been very few studies using 3D hydrodynamic flow focusing for sorting. Herein, we designed a 3D hydrodynamic focusing sorting platform based on independent sheath flow-focusing and pressure-actuated switching. This design offers many advantages in terms of reliable acquisition of weak Raman signals due to the ability to precisely control the speed and position of samples in 3D. With a proof-of-concept demonstration, we show this 3D hydrodynamic focusing-based sorting device has the potential to reach a high degree of accuracy for Raman activated sorting

    A Wideband Series-Fed Circularly Polarized Differential Antenna by Using Crossed Open Slot-Pairs

    A novel method of designing a wideband series-fed circularly polarized (CP) differential antenna by using crossed open slot-pairs is presented in this paper. The near-field distributions and input impedance analyses show that the closely spaced open slot-pairs can radiate as the crossed dipoles and have stable radiating resistance with a compact radiator size. Besides, a wideband half-power phase shifter by using open slot is proposed and utilized to realize CP radiation. The proposed CP antenna is composed of a wide slot-pair and a narrow slot-pair. In the antenna design, the narrow slot-pair is not only excited as a radiator, but also elaborately loaded to provide wideband half-power output and quadrature phase excitation to the wide slot-pair. Both the proposed half-power phase shifter and CP antenna are illustrated by the corresponding equivalent circuits. Based on these analyses, the proposed antenna is designed, fabricated and measured. Compared to the simulated traditionally designed counterpart, 2.1 times wider axial ratio bandwidth is achieved for the proposed antenna. The measured overlapped bandwidth for axial ratio 10 dB is 1.95-3.45 GHz (55.6%). Also, the antenna gain and radiation patterns are measured, which agree well with the simulated results

    Subhealth: definition, criteria for diagnosis and potential prevalence in the central region of China

    BACKGROUND: A full evaluation of health conditions is necessary for the effective implementation of public health interventions. However, terms to address the intermediate state between health and disease are lacking, leading the public to overlook this state and thus increasing the risks of developing disease. METHODS: A cross-sectional health survey of 1,473 randomly recruited Chinese Han adults of both sexes living in the central region of China. The criteria for diagnosis of subhealth was defined as the presence of ≥ 1 of the following abnormalities: body mass index ≥ 25 kg/m(2) or waist circumference ≥ 102 cm in men and 88 cm in women; systolic pressure 120–139 mmHg and/or diastolic pressure 80–89 mmHg; serum triglyceride level ≥ 150 mg/dL and/or total cholesterol level ≥ 200 mg/dL and/or high-density lipoprotein cholesterol level < 40 mg/dL in men and 50 mg/dL in women; serum glucose level 110–125 mg/dL; estimated glomerular filtration rate 60–89 ml/min/1.73 m(2); levels of liver enzymes in liver function tests between 41–59 U/L, or with fatty liver disease but < 33% of affected hepatocytes; levels of oxidative stress biomarkers beyond the reference range of 95%; or problems with both sleep quality and psychological state. RESULTS: The prevalences of subhealth and disease in the central region of China were 36.6% and 43.1%, respectively. The prevalence of disease increased from 26.3% in participants aged 20–39 years, to 47.6% and 78.9% for participants aged 40–59 years and those aged 60 years or older, respectively. Compared with participants aged 20–39, the prevalences of health and subhealth in participants aged 60 years or older decreased by 86.7% and 60.3%, respectively. The prevalence of subhealth was increased in association with increases in lifestyle risk scores, while the prevalences of both health and disease were reduced. CONCLUSION: The prevalences of subhealth and disease are high in central China. Subhealth is associated with high lifestyle risk scores. Both the health care sector and the public should pay more attention to subhealth. Lifestyle modifications and/or psychological interventions are needed to ameliorate these conditions
