    Antioxidative potential of polysaccharide fractions produced from traditional Chinese medicinal macrofungus Cordyceps jiangxiensis in vitro

    Cordyceps jiangxiensis, also called ‘CaoMuWang’, is a medicinal entomopathogenic macrofungus native to eastern China. Polysaccharide fractions from cultured C. jiangxiensis exhibited potent antitumor activity via the induction of cell cycle arrest and apoptotic pathway. Antioxidant pathway is also one action of mechanism of antitumor; thus, the antioxidant abilities of these polysaccharide fractions were overall evaluated by five in vitro assays such as the scavenging abilities on DPPH•, hydroxyl and superoxide anion radicals, the reducing power and the chelating ability on ferrous ions. Among these assays, the polysaccharide fractions presented more excellent scavenging abilities on superoxide anion radicals than that of the positive control. When compared with the positive control, the polysaccharide fractions from C. jiangxiensis only had moderate scavenging activities on both DPPH and hydroxyl free radicals, moderate reducing power and ferrous ion chelating activity. The antioxidant abilities of the different polysaccharide fractions had certain differences at all the tested doses and all had a dose-dependent manner. The results suggested that, polysaccharides are  important antioxidant component in the medicinal Cordyceps fungi and have direct and potent antioxidant ability, and that C. jiangxiensis also is a promising potential source for the development of natural antioxidant.Key words: Cordyceps jiangxiensis, polysaccharide, antioxidant activity

    Tectonic affinity of the west Qinling terrane (central China): North China or Yangtze?

    [33] Neogene (̃14 Ma) basaltic magmatism has occurred in west Qinling, at the northeastern corner of the Tibetan Plateau. Furthermore, U-Pb ages and Hf isotopic data of xenocrystic zircons indicate that the unexposed Neoarchean (2.7-2.5 Ga) basement beneath the Phanerozoic outcrops in west Qinling has affinities with the southern margin of the north China block. The basement has a complex evolution, including the addition of juvenile mantle material at ̃2.7- 2.4 Ga and 1.1-0.8 Ga and reworking at ̃1.8 Ga and possibly at 1.4 Ga. Phanerozoic thermal events at 320-300 Ma, 230 Ma, and 160 Ma also have affected the basement. We interpret the west Qinling orogenic terrane as originally separated from the north China block, joined to the northern Yangtze block during the Meso-Neoproterozoic, and finally involved in the northward subduction and collision of the Yangtze block in the Paleozoic and early Mesozoic and subsequent lithospheric extension in the Jurassic. © 2010 by the American Geophysical Union.published_or_final_versio

    Bayesian network approach to fault diagnosis of a hydroelectric generation system

    This study focuses on the fault diagnosis of a hydroelectric generation system with hydraulic-mechanical-electric structures. To achieve this analysis, a methodology combining Bayesian network approach and fault diagnosis expert system is presented, which enables the time-based maintenance to transform to the condition-based maintenance. First, fault types and the associated fault characteristics of the generation system are extensively analyzed to establish a precise Bayesian network. Then, the Noisy-Or modeling approach is used to implement the fault diagnosis expert system, which not only reduces node computations without severe information loss but also eliminates the data dependency. Some typical applications are proposed to fully show the methodology capability of the fault diagnosis of the hydroelectric generation system

    Efficacy of praziquantel and artemisinin derivatives for the treatment and prevention of human schistosomiasis: a systematic review and meta-analysis

    <p>Abstract</p> <p>Background</p> <p>Praziquantel has been used as first-line drug for chemotherapy of schistosomiasis since 1984. Besides praziquantel, artemether and artesunate have also been used for the control of this infectious disease since late 1990s. In this article, we conducted a systematic review and meta-analysis to evaluate the antischistosomal efficacy of different medication strategies including monotherapy or combination therapies of these drugs.</p> <p>Results</p> <p>A number of 52 trials from 38 articles published in peer-reviewed journals before July 2011 were selected for analysis after searching the following literature databases: the Cochrane Library, PubMed/Medline, ISI Web of Science, Chinese Biomedicine Literature Database, and China National Knowledge Infrastructure. Our meta-analyses showed that a dosage of 30-60 mg/kg praziquantel compared with placebo produced a protection rate of about 76% (95% CI: 67%-83%) for treating human schistosomiasis, which varied from 70% to 76% with no significant differences among the subspecies <it>S. haematobium</it>, <it>S. japonicum </it>or <it>S. mansoni</it>. Protection rates were higher when praziquantel doses were elevated, as concluded from the nRCTs results: the protection rate of praziquantel at 40 mg/kg was 52% (95% CI: 49%-55%), and it increased to 91% (95% CI: 88%-92%) when the dosages were elevated to 60/80/100 mg/kg divided two or more doses. Multiple doses of artemether or artesunate over 1- or 2-week intervals resulted in protection rates of 65% to 97% for preventing schistosomiasis, and increased doses and shorter medication intervals improved their efficacies. Praziquantel and artemisinin derivatives (artemether or artesunate) in combination resulted in a higher protection rate of 84% (95% CI: 64%-91%) than praziquantel monotherapy for treatment. praziquantel and artesunate in combination had a great protection rate of 96% (95% CI: 78%-99%) for preventing schistosomes infection.</p> <p>Conclusions</p> <p>According to the results, praziquantel remains effective in schistosomiasis treatment, and multiple doses would improve its efficacy; meanwhile, praziquantel is also a good drug for preventing acute schistosomiasis morbidity. It's better to use multiple doses of artemether or artesunate with 1- or 2-week intervals for prevention against schistosome infection. Praziquantel and artemether or artesunate in combination perform better in treatment than praziquantel monotherapy, and they are especially suitable for treating the patients with repeated exposure to infected water.</p

    Hepatotoxicity or Hepatoprotection? Pattern Recognition for the Paradoxical Effect of the Chinese Herb Rheum palmatum L. in Treating Rat Liver Injury

    The hepatotoxicity of some Chinese herbs has been a cause for concern in recent years. However, some herbs, such as rhubarb, have been documented as having both therapeutic and toxic effects on the liver, leading to the complex problem of distinguishing the benefits from the risks of using this herb. To comparatively analyze the dose-response relationship between rhubarb and hepatic health, we administrated total rhubarb extract(RE) to normal and carbon tetrachloride(CCl4)-treated rats for 12 weeks at 4 dosage levels(2.00, 5.40, 14.69 and 40.00 g·kg−1, measured as the quantity of crude material), followed by biochemical and histopathological tests of the rats' livers. A composite pattern was extracted by factor analysis, using all the biochemical indices as variables, into a visual representation of two mathematically obtained factors, which could be interpreted as the fibrosis factor and the cellular injury factor, according to the values of the variable loadings. The curative effect of administering the two lowest dosages of RE to CCl4-treated rats was mainly expressed as a decrease in the extent of cellular injury. The hepatoprotective mechanism of RE might be related to its antioxidant effect, the antagonism of the free radical damage to hepatocytes caused by CCl4. By contrast, the RE-induced liver damage was mainly expressed as a significant increase in the amount of fibrosis in both normal rats at all dosage levels and CCl4-treated rats at the two highest dosage levels. Therefore, the hepatotoxic potential of RE could be attributable to the liver cell fibrosis induced by high doses of the herb. This study illustrates the bidirectional potential of rhubarb and demonstrates the feasibility of using factor analysis to study the dose-response relationships between herbal medicines and hepatotoxicity or the healing effects of these herbs by extracting the underlying interrelationships among a number of functional bio-indices in a holistic manner

    An Effective Amperometric Biosensor Based on Gold Nanoelectrode Arrays

    A sensitive amperometric biosensor based on gold nanoelectrode array (NEA) was investigated. The gold nanoelectrode array was fabricated by template-assisted electrodeposition on general electrodes, which shows an ordered well-defined 3D structure of nanowires. The sensitivity of the gold NEA to hydrogen peroxide is 37 times higher than that of the conventional electrode. The linear range of the platinum NEA toward H2O2is from 1 × 10−6to 1 × 10−2 M, covering four orders of magnitudes with detection limit of 1 × 10−7 M and a single noise ratio (S/N) of four. The enzyme electrode exhibits an excellent response performance to glucose with linear range from 1 × 10−5to 1 × 10−2 M and a fast response time within 8 s. The Michaelis–Menten constantkm and the maximum current densityimaxof the enzyme electrode were 4.97 mM and 84.60 μA cm−2, respectively. This special nanoelectrode may find potential application in other biosensors based on amperometric signals
