    Genetic loci mapping for ear axis weight using recombinant inbred line (RIL) population under different nitrogen regimes in maize

    Ear axis weight (EAW) is one of the important agronomic traits in maize (Zea mays L.), related to yield. To understand its genetic basis, a recombinant inbred line (RIL) population, derived from the cross Mo17 × Huangzao4, was used for quantitative trait locus mapping (QTL) for EAW under high and low nitrogen (N) regimes. The results showed that a total of three QTLs were mapped on chromosomes 2 (two) and 4 (one) under the two N regimes, which could explain phenotypic variances from 4.76 to 7.12%. They were near to their linked markers, with mapping interval of 0.2 to 1.0 cM. The two loci on chromosome 2 (bin 2.09) made EAW increase due to positive additive effects, while the other locus on chromosome 4 (bin 4.08) made EAW decrease to some extent, owing to negative additive effects. These results are beneficial for understanding the genetic basis of KNE and developing marker-assisted selection in maize breeding project.Key words: Maize (Zea mays L.), ear axis weight, quantitative trait locus, recombinant inbred line, nitrogen

    A quantitative trait locus for the number of days from sowing to seedling emergence in maize

    Quantitative trait locus (QTL) mapping provides useful information for breeding programs since it allows the estimation of genomic locations and genetic effects of chromosomal regions related to the expression of quantitative traits. The number of days from sowing to seedling emergence (NDSSE) is an important agronomic trait in a maize (Zea mays L.) breeding project which is related to yield. To determine its genetic basis, a recombinant inbred line (RIL) population and two nitrogen (N) regimes were used to detect the QTLs associated with NDSSE; as a result, one QTL was identified under high N regime, on chromosome 9, which could explain 6.20% of phenotypic variance and a decrease of 0.18 of NDSSE due to an additive effect.     Downregulation of the Canonical WNT Signaling Pathway by TGF beta 1 Inhibits Photoreceptor Differentiation of Adult Human Muller Glia with Stem Cell Characteristics

    Muller glia are responsible for the retina regeneration observed in zebrafish. Although the human retina harbors Muller glia with stem cell characteristics, there is no evidence that they regenerate the retina after disease or injury. Transforming growth factor-b (TGFb) and Wnt signaling regulate retinal neurogenesis and inflammation, but their roles in the neural differentiation of human Mu¨ller stem cells (hMSC) are not known. We examined hMSC lines in vitro for the expression of various Wnt signaling components and for their modulation by TGFb1, as well as the effect of this cytokine on the photoreceptor differentiation of these cells. Culture of hMSC with a combination of factors that induce photoreceptor differentiation of hMSC (FGF2, taurine, retinoic acid, and insulin-like growth factor type1; FTRI), markedly upregulated the expression of components of the canonical Wnt signaling pathway, including WNT2B, DKK1, and active b-CATENIN. Although FTRI did not modify mRNA expression of WNT5B, a component of the noncanonical/planar cell polarity Wnt pathway, it upregulated its secretion. Furthermore, TGFb1 not only decreased WNT2B expression, but also inhibited FTRI-induced photoreceptor differentiation of hMSC, as determined by expression of the photoreceptor markers NR2E3, RHODOPSIN, and RECOVERIN. Inhibition of TGFb1 signaling by an ALK5 inhibitor prevented TGFb1-induced changes in the expression of the two Wnt ligands examined. More importantly, inhibition of the canonical WNT signaling by XAV-939 prevented FTRI-induced photoreceptor differentiation. These observations suggest that TGFb may play a key role in preventing neural differentiation of hMSC and may constitute a potential target for induction of endogenous regeneration of the human retina

    High Aspect Ratio (L/D) Riser Viv Prediction Using Wake Oscillator Model

    A two-dimensional (2-D) vortex-induced vibration (VIV) prediction model for high aspect ratio (LID) riser subjected to uniform and sheared flow is studied in this paper. The nonlinear structure equations are considered. The near wake dynamics describing the fluctuating nature of vortex shedding is modeled using classical van der Pol equation. A new approach was applied to calibrate the empirical parameters in the wake oscillator model. Compared the predicted results with the experimental data and computational fluid dynamic (CFD) results. Good agreements are observed. It can be concluded that the present model can be used as simple computational tool in predicting some aspects of VIV of long flexible structures. (C) 2008 Elsevier Ltd.
