    Auxin depletion from leaf primordia contributes to organ patterning

    Stem cells are responsible for organogenesis, but it is largely unknown whether and how information from stem cells acts to direct organ patterning after organ primordia are formed. It has long been proposed that the stem cells at the plant shoot apex produce a signal, which promotes leaf adaxial-abaxial (dorsoventral) patterning. Here we show the existence of a transient low auxin zone in the adaxial domain of early leaf primordia. We also demonstrate that this adaxial low auxin domain contributes to leaf adaxial-abaxial patterning. The auxin signal is mediated by the auxin-responsive transcription factor MONOPTEROS (MP), whose constitutive activation in the adaxial domain promotes abaxial cell fate. Furthermore, we show that auxin flow from emerging leaf primordia to the shoot apical meristem establishes the low auxin zone, and that this auxin flow contributes to leaf polarity. Our results provide an explanation for the hypothetical meristem-derived leaf polarity signal. Opposite to the original proposal, instead of a signal derived from the meristem, we show that a signaling molecule is departing from the primordium to the meristem to promote robustness in leaf patterning

    Yupingfeng polysaccharide promote the growth of chickens via regulating gut microbiota

    IntroductionYupingfeng polysaccharide (YPF-P) is the main substance of alcohol deposition in Yupingfeng powder, which has many biological functions such as enhancing immunity, repairing intestinal barrier and enhancing antioxidant ability. This study employed in vitro growth-promoting drug feed additives and animal experiments to comprehensively evaluate the use of YPF-P in broiler production.MethodsA total of 1,296 151 days-old Qingyuan Partridge chickens were randomly divided into four groups with six replicates and 54 hens per replicate: the control group was fed basal diet, and the experimental groups were fed diets supplemented with 4 g/kg, 8 g/kg, and 12 g/kg YPF-P for 14 days. Broilers were weighed before and at the end of the experiment to calculate total weight gain (GW), average daily gain (ADG), and feed compensation. At the end of the experiment, six chickens from each group were randomly selected for subwing vein blood sampling, which was used to measure serum biochemical indicators GHRH, GH, and IGF-1 by ELISA method. Randomly select chickens from control group and 8 g/kg group for slaughter, and cecal contents were collected for 16S high-throughput sequencing.ResultsDietary supplementation of 8 g/kg YPF-P can significantly increase the final body weight, total weight gain, average daily gain and decrease the feed to gain ratio of chickens. During 151–165 days, serum IGF-1 concentrations increased significantly (p < 0.05). There were no significant changes in serum GH concentration (p > 0.05). In terms of gut microbiota, there was no significant difference between control group and test group in Shannon index and Simpson index. Compared with the control group,the addition of 8 g/kgYPF-P significantly increased the abundance of Firmicutes and significantly decreased the abundance of Bacteroides at the phylum level.At the genus level, the relative abundance of unclassified_Oscillospiraceae was significantly increased and the unclassified_Muribaculaceae, uncultured_Bacteroidales_bacterium, Lactobacillus, Alloprevotella, Ligilactobacillus, Prevotellaceae_UCG_001, and unclassified_Atopobiaceae was significantly decreased.ConclusionThe above results showed that adding 8 mg/kg of YPF-P could increase the average daily gain of Qingyuan Partridge chickens, reduce the ratio of feed to meat, and affect the distribution proportion of intestinal microflora in chickens to some extent

    Indoor Particulate Matter Transfer in CNC Machining Workshop and The Influence of Ventilation Strategies—A Case Study

    Particulate matter in Computer Numerical Control (CNC) machining workshop is harmful to workers’ health. This paper studies particulate matter transfer and the performance of various ventilation strategies in a CNC machining workshop. To obtain the boundary condition of the particle field, instruments were installed to obtain the particle size attenuation characteristics and source strength, respectively. The results show that the 99% cumulative mass concentration of particles is distributed within 1.5 μm, and the release rate of particles from the full enclosure. Next, the indoor flow field and particle field were simulated by numerical simulation with the measured boundary conditions. The working area’s age of air, particle concentration, and ventilation efficiency were compared between four displacement ventilation methods and one mixed ventilation method. The results show that the working area’s mean particle concentration and ventilation efficiency under longitudinal displacement ventilation is better than other methods. At the same time, the mean age of air is slightly worse. In addition, mixed ventilation can obtain lower mean age of air, but the particle concentration is higher in the working area. The bilateral longitudinal ventilation can be improved by placing axial circulation fans with vertical upward outlets in the center of the workshop

    How Low Nucleation Density of Graphene on CuNi Alloy is Achieved

    CuNi alloy foils are demonstrated to be one of the best substrates for synthesizing large area single-crystalline graphene because a very fast growth rate and low nucleation density can be simultaneously achieved. The fast growth rate is understood to be due the abundance of carbon precursor supply, as a result of the high catalytic activity of Ni atoms. However, a theoretical understanding of the low nucleation density remains controversial because it is known that a high carbon precursor concentration on the surface normally leads to a high nucleation density. Here, the graphene nucleation on the CuNi alloy surfaces is systematically explored and it is revealed that: i) carbon atom dissolution into the CuNi alloy passivates the alloy surface, thereby drastically increasing the graphene nucleation barrier; ii) carbon atom diffusion on the CuNi alloy surface is greatly suppressed by the inhomogeneous atomic structure of the surface; and iii) a prominent increase in the rate of carbon diffusion into the bulk occurs when the Ni composition is higher than the percolation threshold. This study reveals the key mechanism for graphene nucleation on CuNi alloy surfaces and provides a guideline for the catalyst design for the synthesis of graphene and other 2D materials

    Serum lactate dehydrogenase activities as systems biomarkers for 48 types of human diseases

    Most human diseases are systems diseases, and systems biomarkers are better fitted for diagnostic, prognostic, and treatment monitoring purposes. To search for systems biomarker candidates, lactate dehydrogenase (LDH), a housekeeping protein expressed in all living cells, was investigated. To this end, we analyzed the serum LDH activities from 172,933 patients with 48 clinically defined diseases and 9528 healthy individuals. Based on the median values, we found that 46 out of 48 diseases, leading by acute myocardial infarction, had significantly increased (p  0.8) for hepatic encephalopathy and lung fibrosis

    EEG-based emotion classification using spiking neural networks

    A novel method of using the spiking neural networks (SNNs) and the electroencephalograph (EEG) processing techniques to recognize emotion states is proposed in this paper. Three algorithms including discrete wavelet transform (DWT), variance and fast Fourier transform (FFT) are employed to extract the EEG signals, which are further taken by the SNN for the emotion classification. Two datasets, i.e., DEAP and SEED, are used to validate the proposed method. For the former dataset, the emotional states include arousal, valence, dominance and liking where each state is denoted as either high or low status. For the latter dataset, the emotional states are divided into three categories (negative, positive and neutral). Experimental results show that by using the variance data processing technique and SNN, the emotion states of arousal, valence, dominance and liking can be classified with accuracies of 74%, 78%, 80% and 86.27% for the DEAP dataset, and an overall accuracy is 96.67% for the SEED dataset, which outperform the FFT and DWT processing methods. In the meantime, this work achieves a better emotion classification performance than the benchmarking approaches, and also demonstrates the advantages of using SNN for the emotion state classifications

    lncRNA profile study reveals the mRNAs and lncRNAs associated with docetaxel resistance in breast cancer cells

    Abstract Resistance to adjuvant systemic treatment, including taxanes (docetaxel and paclitaxel) is a major clinical problem for breast cancer patients. lncRNAs (long non-coding RNAs) are non-coding transcripts, which have recently emerged as important players in a variety of biological processes, including cancer development and chemotherapy resistance. However, the contribution of lncRNAs to docetaxel resistance in breast cancer and the relationship between lncRNAs and taxane-resistance genes are still unclear. Here, we performed comprehensive RNA sequencing and analyses on two docetaxel-resistant breast cancer cell lines (MCF7-RES and MDA-RES) and their docetaxel-sensitive parental cell lines. We identified protein coding genes and pathways that may contribute to docetaxel resistance. More importantly, we identified lncRNAs that were consistently up-regulated or down-regulated in both the MCF7-RES and MDA-RES cells. The co-expression network and location analyses pinpointed four overexpressed lncRNAs located within or near the ABCB1 (ATP-binding cassette subfamily B member 1) locus, which might up-regulate the expression of ABCB1. We also identified the lncRNA EPB41L4A-AS2 (EPB41L4A Antisense RNA 2) as a potential biomarker for docetaxel sensitivity. These findings have improved our understanding of the mechanisms underlying docetaxel resistance in breast cancer and have provided potential biomarkers to predict the response to docetaxel in breast cancer patients

    Spatial-temporal distribution and transport flux of polycyclic aromatic hydrocarbons in a large hydropower reservoir of Southeast China: Implication for impoundment impacts.

    In order to investigate the impacts of dam-related water impoundment on the spatial-temporal variations and transport of anthropogenic organic pollutants, 15 priority polycyclic aromatic hydrocarbons (PAHs) were analyzed in water samples from the Shuikou Reservoir (SKR) of the Minjiang River. The SKR was formed after the construction of the Shuikou Dam, which is the largest hydropower station in Southeast China. The water samples were collected from the backwater zone of the SKR, in both the wet and dry seasons, corresponding to the drainage and impoundment periods of water flow, respectively. The concentrations of the dissolved PAHs in surface water from the wet season (average of 161 ± 97 ng L-1) were significantly higher (ANOVA, p < 0.01) than those from the dry season (average of 43 ± 21 ng L-1). PAH concentrations in the SKR decreased from upstream (industrialized cities) to downstream (rural towns or counties), indicating high PAH loads caused by intensive urbanization effects. The high proportions of 3-ring PAHs in the wet season were from local sources via surface runoff; while the elevated proportions of 4- to 6- ring PAHs in the dry season reflected atmospheric deposition emerged of these PAHs and/or volatilization of 3-ring PAHs enhanced. Molecular diagnostic ratios of PAH isomers in multimedium and principal component analysis indicated that PAH presence in the SKR was mainly attributed to pyrogenic origin. The isomeric ratios of fluoranthene to fluoranthene plus pyrene in the wet season were homogeneous, implying that there were continuous new inputs along the riverine runoff. However, these ratios showed spatial downward trend in the dry season, indicating continued degradation of PAHs occurred along the transport path during the impoundment period. The input and output fluxes of PAHs in the SKR were 5330 kg yr-1 and 2991 kg yr-1, revealing that the reservoir retained contaminants after impoundment of the hydropower dam
