    Recombinant Expression of Serratia marcescens Outer Membrane Phospholipase A (A1) in Pichia pastoris and Immobilization With Graphene Oxide-Based Fe3O4 Nanoparticles for Rapeseed Oil Degumming

    Enzymatic degumming is an effective approach to produce nutritional, safe, and healthy refined oil. However, the high cost and low efficiency of phospholipase limit the application of enzymatic degumming. In this study, an 879 bp outer membrane phospholipase A (A1) (OM-PLA1) gene encoding 292 amino acid residues was isolated from the genome of Serratia marcescens. The recombinant OM-PLA1 profile of appropriately 33 KDa was expressed by the engineered Pichia pastoris GS115. The OM-PLA1 activity was 21.2 U/mL with the induction of 1 mM methanol for 72 h. The expression efficiencies of OM-PLA1 were 0.29 U/mL/h and 1.06 U/mL/OD600. A complex of magnetic graphene oxide (MGO) and OM-PLA1 (MGO-OM-PLA1) was prepared by immobilizing OM-PLA1 with graphene oxide-based Fe3O4 nanoparticles by cross-linking with glutaraldehyde. The content of phosphorus decreased to 5.1 mg/kg rapeseed oil from 55.6 mg/kg rapeseed oil with 0.02% MGO-OM-PLA1 (w/w) at 50°C for 4 h. MGO-OM-PLA1 retained 51.7% of the initial activity after 13 times of continuous recycling for the enzymatic degumming of rapeseed oil. This study provided an effective approach for the enzymatic degumming of crude vegetable oil by developing a novel phospholipase and improving the degumming technology

    Neuronally released vasoactive intestinal polypeptide alters atrial electrophysiological properties and may promote atrial fibrillation

    BACKGROUND: Vagal hyperactivity promotes atrial fibrillation (AF), which has been almost exclusively attributed to acetylcholine. Vasoactive intestinal polypeptide (VIP) and acetylcholine are neurotransmitters co-released during vagal stimulation. Exogenous VIP has been shown to promote AF by shortening action potential duration (APD), increasing APD spatial heterogeneity, and causing intra-atrial conduction block. OBJECTIVE: The purpose of this study was to investigate the effects of neuronally released VIP on atrial electrophysiologic properties during vagal stimulation. METHODS: We used a specific VIP antagonist (H9935) to uncover the effects of endogenous VIP released during vagal stimulation in canine hearts. RESULTS: H9935 significantly attenuated (1) the vagally induced shortening of atrial effective refractory period and widening of atrial vulnerability window during stimulation of cervical vagosympathetic trunks (VCNS) and (2) vagal effects on APD during stimulation through fat-pad ganglion plexus (VGPS). Atropine completely abolished these vagal effects during VCNS and VGPS. In contrast, VGPS-induced slowing of local conduction velocity was completely abolished by either VIP antagonist or atropine. In pacing-induced AF during VGPS, maximal dominant frequencies and their spatial gradients were reduced significantly by H9935 and, more pronouncedly, by atropine. Furthermore, VIP release in the atria during vagal stimulation was inhibited by atropine, which may account for the concealment of VIP effects with muscarinic blockade. CONCLUSION: Neuronally released VIP contributes to vagal effects on atrial electrophysiologic properties and affects the pathophysiology of vagally induced AF. Neuronal release of VIP in the atria is inhibited by muscarinic blockade, a novel mechanism by which VIP effects are concealed by atropine during vagal stimulation

    Analysis of fine grained sand and shale sedimentary characteristics in estuary based on sediment dynamics

    Due to the dual effects of fluvial and tides, the tidal sand bars in estuaries have complex sedimentary characteristics and complex internal structures, making them difficult to predict and describe. In this paper, the sedimentary dynamics numerical simulation method is used to establish a tidal-controlled estuary model. The effects of tidal range and sediment grain size on tidal sand bars are simulated. The length, width, and thickness of tidal sand bars, as well as the length and thickness of the internal shale layer, are also analyzed. The results show that in the environment of a tide-controlled estuary, the tidal range has a more significant effect on tidal sand bars compared to the sediment grain size under the specific conditions used in this study. The main effect of tidal range on tidal sand bars is that the greater the tidal range, the greater the length-to-width ratio of the sandbank, and the higher the degree of sandbank development. In a tidal-controlled estuary environment, the formation and distribution of shale layer structures are also affected by tides: the length of the shale layer increases as the tidal energy increases, but the changes in the thickness are not obvious. Numerical simulations of the development and distribution of the tidal sand bars and shale layers in estuaries based on sedimentary dynamics will provide a basis for the sedimentary evolution of tide-controlled estuaries and will provide guidance for the exploration and development of tidal estuaries

    The Modified Shields Classification and 12 Families with Defined DSPP Mutations

    Mutations in Dentin Sialophosphoprotein (DSPP) are known to cause, in order of increasing severity, dentin dysplasia type-II (DD-II), dentinogenesis imperfecta type-II (DGI-II), and dentinogenesis imperfecta type-III (DGI-III). DSPP mutations fall into two groups: a 5′-group that affects protein targeting and a 3′-group that shifts translation into the −1 reading frame. Using whole-exome sequence (WES) analyses and Single Molecule Real-Time (SMRT) sequencing, we identified disease-causing DSPP mutations in 12 families. Three of the mutations are novel: c.53T>C/p.(Val18Ala); c.3461delG/p.(Ser1154Metfs*160); and c.3700delA/p.(Ser1234Alafs*80). We propose genetic analysis start with WES analysis of proband DNA to identify mutations in COL1A1 and COL1A2 causing dominant forms of osteogenesis imperfecta, 5′-DSPP mutations, and 3′-DSPP frameshifts near the margins of the DSPP repeat region, and SMRT sequencing when the disease-causing mutation is not identified. After reviewing the literature and incorporating new information showing distinct differences in the cell pathology observed between knockin mice with 5′-Dspp or 3′-Dspp mutations, we propose a modified Shields Classification based upon the causative mutation rather than phenotypic severity such that patients identified with 5′-DSPP defects be diagnosed as DGI-III, while those with 3′-DSPP defects be diagnosed as DGI-II

    There is an increasing interest in the question as to which aspects in speech are tightly coupled with co-speech gesture. A general consensus now exists that gestural representation is influenced by event structure, but it is less clear to what extent gesture interacts with syntactic encoding. The present study thus investigates the issue, taking the double object construction and its prepositional paraphrase (both expressing transfer events) in relation to gesture as the starting point. Based on data from American English talk show programs, it is found that the rate of gesture production is closely linked with event structure, whereas gestural Modes of Representation and the dynamicity of Acting gestures are clearly sensitive to the subjective online conceptualization of events or syntactic constructions in encoding events in addition to the event structure. The current findings constitute further evidence in support of the hypothesis which predicts that gestural representation is determined by the online choice of the syntactic encoding/the online construal of events as well as non-linguistic motor-spatial properties of events, but they are at odds with the Free Imagery Hypothesis, which predicts that gestural representation simply derives from pre-linguistic imagery

    Transitivity, events, and gesture: The case of the causative-inchoative alternation

    An increasing number of studies are being devoted to the investigation of what aspects of grammar, and of events, expressed in speech are coordinated with gesture. However, previous studies have focused on gesture use in relation to either transitivity or event properties, without considering how these factors interact. In this study, we consider how gesture use relates to transitivity when the type of event in the causativeinchoative alternation is considered, and also how gesture use relates to properties of the events when the type of transitivity is considered. We found various relations both between gesture use and transitivity on the one hand, and between gesture use and certain properties of events on the other hand. Whereas some of the results contrast with the findings in previous studies about the relation between gesture and transitivity, other results obtained actually reinforce and complement some previous findings. The results concerning event properties and gesture also add to previous studies about which properties of certain motor-spatial events relate to gesture and how they do so. The study thus provides a more nuanced understanding of the relation between gesture and language

    Effect of an Additional 30 Minutes Spent Outdoors during Summer on Daily Steps and Individually Experienced Heat Index

    Spending time outdoors is associated with increased physical activity; however, high ambient temperature/humidity, together with built environment features in urban versus rural environments, may influence physical activity. We conducted an intervention trial with 89 urban and 88 rural participants performing normal activities on Days 1–2 (baseline) and spending an additional 30 min outdoors on Days 3–7 (intervention) in the summer. Participants wore a pedometer with real-time visual feedback to track daily steps taken and a thermometer clipped to their shoe to track temperatures experienced individually. Hygrometer–thermometers were deployed in participants’ neighborhoods to collect finer resolution ambient heat indexes in addition to regional weather station measurements. Using linear mixed effects models and adjusting for ambient conditions and individual-level factors, participants on average walked 637 (95%CI (83, 1192)) more steps and had a 0.59 °C (95%CI (0.30, 0.88)) lower daily mean individually experienced heat index during intervention days compared to baseline days. The intervention benefit of increased physical activity was greater in rural residents who were less active at baseline, compared to urban residents. Our results suggest adding a small amount of additional time outdoors may improve physical activity without increasing participants’ heat exposure, even during summer in a humid subtropical climate

    Effect of PEW and CS on the Thermal, Mechanical, and Shape Memory Properties of UHMWPE

    Modified ultra-high-molecular-weight polyethylene (UHMWPE) with calcium stearate (CS) and polyethylene wax (PEW) is a feasible method to improve the fluidity of materials because of the tense entanglement network formed by the extremely long molecular chains of UHMWPE, and a modified UHMWPE sheet was fabricated by compression molding technology. A Fourier-transform infrared spectroscopy test found that a new chemical bond was generated at 1097 cm−1 in the materials. Besides, further tests on the thermal, thermomechanical, mechanical, and shape memory properties of the samples were also conducted, which indicates that all properties are affected by the dimension and distribution of crystal regions. Moreover, the experimental results indicate that the addition of PEW and CS can effectively improve the mechanical properties. Additionally, the best comprehensive performance of the samples was obtained at the PEW content of 5 wt % and the CS content of 1 wt %. In addition, the effect of temperature on the shape memory properties of the samples was investigated, and the results indicate that the shape fixity ratio (Rf) and the shape recovery ratio (Rr) can reach 100% at 115 °C and 79% at 100 °C, respectively, which can contribute to the development of UHMWPE-based shape memory polymers
