    Predictors of Gull-billed tern (Gelochelidon nilotica) nest survival in artificial coastal saltpans, Bohai Bay, China

    Background Coastal saltpans are a common supratidal human-modified wetland habitat found within many coastal landscape mosaics. Commercial salt production and aquaculture practices often result in the creation of exposed coastal substrates that could provide suitable breeding habitat for waterbird populations; however, few studies have quantified waterbird breeding success in these artificial wetlands. Methods Here we examine the nesting behavior of the Gull-billed tern (Gelochelidon nilotica) breeding in the Nanpu coastal saltpans of Bohai Bay, Yellow Sea, China over three consecutive nesting seasons (2017–2019) by using nest survival model in Program MARK. Results The results revealed that nest survival of Gull-billed terns in coastal saltpans (0.697) was higher than previously published estimates from other regions, with an estimated daily survival rate (DSR) of 0.982 ± 0.001 (±95% CI). High nest survival was mainly attributed to low levels of human disturbances and low predation rates, while exposure to strong winds, flooding and silting were the main factors causing nest failure. Model-averaged estimates revealed that eggs laid in nests located on ‘habitat islands’ with feather or clam shell substrates were most likely to hatch. Initiation date, nest age, clutch size and quadratic effects of nearest-neighbor distance, nearest distance to road and nearest distance to water were all significant predictors of nest success, but the nest survival declined overall from 2017 to 2019 due to the degradation and loss of breeding habitat anthropogenically caused by rising water levels. Discussion Coastal saltpans represent an alternative breeding habitat for the Gull-billed tern populations in Bohai Bay, but conservation management should prioritize flood prevention to improve the extent and quality of breeding habitat, concurrent with efforts to create further ‘habitat islands’ with suitable nesting substrate

    Artificial wetlands as breeding habitats for shorebirds:A case study on Pied Avocets in China’s largest saltpan complex

    Artificial wetlands such as coastal saltpans have replaced a number of coastal natural habitats worldwide and may have accommodated specific waterbird populations in the East Asian–Australasian Flyway (EAAF). The role of saltpans in the EAAF as foraging grounds for shorebirds is widely recognized, although their role as breeding grounds for waterbirds is very limited and contradictory. The Nanpu saltpans in northern Bohai Bay, China, are one of the largest saltpan complexes in the world. In this study, we monitored the nesting success (852 nests) of pied avocets (Recurvirostra avosetta) during three breeding seasons (2015, 2016, and 2018) in the Nanpu saltpans. The nest daily survival rate (DSR) was 0.970; hence, nest survival over the 27 exposure days was 44%. The apparent nest success was 51%. Surprisingly, 55% of nests failed during the laying period. Flooding and nest abandonment were the main causes of nest failure during both the laying and incubation periods. We found a strong positive relationship between the DSR and nest age, with nests that approached hatching having a greater probability of survival than freshly started nests. We also found a strong negative relationship between the DSR and precipitation, with the highest DSR observed for nests that experienced no precipitation. The DSR decreased over the course of the 71-days nesting season and followed a linear trend. The DSR was also density dependent and decreased slightly when nests were denser. A literature review showed that nest survival in the Nanpu saltpans was average compared with that of other studies and that nest success in artificial wetlands was significantly higher than that in natural wetlands or both habitats. Nevertheless, nest success decreased with the study date, suggesting that that breeding conditions for the pied avocet are worsening with time. The loss of saltpans could negatively affect the population of avocets and other ground-nesting waterbird species; therefore, conservation actions and research efforts should be strengthened to understand and conserve these functional wetlands for waterbirds

    Sexual attraction enhances glutamate transmission in mammalian anterior cingulate cortex

    Functional human brain imaging studies have indicated the essential role of cortical regions, such as the anterior cingulate cortex (ACC), in romantic love and sex. However, the neurobiological basis of how the ACC neurons are activated and engaged in sexual attraction remains unknown. Using transgenic mice in which the expression of green fluorescent protein (GFP) is controlled by the promoter of the activity-dependent gene c-fos, we found that ACC pyramidal neurons are activated by sexual attraction. The presynaptic glutamate release to the activated neurons is increased and pharmacological inhibition of neuronal activities in the ACC reduced the interest of male mice to female mice. Our results present direct evidence of the critical role of the ACC in sexual attraction, and long-term increases in glutamate mediated excitatory transmission may contribute to sexual attraction between male and female mice

    Selective activation of microglia in spinal cord but not higher cortical regions following nerve injury in adult mouse

    Neuronal plasticity along the pathway for sensory transmission including the spinal cord and cortex plays an important role in chronic pain, including inflammatory and neuropathic pain. While recent studies indicate that microglia in the spinal cord are involved in neuropathic pain, a systematic study has not been performed in other regions of the central nervous system (CNS). In the present study, we used heterozygous Cx3cr1GFP/+mice to characterize the morphological phenotypes of microglia following common peroneal nerve (CPN) ligation. We found that microglia showed a uniform distribution throughout the CNS, and peripheral nerve injury selectively activated microglia in the spinal cord dorsal horn and related ventral horn. In contrast, microglia was not activated in supraspinal regions of the CNS, including the anterior cingulate cortex (ACC), prefrontal cortex (PFC), primary and secondary somatosensory cortex (S1 and S2), insular cortex (IC), amygdala, hippocampus, periaqueductal gray (PAG) and rostral ventromedial medulla (RVM). Our results provide strong evidence that nerve injury primarily activates microglia in the spinal cord of adult mice, and pain-related cortical plasticity is likely mediated by neurons

    Birds and marine mammals in Antarctica, especially penguins and seals, are top consumers and critical elements of the Southern Ocean ecosystem. As a region undergoing rapid global change, new challenges will be posed to the survival of these vertebrates species. Global climate change causes many alterations, such as ocean temperature rise, altered sea ice distribution, and abnormal climate events along with effects of intensive human activities, such as fishing. These not only directly affect the spatiotemporal distributions and population dynamics of Antarctic birds and marine mammals but also indirectly influence them via modification of their food resources. At present, the impact of climate change on birds and marine mammals in the Antarctica is focusing on a number of species in a few areas. Response mechanisms of these species are still very limited and therefore require further long-term and continuous monitoring and research

    Comparative study of differentiating human pluripotent stem cells into vascular smooth muscle cells in hydrogel-based culture methods

    Vascular smooth muscle cells (VSMCs), which provides structural integrity and regulates the diameter of vasculature, are of great potential for modeling vascular-associated diseases and tissue engineering. Here, we presented a detailed comparison of differentiating human pluripotent stem cells (hPSCs) into VSMCs (hPSCs-VSMCs) in four different culture methods, including 2-dimensional (2D) culture, 3-dimensional (3D) PNIPAAm-PEG hydrogel culture, 3-dimensional (3D) alginate hydrogel culture, and transferring 3- dimensional alginate hydrogel culture to 2-dimensional (2D) culture. Both hydrogel-based culture methods could mimic in vivo microenvironment to protect cells from shear force, and avoid cells agglomeration, resulting in the extremely high culture efficiency (e.g., high viability, high purity and high yield) compared with 2D culture. We demonstrated hPSC-VSMCs produced from hydrogel-based culture methods had better contractile phenotypes and the potential of vasculature formation. The transcriptome analysis showed the hPSC-VSMCs derived from hydrogel-based culture methods displayed more upregulated genes in vasculature development, angiogenesis and blood vessel development, extracellular matrix compared with 2D culture. Taken together, hPSC-VSMCs produced from hydrogel-based culture system could be applied in various biomedical fields, and further indicated the suitable development of alginate hydrogel for industrial production by taking all aspects into consideration

    Spectrometers with ever-smaller footprints are sought after for a wide range of applications in which minimized size and weight are paramount, including emerging in situ characterization techniques. We report on an ultracompact microspectrometer design based on a single compositionally engineered nanowire. This platform is independent of the complex optical components or cavities that tend to constrain further miniaturization of current systems. We show that incident spectra can be computationally reconstructed from the different spectral response functions and measured photocurrents along the length of the nanowire.     Modeling Rett Syndrome Using TALEN-Edited MECP2 Mutant Cynomolgus Monkeys

    Gene-editing technologies have made it feasible to create nonhuman primate models for human genetic disorders. Here, we report detailed genotypes and phenotypes of TALEN-edited MECP2 mutant cynomolgus monkeys serving as a model for a neurodevelopmental disorder, Rett syndrome (RTT), which is caused by loss-of-function mutations in the human MECP2 gene. Male mutant monkeys were embryonic lethal, reiterating that RTT is a disease of females. Through a battery of behavioral analyses, including primate-unique eye-tracking tests, in combination with brain imaging via MRI, we found a series of physiological, behavioral, and structural abnormalities resembling clinical manifestations of RTT. Moreover, blood transcriptome profiling revealed that mutant monkeys resembled RTT patients in immune gene dysregulation. Taken together, the stark similarity in phenotype and/or endophenotype between monkeys and patients suggested that gene-edited RTT founder monkeys would be of value for disease mechanistic studies as well as development of potential therapeutic interventions for RTT

    The impact of different benefit packages of Medical Financial Assistance Scheme on health service utilization of poor population in Rural China

    <p>Abstract</p> <p>Background</p> <p>Since 2003 and 2005, National Pilot Medical Financial Assistance Scheme (MFA) has been implemented in rural and urban areas of China to improve the poorest families' accessibility to health services. Local governments of the pilot areas formulated various benefit packages. Comparative evaluation research on the effect of different benefit packages is urgently needed to provide evidence for improving policy-making of MFA. This study was based on a MFA pilot project, which was one component of Health VIII Project conducted in rural China. This article aimed to compare difference in health services utilization of poor families between two benefit package project areas: H8 towns (package covering inpatient service, some designated preventive and curative health services but without out-patient service reimbursement in Health VIII Project,) and H8SP towns (package extending coverage of target population, covering out- patient services and reducing co-payment rate in Health VIII Supportive Project), and to find out major influencing factors on their services utilization.</p> <p>Methods</p> <p>A cross-sectional survey was conducted in 2004, which used stratified cluster sampling method to select poor families who have been enrolled in MFA scheme in rural areas of ChongQing. All family members of the enrolled households were interviewed. 748 and 1129 respondents from two kinds of project towns participated in the survey. Among them, 625 and 869 respondents were included (age≥15) in the analysis of this study. Two-level linear multilevel model and binomial regressions with a log link were used to assess influencing factors on different response variables measuring service utilization.</p> <p>Results</p> <p>In general, there was no statistical significance in physician visits and hospitalizations among all the respondents between the two kinds of benefit package towns. After adjusting for major confounding factors, poor families in H8SP towns had much higher frequency of MFA use (β = 1.17) and less use of hospitalization service (OR = 0.7 (H8SP/H8), 95%CI (0.5, 1.0)) among all the respondents. While calculating use of hospital services among those who needed, there was significant difference (p = 0.032) in percentage of hospitalization use between H8SP towns (46%) and H8 towns (33%). Meanwhile, the non-use but ought-to-use hospitalization ratio of H8SP (54%) was lower than that of H8 (67 %) towns. This indicated that hospitalization utilizations had improved in H8SP towns among those who needed. Awareness of MFA detailed benefit package and presence of physician diagnosed chronic disease had significant association with frequency of MFA use and hospitalizations. There was no significant difference in rate of borrowing money for illness treatment between the two project areas. Large amount of medical debt had strong association with hospitalization utilization.</p> <p>Conclusions</p> <p>The new extended benefit package implemented in pilot towns significantly increased the poor families' accessibility to MFA package in H8SP than that of H8 towns, which reduced poor families' demand of hospitalization services for their chronic diseases, and improved the poor population's utilization of out-patient services to some degree. It can encourage poor people to use more outpatient services thus reduce their hospitalization need. Presence of chronic disease and hospitalization had strong association with the presence of large amount of medical debt, which indicated that: although establishment of MFA had facilitated accessibility of poor families to this new system, and improved service utilization of poor families to some degree, but its role in reducing poor families' medical debt resulted from chronic disease and hospitalization was still very limited. Besides, the following requirements of MFA: co-payment for in-patient services, ceiling and deductibles for reimbursement, limitations on eligibility for diseases reimbursement, also served as most important obstacles for poor families' access to health care.</p> <p>Therefore, there is great need to improve MFA benefit package design in the future, including extending to cover out-patient services, raising ceiling for reimbursement, removing deductibles of MFA, reducing co-payment rate, and integrating MFA with New Rural Cooperative Medical Scheme more closely so as to provide more protection to the poor families.</p

    Conservation Agriculture Could Improve the Soil Dry Layer Caused by the Farmland Abandonment to Forest and Grassland in the Chinese Loess Plateau Based on EPIC Model

    Converting farmland to forest and grassland alleviated water loss and soil erosion. However, water-intensive grasslands and woodlands could form dry soil layers in the arid or semi-arid zones. Therefore, it is necessary to explore a management method to solve this pedological problem. In this study, based on the Environment Policy Integrated Climate (EPIC) model, the crop productivity and soil dry layer was predicted from 2018 to 2038 in alfalfa and apple land. Then, conservation agriculture and conventional tillage systems were used to repair the soil dry layer in apple and alfalfa systems from 2039–2050 in order to explore their potential. Model verification showed that EPIC simulations of yield, ET, and SWC were generally reliable. The predicted results showed that soil drought was more intense in alfalfa systems. Alfalfa’s annual decrease rate and total amount in the soil available water (SAW) were 27.31 mm year−1 and 652.76 mm, higher than 13.62 mm year−1 and 476 mm of the apple system, and the DSLT of apple’s system was thicker, but DSL-SWC was higher than alfalfa. In the recovery process, the restoration degree of soil desiccation in conservation agriculture was significantly higher than in conventional tillage systems (p &lt; 0.05). In addition, the recovery effect increased with the increase of planting times of shallow root crops, such as potato and soybean. The recovery rate was 27.1 ± 1.72 mm year−1, DSLT was 750 ± 51.2 cm in conventional tillage systems, and the recovery rate was 44.7 ± 1.99 mm year−1, DSLT was 258.3 ± 74.9 cm in conservation agriculture systems. This study provides an effective farmland management method to alleviate soil desiccation and further reveals the new role of the Epic Model in future drought assessment
