Molecular Mechanism for the Hofmeister Effect Derived from NMR and DSC Measurements on Barnase
The effects of sodium thiocyanate, sodium chloride, and sodium sulfate on the ribonuclease barnase were studied using differential scanning calorimetry (DSC) and NMR. Both measurements reveal specific and saturable binding at low anion concentrations (up to 250 mM), which produces localized conformational and energetic effects that are unrelated to the Hofmeister series. The binding of sulfate slows intramolecular motions, as revealed by peak broadening in 13 C heteronuclear single quantum coherence spectroscopy. None of the anions shows significant binding to hydrophobic groups. Above 250 mM, the DSC results are consistent with the expected Hofmeister effects in that the chaotropic anion thiocyanate destabilizes barnase. In this higher concentration range, the anions have approximately linear effects on protein NMR chemical shifts, with no evidence for direct interaction of the anions with the protein surface. We conclude that the effects of the anions on barnase are mediated by solvent interactions. The results are not consistent with the predictions of the preferential interaction, preferential hydration, and excluded volume models commonly used to describe Hofmeister effects. Instead, they suggest that the Hofmeister anion effects on both stability and solubility of barnase are due to the way in which the protein interacts with water molecules, and in particular with water dipoles, which are more ordered around sulfate anions and less ordered around thiocyanate anions
Treatment of hypertension following endotracheal intubation A study comparing the efficacy of labetalol, practolol and placebo
Labetalol, a new adrenergic receptor antagonist, has both a- and B-blocking properties. Intravenous labetalol (0,25 and 0,5 mg/kg), practolol (0,4 mg/kg) and saline (1 ml), injected prior to anaesthesia, were compared with respect to their effect on the haemodynamic consequences of direct laryngoscopy followed by the passage of an endotracheal tube. When compared with intravenous saline injection, both labetalol and practolol obtunded the tachycardia induced by endotracheal intubation. The higher dose of labetalol was more effective in reducing the hypertensive response than the lower dose of practolol. However, none of the regimens completely abolished the adverse haemodynamic consequences of laryngotracheal, manipulations
Computer simulations of hard pear-shaped particles
We report results obtained from Monte Carlo simulations investi-
gating mesophase formation in two model systems of hard pear-shaped
particles. The first model considered is a hard variant of the trun-
cated Stone-Expansion model previously shown to form nematic and
smectic mesophases when embedded within a 12-6 Gay-Berne-like po-
tential [1]. When stripped of its attractive interactions, however, this system is found to lose its liquid crystalline phases. For particles of length to breadth ratio k = 3, glassy behaviour is seen at high pressures, whereas for k = 5 several bi-layer-like domains are seen, with high intradomain order but little interdomain orientational correlation. For the second model, which uses a parametric shape parameter based on the generalised Gay-Berne formalism, results are presented for particles with elongation k = 3; 4 and 5. Here, the systems with k = 3 and 4 fail to display orientationally ordered phases, but that with k = 5 shows isotropic, nematic and, unusually for a hard-particle model, interdigitated smectic A2 phases.</p
Extended Superscaling of Electron Scattering from Nuclei
An extended study of scaling of the first and second kinds for inclusive
electron scattering from nuclei is presented. Emphasis is placed on the
transverse response in the kinematic region lying above the quasielastic peak.
In particular, for the region in which electroproduction of resonances is
expected to be important, approximate scaling of the second kind is observed
and the modest breaking of it is shown probably to be due to the role played by

Charmless Decays Based on the six-quark Effective Hamiltonian with Strong Phase Effects II
We provide a systematic study of charmless decays (
and denote pseudoscalar and vector mesons, respectively) based on an
approximate six-quark operator effective Hamiltonian from QCD. The calculation
of the relevant hard-scattering kernels is carried out, the resulting
transition form factors are consistent with the results of QCD sum rule
calculations. By taking into account important classes of power corrections
involving "chirally-enhanced" terms and the vertex corrections as well as weak
annihilation contributions with non-trivial strong phase, we present
predictions for the branching ratios and CP asymmetries of decays into
PP, PV and VV final states, and also for the corresponding polarization
observables in VV final states. It is found that the weak annihilation
contributions with non-trivial strong phase have remarkable effects on the
observables in the color-suppressed and penguin-dominated decay modes. In
addition, we discuss the SU(3) flavor symmetry and show that the symmetry
relations are generally respected
Clumped-isotope palaeothermometry and LA-ICP-MS U–Pb dating of lava-pile hydrothermal calcite veins
Calcite veins are a common product of hydrothermal fluid circulation. Clumped-isotope palaeothermometry is a promising technique for fingerprinting the temperature of hydrothermal fluids, but clumped-isotope systematics can be reset at temperatures of > ca. 100 °C. To model whether the reconstructed temperatures represent calcite precipitation or closed-system resetting, the precipitation age must be known. LA-ICP-MS U–Pb dating of calcite is a recently developed approach to direct dating of calcite and can provide precipitation ages for modelling clumped-isotope systematics in calcite veins. In this study, clumped-isotope and LA-ICP-MS U–Pb calcite analyses were combined in basalt-hosted calcite veins from three settings in Scotland. Samples from all three localities yielded precipitation temperatures of ca. 75–115 °C from clumped-isotope analysis, but veins from only two of the sites were dateable, yielding precipitation ages of 224 ± 8 Ma and 291 ± 33 Ma (2σ). Modelling from the dated samples enabled confident interpretation that no closed-system resetting had occurred in these samples. However, the lack of a precipitation age from the third location meant that a range of possible thermal histories had to be modelled meaning that confidence that resetting had not occurred was lower. This highlights the importance of coupling clumped-isotope thermometry and LA-ICP-MS U–Pb calcite dating in determining the temperature of hydrothermal fluids recorded in calcite veins. This paired approach is shown to be robust in constraining the timing and precipitation temperature of calcite formation, and thus for tracking hydrothermal processes
