    Evaluating monitoring methods for cetaceans

    With increasing human pressures on wildlife comes a responsibility to monitor them effectively, particularly in an environment of declining research funds. Scarce funding resources compromise the level and efficacy of monitoring possible to detect trends in abundance, highlighting the priority for developing cost-effective programs. A systematic and rigorous sampling regime was developed to estimate abundance of a small, genetically isolated spinner dolphin (Stenella longirostris) population exposed to high levels of human activities. Five monitoring scenarios to detect trends in abundance were evaluated by varying sampling effort, precision, power, and sampling interval. Scenario 1 consisted of monthly surveys, each of 12 days, used to obtain the initial two consecutive annual abundance estimates. Scenarios 2, 3, and 4 consisted of a reduced effort, while Scenario 5 doubled the effort of Scenario 1. Scenarios with the greatest effort (1 and 5) produced the most precise abundance estimates (CV = 0.09). Using a CV = 0.09 and power of 80%, it would take 9 years to detect a 5% annual change in abundance compared with 12 years at a power of 95%. Under this best-case monitoring scenario, if the trend was a decline, the population would have decreased by 37% and 46%, respectively, prior to detection of a significant decline: With the potential of a large decline in a small population prior to detection, the lower power level should be used to trigger a management intervention. The approach presented here is applicable across taxa for which individuals can be identified, including terrestrial and aquatic mammals, birds, and reptiles

    Loss of the homeostatic protein BPIFA1, leads to exacerbation of otitis media severity in the Junbo mouse model

    Otitis Media (OM) is characterized by epithelial abnormalities and defects in innate immunity in the middle ear (ME). Although, BPIFA1, a member of the BPI fold containing family of putative innate defence proteins is abundantly expressed by the ME epithelium and SNPs in Bpifa1 have been associated with OM susceptibility, its role in the ME is not well characterized. We investigated the role of BPIFA1 in protection of the ME and the development of OM using murine models. Loss of Bpifa1 did not lead to OM development. However, deletion of Bpifa1 in Evi1Jbo/+mice, a model of chronic OM, caused significant exacerbation of OM severity, thickening of the ME mucosa and increased collagen deposition, without a significant increase in pro-inflammatory gene expression. Our data suggests that BPIFA1 is involved in maintaining homeostasis within the ME under steady state conditions and its loss in the presence of inflammation, exacerbates epithelial remodelling leading to more severe OM

    Human muscle-derived cell populations isolated by differential adhesion rates: Phenotype and contribution to skeletal muscle regeneration in Mdx/SCID mice

    Muscle-derived stem cells (MDSCs) isolated from murine skeletal tissue by the preplate method have displayed the capability to commit to the myogenic lineage and regenerate more efficiently than myoblasts in skeletal and cardiac muscle in murine Duchenne Muscular Dystrophy mice (mdx). However, until now, these studies have not been translated to human muscle cells. Here, we describe the isolation, by a preplate technique, of candidate human MDSCs, which exhibit myogenic and regenerative characteristics similar to their murine counterparts. Using the preplate isolation method, we compared cells that adhere faster to the flasks, preplate 2 (PP2), and cells that adhere slower, preplate 6 (PP6). The human PP6 cells express several markers of mesenchymal stem cells and are distinct from human PP2 (a myoblast-like population) based on their expression of CD146 and myogenic markers desmin and CD56. After transplantation to the gastrocnemius muscle of mdx/SCID mice, we observe significantly higher levels of PP6 cells participating in muscle regeneration as compared with the transplantation of PP2 cells. This study supports some previous findings related to mouse preplate cells, and also identifies some differences between mouse and human muscle preplate cells

    Quantitative trait loci for bone traits segregating independently of those for growth in an F-2 broiler X layer cross

    An F broiler-layer cross was phenotyped for 18 skeletal traits at 6, 7 and 9 weeks of age and genotyped with 120 microsatellite markers. Interval mapping identified 61 suggestive and significant QTL on 16 of the 25 linkage groups for 16 traits. Thirty-six additional QTL were identified when the assumption that QTL were fixed in the grandparent lines was relaxed. QTL with large effects on the lengths of the tarsometatarsus, tibia and femur, and the weights of the tibia and femur were identified on GGA4 between 217 and 249 cM. Six QTL for skeletal traits were identified that did not co-locate with genome wide significant QTL for body weight and two body weight QTL did not coincide with skeletal trait QTL. Significant evidence of imprinting was found in ten of the QTL and QTL x sex interactions were identified for 22 traits. Six alleles from the broiler line for weight- and size-related skeletal QTL were positive. Negative alleles for bone quality traits such as tibial dyschondroplasia, leg bowing and tibia twisting generally originated from the layer line suggesting that the allele inherited from the broiler is more protective than the allele originating from the layer

    The Hamiltonian limit of (3+1)D SU(3) lattice gauge theory on anisotropic lattices

    The extreme anisotropic limit of Euclidean SU(3) lattice gauge theory is examined to extract the Hamiltonian limit, using standard path integral Monte Carlo (PIMC) methods. We examine the mean plaquette and string tension and compare them to results obtained within the Hamiltonian framework of Kogut and Susskind. The results are a significant improvement upon previous Hamiltonian estimates, despite the extrapolation procedure necessary to extract observables. We conclude that the PIMC method is a reliable method of obtaining results for the Hamiltonian version of the theory. Our results also clearly demonstrate the universality between the Hamiltonian and Euclidean formulations of lattice gauge theory. It is particularly important to take into account the renormalization of both the anisotropy, and the Euclidean coupling βE \beta_E , in obtaining these results.Comment: 10 pages, 11 figure
