    Initial experience using a femtosecond laser cataract surgery system at a UK National Health Service cataract surgery day care centre

See: http://creativecommons.org/licenses/by-nc/4.0/OBJECTIVES: To describe the initial outcomes following installation of a cataract surgery laser system.SETTING: National Health Service cataract surgery day care unit in North London, UK.PARTICIPANTS: 158 eyes of 150 patients undergoing laser-assisted cataract surgery.INTERVENTIONS: Laser cataract surgery using the AMO Catalys femtosecond laser platform.PRIMARY AND SECONDARY OUTCOME MEASURES: PRIMARY OUTCOME MEASURE: intraoperative complications including anterior and posterior capsule tears.SECONDARY OUTCOME MEASURES: docking to the laser platform, successful treatment delivery, postoperative visual acuities.RESULTS: Mean case age was 67.7±10.8 years (range 29-88 years). Docking was successful in 94% (148/158 cases), and in 4% (6/148 cases) of these, the laser delivery was aborted part way during delivery due to patient movement. A total of 32 surgeons, of grades from junior trainee to consultant, performed the surgeries. Median case number per surgeon was 3 (range from 1-20). The anterior capsulotomy was complete in 99.3% of cases, there were no anterior capsule tears (0%). There were 3 cases with posterior capsule rupture requiring anterior vitrectomy, and 1 with zonular dialysis requiring anterior vitrectomy (4/148 eyes, 2.7%). These 4 cases were performed by trainee surgeons, and were either their first laser cataract surgery (2 surgeons) or their first and second laser cataract surgeries (1 surgeon).CONCLUSIONS: Despite the learning curve, docking and laser delivery were successfully performed in almost all cases, and surgical complication rates and visual outcomes were similar to those expected based on national data. Complications were predominately confined to trainee surgeons, and with the exception of intraoperative pupil constriction appeared unrelated to the laser-performed steps.Peer reviewe

    The U.S. Deaf community is a sociolinguistic minority group of at least 500,000 individuals who communicate using American Sign Language (ASL).1 ASL is fully distinct from English – i.e., it is not “English on the hands.” ASL is a natural, formal language with its own syntax, morphology, and structure. Members of the Deaf community identify as members of a cultural minority group with shared language, experience, history, art, and literature. This tip sheet focuses on best practices for sharing research findings with culturally Deaf individuals who primarily use ASL. However, many of the strategies described below align with principles for universal accessibility and will, therefore, apply to a diverse range of hearing people and people with hearing loss

    Enhanced Retention In The Passive-Avoidance Task By 5-HT1A Receptor Blockade Is Not Associated With Increased Activity Of The Central Nucleus Of The Amygdala

    The effect of blockade of S-HT1A receptors was investigated on (1) retention in a mildly aversive passive-avoidance task, and (2) spontaneous single-unit activity of central nucleus of the amygdala (CeA) neurons, a brain site implicated in modulation of retention. Systemic administration of the selective S-HT1A antagonist NAN-190 immediately after training markedly-and dose-dependently-facilitated retention in the passive-avoidance task; enhanced retention was time-dependent and was not attributable to variations in wattages of shock received by animals. Systemic administration of NAN-190 had mixed effects on spontaneous single-unit activity of CeA neurons recorded extracellularly in vivo; microiontophoretic application of S-HT, in contrast, consistently and potently suppressed CeA activity. The present findings-that S-HT1A receptor blockade by NAN-190 (1) enhances retention in the passive-avoidance task, and (2) does not consistently increase spontaneous neuronal activity of the CeA-provide evidence that a serotonergic system tonically inhibits modulation of retention in the passive-avoidance task through activation of the S-HT1A receptor subtype at brain sites located outside the CeA

    King-plot analysis of isotope shifts in simple diatomic molecules

    We demonstrate that the isotope shift in isotopomers of diatomic molecules, where the nucleus of one of its constituent atoms is replaced by another isotope, can be expressed as the sum of a field shift and a mass shift, similar to the atomic case. We show that a linear relation holds between atomic and molecular isotopes shifts, thus extending the King-plot analysis to molecular isotope shifts. Optical isotope shifts in YbF and ZrO and infrared isotope shifts in SnH are analyzed with a molecular King-plot approach, utilizing Yb+^{+} and Zr+^{+} ionic isotope shifts and charge radii of Sn obtained with non-optical methods. The changes in the mean-squared nuclear charge radii δr2\delta \langle r^2 \rangle of 170174,176^{170-174,176}Yb and 9092,94,96^{90-92,94,96}Zr extracted from the molecular transitions are found to be in excellent agreement with the values from the spectroscopy of Yb+^{+} and Zr+^{+}, respectively. On the contrary, in the case of the vibrational-rotational transition in SnH, no sensitivity to the nuclear volume could be deduced within the experimental resolution, which makes it unsuitable for the extraction of nuclear charge radii but provides insights into the molecular electronic wave function not accessible via other methods. The new opportunities offered by the molecular King-plot analysis for research in nuclear structure and molecular physics are discussed.Comment: Accepted at Physical Review X.     Acute exposure to ultraviolet radiation targets proteins involved in collagen fibrillogenesis

    Introduction: Exposure to chronic, low-dose UV irradiation (UVR) can lead to premature ageing of the skin. Understanding which proteins are affected by acute UVR and photo-dynamically produced reactive oxygen species (ROS) could help to inform strategies to delay photoageing. Conventional biochemical analyses can be used to characterize UVR/ROS-induced damage on a protein-by-protein basis and we have previously shown using SDS-PAGE that collagen I and plasma fibronectin are respectively resistant and susceptible to physiological doses of UVR. The aim of this study was to screen a complex proteome for UVR-affected proteins.Methods: This study employed a sensitive mass spectrometry technique (peptide location fingerprinting: PLF) which can identify structure associated differences following trypsin digestion to characterize the impact of UVR exposure on purified collagen I and tissue fibronectin and to identify UVR-susceptible proteins in an ECM-enriched proteome.Results: Using LC/MS-MS and PLF we show that purified mature type-I collagen is resistant to UVR, whereas purified tissue fibronectin is susceptible. UV irradiation of a human dermal fibroblast-deposited ECM-enriched proteome in vitro, followed by LC/MS-MS and PLF analysis revealed two protein cluster groups of UV susceptible proteins involved in i) matrix collagen fibril assembly and ii) protein translation and motor activity. Furthermore, PLF highlighted UV susceptible domains within targeted matrix proteins, suggesting that UV damage of matrix proteins is localized.Discussion: Here we show that PLF can be used to identify protein targets of UVR and that collagen accessory proteins may be key targets in UVR exposed tissues

    Sampling nucleotide diversity in cotton

    <p>Abstract</p> <p>Background</p> <p>Cultivated cotton is an annual fiber crop derived mainly from two perennial species, <it>Gossypium hirsutum </it>L. or upland cotton, and <it>G. barbadense </it>L., extra long-staple fiber Pima or Egyptian cotton. These two cultivated species are among five allotetraploid species presumably derived monophyletically between <it>G. arboreum </it>and <it>G. raimondii</it>. Genomic-based approaches have been hindered by the limited variation within species. Yet, population-based methods are being used for genome-wide introgression of novel alleles from <it>G. mustelinum </it>and <it>G. tomentosum </it>into <it>G. hirsutum </it>using combinations of backcrossing, selfing, and inter-mating. Recombinant inbred line populations between genetics standards TM-1, (<it>G. hirsutum</it>) × 3-79 (<it>G. barbadense</it>) have been developed to allow high-density genetic mapping of traits.</p> <p>Results</p> <p>This paper describes a strategy to efficiently characterize genomic variation (SNPs and indels) within and among cotton species. Over 1000 SNPs from 270 loci and 279 indels from 92 loci segregating in <it>G. hirsutum </it>and <it>G. barbadense </it>were genotyped across a standard panel of 24 lines, 16 of which are elite cotton breeding lines and 8 mapping parents of populations from six cotton species. Over 200 loci were genetically mapped in a core mapping population derived from TM-1 and 3-79 and in <it>G. hirsutum </it>breeding germplasm.</p> <p>Conclusion</p> <p>In this research, SNP and indel diversity is characterized for 270 single-copy polymorphic loci in cotton. A strategy for SNP discovery is defined to pre-screen loci for copy number and polymorphism. Our data indicate that the A and D genomes in both diploid and tetraploid cotton remain distinct from each such that paralogs can be distinguished. This research provides mapped DNA markers for intra-specific crosses and introgression of exotic germplasm in cotton.</p

    Engaging Communities in Emergency Risk and Crisis Communication: Mixed-Method Systematic Review and Evidence Synthesis

    The World Health Organization (WHO) commissioned systematic reviews to assist with the development of new emergency risk communication guidelines that will impact responses and distribution of resources at all levels. This mixed-method evidence synthesis, guided by Cochrane principles and methods, examined the extant research in countries throughout the world, published from 2003 to 2016, related to the best practices to engage communities in preparing for and responding to emergency events with public health implications. Although few studies directly examined which strategies or tactics effectively engage public participation, many studies reinforced the importance of community participation. The findings support the perspective that emergency events are communicatively understood by all publics and that they benefit from emergency risk communication before, during, and after such events, especially when grounded in local contexts. Although the importance of local context limits the generalizability of risk communication, it is important to continue studying strategies and tactics to cultivate participation among all stakeholders
