    Analysis of Histocial Distribution of SAV in the Eastern Shore Coastal Basins and Mid-Bay Island Complexes as Evidence of Historical Water Quality Conditions and a Restored Bay Ecosystem

    Historical black and white format photographs at scales of approximately 1:20,000, dating from 1952 to 1956 were used to delineate the maximum coverage of SAV in the study region. Coverage of photography from decades before and after this period were found to generally to be of poorer quality and show less SAV presence. Photo-interpretation of the aerial photographs was accomplished using a head-up, on-screen digitizing system at fixed image scale of 1:12,000 and followed as closely as possible the methods currently used to delineate SAV beds throughout the Chesapeake Bay as well as the delineation of historical SAV coverage for other region. A total of 13,046 hectares of sub-tidal bottom in the Eastern Shore bay region between the tip of Fisherman’s Island to the Virginia-Maryland border, including the mid-bay island complex, were found to display SAV signatures.     The human health effects of exposure to polybrominated biphenyls

    Polybrominated biphenyls (PBB) were inadvertently introduced into the food chain in Michigan in 1973. Fifty-one people with known exposure to PBB were studied. Twenty-three farmers with disabling health complaints, and 28 chemical workers involved in the PBB manufacturing process were systematically evaluated to determine if these highrisk groups suffered adverse effects from their known exposure to PBB. The farmers had a high frequency of constitutional symptoms, hepatomegaly and skin rashes, findings not commonly noted in the chemical workers. Biochemical and hematologic testing revealed few abnormalities, and electromyograms, nerve conduction velocities, endocrine studies, and lymphocyte transformation studies provided no objective findings that correlated with subjective complaints. There was no relationship between PBB levels and physical or laboratory abnormalities. Present evidence suggests that people exposed to PBB have few objective findings at this time, and reactive depression may be responsible for the high prevalence of constitutional symptoms.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/24421/1/0000692.pd

    Aerial surveys of coastal habitats can uniquely inform the science and management of shallow, coastal zones, and when repeated annually,theyrevealchangesthatareotherwisedifficulttoassess fromground-basedsurveys.Thispaperreviewstheutilityofalongterm(1984–present)annualaerialmonitoringprogramforsubmersedaquaticvegetation(SAV)inChesapeakeBay,itstidaltributaries, and nearby Atlantic coastal bays, USA. We present a series of applications that highlight the program’s importance in assessing anthropogenic impacts, gauging water quality status and trends, establishing and evaluating restoration goals, and understanding the impactofcommercialfishingpracticesonbenthichabitats.Theseexamplesdemonstratehowperiodicallyquantifyingcoverageofthis important foundational habitat answers basic research questions locally, as well as globally, and provides essential information to resource managers. New technologies are enabling more frequent and accurate aerial surveys at greater spatial resolution and lower cost. These advances will support efforts to extend the applications described here to similar issues in other areas

    The validation of pharmacogenetics for the identification of Fabry patients to be treated with migalastat

    PURPOSE: Fabry disease is an X-linked lysosomal storage disorder caused by mutations in the α-galactosidase A gene. Migalastat, a pharmacological chaperone, binds to specific mutant forms of α-galactosidase A to restore lysosomal activity. METHODS: A pharmacogenetic assay was used to identify the α-galactosidase A mutant forms amenable to migalastat. Six hundred Fabry disease-causing mutations were expressed in HEK-293 (HEK) cells; increases in α-galactosidase A activity were measured by a good laboratory practice (GLP)-validated assay (GLP HEK/Migalastat Amenability Assay). The predictive value of the assay was assessed based on pharmacodynamic responses to migalastat in phase II and III clinical studies. RESULTS: Comparison of the GLP HEK assay results in in vivo white blood cell α-galactosidase A responses to migalastat in male patients showed high sensitivity, specificity, and positive and negative predictive values (≥0.875). GLP HEK assay results were also predictive of decreases in kidney globotriaosylceramide in males and plasma globotriaosylsphingosine in males and females. The clinical study subset of amenable mutations (n = 51) was representative of all 268 amenable mutations identified by the GLP HEK assay. CONCLUSION: The GLP HEK assay is a clinically validated method of identifying male and female Fabry patients for treatment with migalastat
